Stabilization Of Porous Chitosan Improves The Performance Of Its Association With Platelet-rich Plasma As A Composite Scaffold.
This study offers innovative perspectives for optimizing of scaffolds based on correlation structure-function aimed the regenerative medicine. Thus, we evaluated in vitro performance of stabilized porous chitosan (SPCHTs) associated with activated platelet-rich plasma (aP-PRP) as a composite scaffold for the proliferation and osteogenic differentiation of human adipose-derived mesenchymal stem cells (h-AdMSCs). The porous structure of chitosan (PCHT) was prepared similarly to solid sponges by controlled freezing (-20°C) and lyophilization of a 3% (w/v) chitosan solution. Stabilization was performed by treating the PCHT with sodium hydroxide (TNaOH), an ethanol series (TEtOH) or by crosslinking with tripolyphosphate (CTPP). The aP-PRP was obtained from the controlled centrifugation of whole blood and activated with autologous serum and calcium. Imaging of the structures showed fibrin networks inside and on the surface of SPCHTs as a consequence of electrostatic interactions. SPCHTs were non-cytotoxic, and the porosity, pore size and Young's modulus were approximately 96%, 145μm and 1.5MPa for TNaOH and TEtOH and 94%, 110μm and 1.8MPa for CTPP, respectively. Stabilization maintained the integrity of the SPCHTs for at least 10days of cultivation. SPCHTs showed controlled release of the growth factors TGF-β1 and PDGF-AB. Although generating different patterns, all of the stabilization treatments improved the proliferation of seeded h-AdMSCs on the composite scaffold compared to aP-PRP alone, and differentiation of the composite scaffold treated with TEtOH was significantly higher than for non-stabilized PCHT. We conclude that the composite scaffolds improved the in vitro performance of PRP and have potential in regenerative medicine.60538-54

Long-term persistence of oligoclonal serum IgM repertoires in patients treated with allogeneic bone marrow transplantation (BMT)
Immunoglobulin gene rearrangements in patients treated with BMT have restricted repertoire diversity. Clonal variability remains low for 3 months and reconstitution of the humoral immune system appears to follow a wave-like pattern. In the present study we analysed serum IgM and IgG repertoires in 44 patients from 1 week to 3 years after transplantation. We applied a quantitative immunoblot technique in combination with a newly developed method for estimation of repertoire diversity in complex mixtures of antibodies. Our results demonstrate that 60% of BMT patients have severely reduced diversity in the IgM repertoire during and after the first year post-BMT, compared with healthy controls. In contrast, the majority of patients have a polyclonal IgG repertoire, similar to that of healthy controls. Serum IgM repertoires remain oligoclonal even though the serum concentration of total IgM is within normal range around 6 months post-BMT. During the first years after transplantation IgM as well as IgG repertoires are less diverse in patients receiving a BM graft from a sibling donor compared with those receiving a graft from an HLA-matched unrelated donor. Patients in the latter group show a higher incidence of infections and minor antigen mismatches which may promote the development of a diverse immunoglobulin repertoire post-BMT
Ayahuasca e redução do uso abusivo de psicoativos: eficácia terapêutica?
Trata-se de uma avaliação do possível papel do uso da ayahuasca, em contexto religioso, como auxiliar na redução do consumo abusivo de psicoativos, a partir de uma pesquisa de estudo de caso. Foi realizada uma entrevista aberta com uma usuária regular de cocaína, nicotina e álcool que abandonou este comportamento após entrar em contato com a ayahuasca num contexto ritualizado. O caso foi analisado à luz da comparação deste com a literatura existente sobre o assunto. Foi traçada uma relação entre o início do uso da ayahuasca e o abandono do uso de cocaína, nicotina e álcool pela entrevistada, a partir da avaliação das representações simbólicas e das descrições de suas primeiras experiências com a bebida
