    Detection of equine atypical myopathy-associated hypoglycin A in plant material: Optimisation and validation of a novel LC-MS based method without derivatisation

    Hypoglycin A (HGA) toxicity, following ingestion of material from certain plants, is linked to an acquired multiple acyl-CoA dehydrogenase deficiency known as atypical myopathy, a commonly fatal form of equine rhabdomyolysis seen worldwide. Whilst some plants are known to contain this toxin, little is known about its function or the mechanisms that lead to varied HGA concentrations between plants. Consequently, reliable tools to detect this amino acid in plant samples are needed. Analytical methods for HGA detection have previously been validated for the food industry, however, these techniques rely on chemical derivatisation to obtain accurate results at low HGA concentrations. In this work, we describe and validate a novel method, without need for chemical derivatisation (accuracy = 84–94%; precision = 3–16%; reproducibility = 3–6%; mean linear range R2 = 0.999). The current limit of quantitation for HGA in plant material was halved (from 1μg/g in previous studies) to 0.5μg/g. The method was tested in Acer pseudoplatanus material and other tree and plant species. We confirm that A. pseudoplatanus is most likely the only source of HGA in trees found within European pastures

In addition, the materials can contain or develop cracks. Once a crack has initiated, the conditions under which the components operate can cause these cracks to propagate.     Conformational transformations induced by the charge-curvature interaction at finite temperature

    The role of thermal fluctuations on the conformational dynamics of a single closed filament is studied. It is shown that, due to the interaction between charges and bending degrees of freedom, initially circular aggregates may undergo transformation to polygonal shape. The transition occurs both in the case of hardening and softening charge-bending interaction. In the former case the charge and curvature are smoothly distributed along the chain while in the latter spontaneous kink formation is initiated.     Determination of material properties in the Chaboche unified viscoplasticity model

    An experimental programme of cyclic mechanical testing of a 316 stainless steel, at temperatures up to 600°C, under isothermal conditions, for the identification of material constitutive constants, has been carried out using a thermo-mechanical fatigue (TMF) test machine with induction coil heating. The constitutive model adopted is a modified Chaboche unified viscoplasticity model, which can deal with both cyclic effects, such as combined isotropic and kinematic hardening, and rate-dependent effects, associated with viscoplasticity. The characterization of 316 stainless steel is presented and compared with results from cyclic isothermal tests. A least-squares optimization algorithm has been developed and implemented for determining the material constants in order to further improve the general fit of the model to experimental data, using the initially obtained material constants as the starting point in this optimization process. The model predictions using both the initial and optimized material constants are compared to experimental data

    Methods to elicit probability distributions from experts: a systematic review of reported practice in health technology assessment

    The final publication is available at Springer via the DOI in this record.BACKGROUND: Elicitation is a technique that can be used to obtain probability distribution from experts about unknown quantities. We conducted a methodology review of reports where probability distributions had been elicited from experts to be used in model-based health technology assessments. METHODS: Databases including MEDLINE, EMBASE and the CRD database were searched from inception to April 2013. Reference lists were checked and citation mapping was also used. Studies describing their approach to the elicitation of probability distributions were included. Data was abstracted on pre-defined aspects of the elicitation technique. Reports were critically appraised on their consideration of the validity, reliability and feasibility of the elicitation exercise. RESULTS: Fourteen articles were included. Across these studies, the most marked features were heterogeneity in elicitation approach and failure to report key aspects of the elicitation method. The most frequently used approaches to elicitation were the histogram technique and the bisection method. Only three papers explicitly considered the validity, reliability and feasibility of the elicitation exercises. CONCLUSION: Judged by the studies identified in the review, reports of expert elicitation are insufficient in detail and this impacts on the perceived usability of expert-elicited probability distributions. In this context, the wider credibility of elicitation will only be improved by better reporting and greater standardisation of approach. Until then, the advantage of eliciting probability distributions from experts may be lost.NIH
