    Strong interaction of a turbulent spot with a shock-induced separation bubble

    Direct numerical simulations have been conducted to study the passage of a turbulent spot through a shock-induced separation bubble. Localized blowing is used to trip the boundary layer well upstream of the shock impingement, leading to mature turbulent spots at impingement, with a length comparable to the length of the separation zone. Interactions are simulated at free stream Mach numbers of two and four, for isothermal (hot) wall boundary conditions. The core of the spot is seen to tunnel through the separation bubble, leading to a transient reattachment of the flow. Recovery times are long due to the influence of the calmed region behind the spot. The propagation speed of the trailing interface of the spot decreases during the interaction and a substantial increase in the lateral spreading of the spot was observed. A conceptual model based on the growth of the lateral shear layer near the wingtips of the spot is used to explain the change in lateral growth rat

    Polarization restricts hepatitis C virus entry into HepG2 hepatoma cells

    The primary reservoir for hepatitis C virus (HCV) replication is believed to be hepatocytes, which are highly polarized with tight junctions (TJ) separating their basolateral and apical domains. HepG2 cells develop polarity over time, resulting in the formation and remodeling of bile canalicular (BC) structures. HepG2 cells expressing CD81 provide a model system to study the effects of hepatic polarity on HCV infection. We found an inverse association between HepG2-CD81 polarization and HCV pseudoparticle entry. As HepG2 cells polarize, discrete pools of claudin-1 (CLDN1) at the TJ and basal/lateral membranes develop, consistent with the pattern of receptor staining observed in liver tissue. The TJ and nonjunctional pools of CLDN1 show an altered association with CD81 and localization in response to the PKA antagonist Rp-8-Br-cyclic AMPs (cAMPs). Rp-8-Br-cAMPs reduced CLDN1 expression at the basal membrane and inhibited HCV infection, supporting a model where the nonjunctional pools of CLDN1 have a role in HCV entry. Treatment of HepG2 cells with proinflammatory cytokines, tumor necrosis factor alpha and gamma interferon, perturbed TJ integrity but had minimal effect(s) on cellular polarity and HCV infection, suggesting that TJ integrity does not limit HCV entry into polarized HepG2 cells. In contrast, activation of PKC with phorbol ester reduced TJ integrity, ablated HepG2 polarity, and stimulated HCV entry. Overall, these data show that complex hepatocyte-like polarity alters CLDN1 localization and limits HCV entry, suggesting that agents which disrupt hepatocyte polarity may promote HCV infection and transmission within the liver

    Intrahistiocytic Storage of Clofazimine Crystals in a Cat

    A 13-year-old castrated male Maine coon cat with a 5-year history of atypical mycobacteriosis was euthanized and submitted for necropsy. The cat had been kept in clinical remission since diagnosis using a combination of the antimycobacterial drug clofazimine and additional multimodal antimicrobial therapy. Grossly, tissues were diffusely discolored red-brown to yellow. Histologically, the myocardial interstitum was expanded by numerous, often multinucleated cells, which were distended by uniformly shaped acicular cytoplasmic spaces. These cells were immunopositive for CD18 and immunonegative for desmin, suggesting a histiocytic rather than muscular origin. Macrophages in other tissues contained similar acicular spaces. Ultrastructurally, the spaces were surrounded by 2 lipid membranes, resembling an autophagosome. Based on the clinical history and histologic, immunohistochemical, and ultrastructural data, we diagnosed clofazimine crystal storage. To our knowledge, this is the first report of clofazimine storage in a cat or within myocardial interstitial macrophages

    Mapping giant magnetic fields around dense solid plasmas by high resolution magneto-optical microscopy

    We investigate distribution of magnetic fields around dense solid plasmas generated by intense p-polarized laser (~10^{16} W.cm^{-2}, 100 fs) irradiation of magnetic tapes, using high sensitivity magneto optical microscopy. We present evidence for giant axial magnetic fields and map out for the first time the spatial distribution of these fields. By using the axial magnetic field distribution as a diagnostic tool we uncover evidence for angular momentum associated with the plasma. We believe this study holds significance for investigating the process under which a magnetic material magnetizes or demagnetizes under the influence of ultrashort intense laser pulses.Comment: 17 pages of text with 4 figure

    This report details work carried out under NERC grants NE/M008339/1 and NE/NO13026/1 which were collaborations between the British Geological Survey and Yorkshire Water, with an additional knowledge transfer component involving Scottish Water and Dŵr Cymru Welsh Water. The work examines whether models developed using environmental, topographical and geohazard information could complement existing management tools, and increase the understanding as to how pipe networks of different materials interact with their broader environment. This can be seen as a first step in identifying ways in which greater resilience could be built into pipe networks

    Magnetic field generation by coherent turbulence structures

    It is thought that the small-scale magnetic fields observed in accretion discs, galaxies and galactic clusters are generated by a dynamo process in which the turbulent plasma amplifies small initial magnetic fluctuations. Numerical simulations of turbulence have revealed that turbulence consists of filamentlike vortex structures superimposed on an incoherent background, which carry a considerable amount of the energy. The natural questions to ask are whether these coherent structures can generate a magnetic field and, if so, if the generated magnetic field is also filament-like. After setting up a turbulence model which consists only of vortex filaments, we show in an unambiguous way that the coherent structure can sustain kinematic dynamo action and that the magnetic field thus generated consists of relatively thick ribbons (flattened tubes) located in between vortices

    Early Potent Protection against Heterologous SIVsmE660 Challenge Following Live Attenuated SIV Vaccination in Mauritian Cynomolgus Macaques

    Background: Live attenuated simian immunodeficiency virus (SIV) vaccines represent the most effective means of vaccinating macaques against pathogenic SIV challenge. However, thus far, protection has been demonstrated to be more effective against homologous than heterologous strains. Immune correlates of vaccine-induced protection have also been difficult to identify, particularly those measurable in the peripheral circulation. Methodology/Principal Findings: Here we describe potent protection in 6 out of 8 Mauritian-derived cynomolgus macaques (MCM) against heterologous virus challenge with the pathogenic, uncloned SIVsmE660 viral stock following vaccination with live attenuated SIVmac251/C8. MCM provided a characterised host genetic background with limited Major Histocompatibility Complex (MHC) and TRIM5α allelic diversity. Early protection, observed as soon as 3 weeks post-vaccination, was comparable to that of 20 weeks vaccination. Recrudescence of vaccine virus was most pronounced in breakthrough cases where simultaneous identification of vaccine and challenge viruses by virus-specific PCR was indicative of active co-infection. Persistence of the vaccine virus in a range of lymphoid tissues was typified by a consistent level of SIV RNA positive cells in protected vaccinates. However, no association between MHC class I /II haplotype or TRIM5α polymorphism and study outcome was identified. Conclusion/Significance: This SIV vaccine study, conducted in MHC-characterised MCM, demonstrated potent protection against the pathogenic, heterologous SIVsmE660 challenge stock after only 3 weeks vaccination. This level of protection against this viral stock by intravenous challenge has not been hitherto observed. The mechanism(s) of protection by vaccination with live attenuated SIV must account for the heterologous and early protection data described in this study, including those which relate to the innate immune system
