    Epifluorescence microscope methods for bacterial enumeration in a 4-chlorophenol degrading consortium

    Epifluorescence microscope methods, namely BacLight, direct epifluorescence filter technique and Rhodamine 123, consistently underestimated plate bacterial counts in a 4-chlorophenol degrading consortium. Cells capable of passing through 0.2 mgrm filters, referred as `ultramicrocells', were found. Although cell counts were higher when traditional methods were used, BacLight and direct epifluorescence filter technique were convenient techniques for the systematic monitoring of bacteria involved in biodegradation processes, as results were consistent and available within a short time

    Root system traits and its relationship with photosynthesis and productivity in four maize genotypes under drought

    The present study combined evaluations of agronomic parameters such as roots morphometry (using the WinRhizo program) and leaf gas exchange, in order to detect features in the root system which allow the maintenance of photosynthetic rates and productivity in four maize genotypes contrasting for tolerance to water deficit (WD), two tolerant (DKB 390 and BRS1055) and two sensitive (BRS 1010 and 2B710). The genotypes showed similar tolerance to dehydration of leaf tissue, but the tolerant genotypes DKB 390 and BRS1055 presented higher photosynthetic rate and yield compared to the sensitive BRS 1010 and 2B710. Nevertheless, divergent strategies of adaptation to drought among tolerant genotypes were observed. The genotype DKB 390 presented physiological mechanisms in shoots responsible for minimizing water loss, which decreases the dependence of root adjustments to increase the absorption of water. In turn, the BRS 1055 genotype showed a drought avoidance strategy by producing fine roots associated with a higher leaf area

    Ultrasonography detection of pregnancy in outdoor iberian sows

    In a flock of pure Iberian sows, with outdoor breeding and total freedom, ultrasonography detection of pregnancy length is studied (11.1±0.6 seconds), according to pregnancy or not pregnancy (9.7 vs 20.4 seconds). Also, possible influence of cycle number is analysed. By other side permanence of ear tags after a year is valued (99.5%). Results show that indoor and intensive production is not justified in order to attain more reproductive control.En un rebaño de cerdas Ibéricas puras, criadas en régimen de total libertad (sin empleo de jaulas), se estudia el tiempo empleado en el diagnóstico de gestación con ecógrafo (11,1±0,6 segundos por cerda), se analiza la duración del diagnóstico dependiendo de si hay preñez o no (9,7 vs 20,4 segundos) y la posible influencia del número de ciclo. Por otra parte se valora la permanencia de los crotales de identificación individual tras un año (99,5%). Los resultados indican que la intensificación de la producción en esta raza no está justificada por la consecución de un mayor control reproductivo

    Transforming growth factor-β (TGF-β) maintains follicular ultrastructure and stimulates preantral follicle growth in caprine ovarian tissue cultured in vitro

    O objetivo desse estudo foi investigar se o TGF-β afeta a sobrevivência, ativação e crescimento de folículos primordiais caprinos inclusos no córtex ovariano após o cultivo in vitro. Ovários de cabras foram coletados em abatedouro e fragmentos de tecido ovariano foram cultivados por um e sete dias em meio essencial mínimo alfa (α-MEM+) sozinho ou suplementado com TGF-β (1, 5, 10 ou 50ng/mL). Fragmentos ovarianos não cultivados e cultivados foram processados para análise histológica e ultraestrutural. Os resultados mostraram que, comparado ao controle fresco, houve diminuição no percentual de folículos morfologicamente normais em todos os tratamentos somente após sete dias de cultivo. O TGF-β não afetou a ativação folicular independente da concentração testada, contudo, o diâmetro folicular foi superior (P<0.05) no tratamento com 10ng/mL de TGF-β quando comparado ao controle fresco e aos demais tratamentos. Além disso, essa mesma concentração manteve a ultraestrutura normal dos folículos após sete dias de cultivo. Em conclusão, o TGF-β apresentou efeito adicional no crescimento folicular e na manutenção da integridade ultraestrutural de folículos pré-antrais caprinos inclusos no tecido ovariano quando utilizado na concentração de 10ng/mL durante sete dias de cultivo.The objectives of this study were to investigate whether TGF-β affect the survival, activation and further growth of goat primordial follicles enclosed in ovarian cortex after in vitro culture. Goat ovaries were collected from an abattoir and pieces of ovarian tissues were cultured for one or seven days in a supplemented alpha Minimum Essential Medium, alone or containing TGF-β (1, 5, 10 or 50ng/mL). Ovarian tissues from the fresh control as well as those cultured were processed for histological and ultrastructural studies. The results showed that when compared with fresh control, there was decrease in the percentages of histologically normal follicles in all treatments only after seven days culture. TGF-β did not affect the activation of preantral follicles regardless of its concentration, however, larger follicles diameter (P<0.05) was observed using 10ng/mL TGF-β than in the fresh control and other treatments. Moreover, this concentration maintained the normal ultrastructure after seven days of culture. In conclusion, TGF-β showed additional effect on the follicle growth and the maintenance of ultrastructural integrity of goat preantral follicles enclosed in ovarian tissue when used at 10ng/mL during seven days of culture

    Effective Field Theory for Layered Quantum Antiferromagnets with Non-Magnetic Impurities

    We propose an effective two-dimensional quantum non-linear sigma model combined with classical percolation theory to study the magnetic properties of site diluted layered quantum antiferromagnets like La2_{2}Cu1x_{1-x}Mx_xO4_{4} (M==Zn, Mg). We calculate the staggered magnetization at zero temperature, Ms(x)M_s(x), the magnetic correlation length, ξ(x,T)\xi(x,T), the NMR relaxation rate, 1/T1(x,T)1/T_1(x,T), and the N\'eel temperature, TN(x)T_N(x), in the renormalized classical regime. Due to quantum fluctuations we find a quantum critical point (QCP) at xc0.305x_c \approx 0.305 at lower doping than the two-dimensional percolation threshold xp0.41x_p \approx 0.41.     We study the existence of stationary classical solutions of the incompressible Euler equation in the plane that approximate singular stationnary solutions of this equation. The construction is performed by studying the asymptotics of equation -\eps^2 \Delta u^\eps=(u^\eps-q-\frac{\kappa}{2\pi} \log \frac{1}{\eps})_+^p with Dirichlet boundary conditions and qq a given function. Actualmente se están realizando importantes esfuerzos para materializarlo en base a las posiciones de radio-fuentes de origen extragaláctico, medidas con interferómetros de linea-base muy larga.     Neurolysin knockout mice generation and initial phenotype characterization

    The oligopeptidase neurolysin (EC 3.4.24.16; Nln) was first identified in rat brain synaptic membranes and shown to ubiquitously participate in the catabolism of bioactive peptides such as neurotensin and bradykinin. Recently, it was suggested that Nln reduction could improve insulin sensitivity. Here, we have shown that Nln knockout mice (KO) have increased glucose tolerance, insulin sensitivity and gluconeogenesis. KO mice have increased liver mRNA for several genes related to gluconeogenesis. Isotopic label semi-quantitative peptidomic analysis suggests increase in specific intracellular peptides in gastrocnemius and epididymal adipose tissue, which likely is involved with the increased glucose tolerance and insulin sensitivity in the KO mice. These results suggest the exciting new possibility that Nln is a key enzyme for energy metabolism and could be a novel therapeutic target to improve glucose uptake and insulin sensitivity
