    A practical and low cost microbiotest to assess the phytotoxic potential of growing media and soil

    For routine toxicity testing of composts and growing media, two different types of assays are usually applied. One approach is the use of growing trials in pots with various mixtures of the material. In extract-based tests, seeds are germinated in petri dishes and exposed to an exudate from the material to be tested. In this study, one of these methods, the Phytotoxkit microbiotest, was compared to the standard phytotoxicity test with Lactuca sativa as used by the RHP foundation. The Phytotoxkit test was performed in transparent test containers which allow for direct observation and length measurements of the seedlings by means of image analysis. Two dicotyles (Lepidium sativum and Sinapis alba) and one monocotyle (Sorghum saccharatum) were used. When measuring a presumably phytotoxic bark, in comparison with a reference bark, the Phytotoxkit showed a seedling length inhibition of 60¿80% for the dicotyles and less than 20% for the monocotyle after three days. The RHP growing trial showed 30% weight inhibition of the lettuce after two weeks of plant growth when comparing the same barks in mixtures. This preliminary comparison showed that the Phytotoxkit microbiotest is a quick and practical bioassay with a high resolution which has a potential to become an international standard

    The Freshwater Information Platform: a global online network providing data, tools and resources for science and policy support

    Freshwaters are among the most complex, dynamic, and diverse ecosystems globally. Despite their small share of the earth’s surface (less than 1%) they are home to over 10% of all known animal species. Biodiversity decrease in general and freshwater biodiversity decline in particular have recently received increasing attention, and various policy instruments are now targeting the conservation, protection and enhancement of biodiversity and associated ecosystem services. Surveillance programs as well as a variety of research projects have been producing a tremendous amount of freshwater-related information. Though there have been various attempts to build infrastructures for online collection of such data, tools and reports, they often provide only limited access to resources that can readily be extracted for conducting large scale analyses. Here, we present the Freshwater Information Platform, an open system of relevant freshwater biodiversity-related information. We provide a comprehensive overview of the platform’s core components, highlight their values, present options for their use, and discuss future developments. This is complemented by information on the platform’s current management structure, options for contributing data and research results and an outlook for the future

    Cell line derived xenograft mouse models are a suitable in vivo model for studying tumor budding in colorectal cancer

    Tumor budding (TB) is an important prognostic parameter in colorectal cancer (CRC) and associated with metastasis. However, the mechanisms of TB have not been fully elucidated and a major limitation is the absence of in vivo models. Here, we determine the suitability of human cell line derived xenografts (CDX) as models of TB in CRC. Pan-cytokeratin (CK)-stained next-generation Tissue Microarrays (ngTMA) of two CDX models (HT-29, n = 12 and HCT-8, n = 8) and human CRC (n = 27 high-grade and 25 low-grade budding tumors, each) were evaluated for TB. Immunohistochemistry for E-cadherin, β-catenin, Ki-67, ZEB1, and TWIST1 was performed. HT-29 and HCT-8 were predominantly high-grade and no/low-grade TB tumors, respectively. TB counts in the tumor center (intratumoral budding, ITB) were significantly higher in HT-29 CDX tumors compared to human CRC (p = 0.0099). No difference was found in TB counts at the invasion front (peritumoral budding, PTB; p=0.07). ITB and PTB were strongly correlated (r = 0.438 and r = 0.62 in CDX and human CRC, respectively). Immunohistochemistry profiles were comparable in CDX and human CRC tissues. TB in the CDX mouse models is phenotypically similar to human CRCs and highlights comparable protein profiles.     Effects of Grassland Management on Herbage Lipid Composition and Consequences for Fatty Acids in Milk

    Herbage provides bulk feed and is the basis for ruminant nutrition. Herbage lipids, especially C18:3, are a major source of beneficial fatty acids (FA) in milk. These desired FA are unsaturated FA such as CLA (conjugated linoleic acid), especially the isomer rumenic acid, and also vaccenic acid, both trans omega-7 FA (Ellen & Elgersma, 2004). As information on lipids in forages is scarce, effects were studied of N application level and regrowth period on the lipid concentration and FA composition of perennial ryegrass (Lolium perenne L.), the most important forage in temperate climate zones. A linear relation had previously been found between C18:3 intake of cows stall-fed with fresh grass and the amount of omega-7 FA in milk (Elgersma et al., 2003)

    Facial emotion detection in Vestibular Schwannoma patients with and without facial paresis

    This study investigates whether there exist differences in facial emotion detection accuracy in patients suffering from Vestibular Schwannoma (VS) due to their facial paresis. Forty-four VS patients, half of them with, and half of them without a facial paresis, had to classify pictures of facial expressions as being emotional or non-emotional. The visual information of images was systematically manipulated by adding different levels of visual noise. The study had a mixed design with emotional expression (happy vs. angry) and visual noise level (10% to 80%) as repeated measures and facial paresis (present vs. absent) and degree of facial dysfunction as between subjects' factors. Emotion detection accuracy declined when visual information declined, an effect that was stronger for anger than for happy expressions. Overall, emotion detection accuracy for happy and angry faces did not differ between VS patients with or without a facial paresis, although exploratory analyses suggest that the ability to recognize emotions in angry facial expressions was slightly more impaired in patients with facial paresis. The findings are discussed in the context of the effects of facial paresis on emotion detection, and the role of facial mimicry, in particular, as an important mechanism for facial emotion processing and understanding.info:eu-repo/semantics/publishedVersio
