Growing our own - recruiting high quality research students by stealth
Most design students assume that their education is preparing them for a career in professional practice. Until recently the idea of working as researchers or studying for a research degree was not an option.
In the early 1990s, the Art and Design Research Centre at Sheffield Hallam University was set up with a focus on "designerly" research. At that time we did not have a ready supply of suitable experienced researchers, and design graduates were not coming forward as candidates for research degrees.
The university's policy at the time required us to include a substantial research methods element in all postgraduate courses, including the vocationally focused MA Design programme. As a result of adopting this policy in a very whole-hearted way, making research methods central to the MA and framing the creative practice of design as both research-driven and investigative, we have "subverted" a number of postgraduate students who might never have considered a research degree, but are now registered for, or in the process of transferring to, PhD studies.
This paper describes the features of the MA programme that foster an investigative culture and provides some examples of PhD students whose research has grown from opportunities encountered in the MA. It also discusses the relationship between professional practice and research and the ways in which a research-centred education can prepare graduates for professional leadership. </p
Tauroursodeoxycholic acid exerts anticholestatic effects by a cooperative cPKC alpha-/PKA-dependent mechanism in rat liver.
Objective: Ursodeoxycholic acid (UDCA) exerts anticholestatic effects in part by protein kinase C (PKC)-dependent mechanisms. Its taurine conjugate, TUDCA, is a cPKCa agonist. We tested whether protein kinase A (PKA) might contribute to the anticholestatic action of TUDCA via cooperative cPKCa-/PKA-dependent mechanisms
in taurolithocholic acid (TLCA)-induced cholestasis.
Methods: In perfused rat liver, bile flow was determined gravimetrically, organic anion secretion spectrophotometrically,
lactate dehydrogenase (LDH) release enzymatically, cAMP response-element binding protein (CREB) phosphorylation by immunoblotting, and cAMP by immunoassay. PKC/PKA inhibitors were tested radiochemically. In vitro phosphorylation of the conjugate export pump, Mrp2/Abcc2, was studied in rat hepatocytes and human Hep-G2 hepatoma cells.
Results: In livers treated with TLCA (10 mmol/l)+TUDCA (25 mmol/l), combined inhibition of cPKC by the cPKCselective
inhibitor Go¨6976 (100 nmol/l) or the nonselective PKC inhibitor staurosporine (10 nmol/l) and of PKA by H89 (100 nmol/l) reduced bile flow by 36% (p,0.05) and 48% (p,0.01), and secretion of the Mrp2/
Abcc2 substrate, 2,4-dinitrophenyl-S-glutathione, by 31% (p,0.05) and 41% (p,0.01), respectively; bile flow was
unaffected in control livers or livers treated with TUDCA only or TLCA+taurocholic acid. Inhibition of cPKC or PKA alone did not affect the anticholestatic action of TUDCA. Hepatic cAMP levels and CREB phosphorylation as readout of PKA activity were unaffected by the bile acids
tested, suggesting a permissive effect of PKA for the anticholestatic action of TUDCA. Rat and human hepatocellular Mrp2 were phosphorylated by phorbol ester pretreatment and recombinant cPKCa, nPKCe, and PKA, respectively, in a staurosporine-sensitive manner.
Conclusion: UDCA conjugates exert their anticholestatic action in bile acid-induced cholestasis in part via cooperative post-translational cPKCa-/PKA-dependent
mechanisms. Hepatocellular Mrp2 may be one target of bile acid-induced kinase activation
Botulinum neurotoxin type C protease induces apoptosis in differentiated human neuroblastoma cells
Neuroblastomas constitute a major cause of cancer-related deaths in young children. In recent years, a number of translation-inhibiting enzymes have been evaluated for killing neuroblastoma cells. Here we investigated the potential vulnerability of human neuroblastoma cells to protease activity derived from botulinum neurotoxin type C. We show that following retinoic acid treatment, human neuroblastoma cells, SiMa and SH-SY5Y, acquire a neuronal phenotype evidenced by axonal growth and expression of neuronal markers. Botulinum neurotoxin type C which cleaves neuron-specific SNAP25 and syntaxin1 caused apoptotic death only in differentiated neuroblastoma cells. Direct comparison of translation-inhibiting enzymes and the type C botulinum protease revealed one order higher cytotoxic potency of the latter suggesting a novel neuroblastoma-targeting pathway. Our mechanistic insights revealed that loss of ubiquitous SNAP23 due to differentiation coupled to SNAP25 cleavage due to botulinum activity may underlie the apoptotic death of human neuroblastoma cells
Can Streamer Blobs prevent the Buildup of the Interplanetetary Magnetic Field?
Coronal Mass Ejections continuously drag closed magnetic field lines away
from the Sun, adding new flux to the interplanetary magnetic field (IMF). We
propose that the outward-moving blobs that have been observed in helmet
streamers are evidence of ongoing, small-scale reconnection in streamer current
sheets, which may play an important role in the prevention of an indefinite
buildup of the IMF. Reconnection between two open field lines from both sides
of a streamer current sheet creates a new closed field line, which becomes part
of the helmet, and a disconnected field line, which moves outward. The blobs
are formed by plasma from the streamer that is swept up in the trough of the
outward moving field line. We show that this mechanism is supported by
observations from SOHO/LASCO. Additionally, we propose a thorough statistical
study to quantify the contribution of blob formation to the reduction of the
IMF, and indicate how this mechanism may be verified by observations with
SOHO/UVCS and the proposed NASA STEREO and ESA Polar Orbiter missions.Comment: 7 pages, 2 figures; accepted by The Astrophysical Journal Letters;

Signal specific electric potential sensors for operation in noisy environments
Limitations on the performance of electric potential sensors are due to saturation caused by environmental electromagnetic noise. The work described involves tailoring the response of the sensors to reject the main components of the noise, thereby enhancing both the effective dynamic range and signal to noise. We show that by using real-time analogue signal processing it is possible to detect a human heartbeat at a distance of 40 cm from the front of a subject in an unshielded laboratory. This result has significant implications both for security sensing and biometric measurements in addition to the more obvious safety related applications
An exploratory study of how a Korean chaebol’s learning culture facilitates customer service
This paper explains how the organizational learning concept is used by managers in a global Korean company to promote group work, information sharing and an open communication style in order to produce a high level of customer service. Previously collected data from a set of in-depth personal interviews undertaken with three senior managers in a Korean electronics company were analyzed and interpreted using the grounded theory approach, and a number of propositions are put forward. The research findings show that managers in a chaebol deploy organizational learning to identify skilled and knowledgeable staff, and improve the organization’s capability by placing emphasis on developing harmonious, mutually oriented relationships that permeate throughout the organization. Top management demand that staff identify with government economic objectives and align the organization’s strategy accordingly so that the products produced are marketable. To achieve this, the organization fosters continual interaction among managers throughout the organization’s hierarchy. The chaebol’s organizational learning model encapsulates a “corollary” (continual communication) and “tools” (cultural influence and relationship management), and manifests in a unique strategy that allows management systems to evolve naturally
Bioclipse: an open source workbench for chemo- and bioinformatics
BACKGROUND: There is a need for software applications that provide users with a complete and extensible toolkit for chemo- and bioinformatics accessible from a single workbench. Commercial packages are expensive and closed source, hence they do not allow end users to modify algorithms and add custom functionality. Existing open source projects are more focused on providing a framework for integrating existing, separately installed bioinformatics packages, rather than providing user-friendly interfaces. No open source chemoinformatics workbench has previously been published, and no sucessful attempts have been made to integrate chemo- and bioinformatics into a single framework. RESULTS: Bioclipse is an advanced workbench for resources in chemo- and bioinformatics, such as molecules, proteins, sequences, spectra, and scripts. It provides 2D-editing, 3D-visualization, file format conversion, calculation of chemical properties, and much more; all fully integrated into a user-friendly desktop application. Editing supports standard functions such as cut and paste, drag and drop, and undo/redo. Bioclipse is written in Java and based on the Eclipse Rich Client Platform with a state-of-the-art plugin architecture. This gives Bioclipse an advantage over other systems as it can easily be extended with functionality in any desired direction. CONCLUSION: Bioclipse is a powerful workbench for bio- and chemoinformatics as well as an advanced integration platform. The rich functionality, intuitive user interface, and powerful plugin architecture make Bioclipse the most advanced and user-friendly open source workbench for chemo- and bioinformatics. Bioclipse is released under Eclipse Public License (EPL), an open source license which sets no constraints on external plugin licensing; it is totally open for both open source plugins as well as commercial ones. Innovate to eliminate: a prerequisite in NTD programmes
Innovation plays a critical role in progress towards achievement of the World Health Organization's road map for neglected tropical diseases 2021-2030. As disease prevalence decreases, the cost to identify and treat remaining cases goes up. Additionally, as programmes move to the surveillance phase, diagnostic tests need to be highly sensitive and affordable. Until the early end to the Ascend West and Central Africa programme, the Ascend Learning and Innovation Fund supported five projects from 2019 to 2021. Designed for innovation, the fund encompassed a range of activities, including operational research, product development and social behavioural change. This flexibility allowed innovation to bridge the gap between strategic policy and practical implementation, piloting and proving business models to respond to information found through Ascend
Characteristics of improvements in balance control using vibro-tactile biofeedback of trunk sway for multiple sclerosis patients
Background and aims: Previously, we determined that training with vibrotactile feedback (VTfb) of trunk sway improves MS patients’ balance impairment. Here, we posed 5 questions: 1) How many weeks of VTfb training are required to obtain the best short-term carry over effect (CoE) with VTfb? 2) How long does the CoE last once VTfb training terminates? 3) Is the benefit similar for stance and gait? 4) Is position or velocity based VTfb more effective in reducing trunk sway? 5) Do patients’ subjective assessments of balance control improve? Methods: Balance control of 16 MS patients was measured with gyroscopes at the lower trunk. The gyroscopes drove directionally active VTfb in a head-band. Patients trained twice per week with VTfb for 4 weeks to determine when balance control with and without VTfb stopped improving. Thereafter, weekly assessments without VTfb over 4 weeks and at 6 months determined when CoEs ended. Results: A 20% improvement in balance to normal levels occurred with VTfb. Short term CoEs improved from 15 to 20% (p ≤0.001). Medium term (1–4 weeks) CoEs were constant at 19% (p ≤0.001). At 6 months improvement was not significant, 9%. Most improvement was for lateral sway. Equal improvement occurred when angle position or velocity drove VTfb. Subjectively, balance improvements peaked after 3 weeks of training (32%, p ≤0.05). Conclusions: 3–4 weeks VTfb training yields clinically relevant sway reductions and subjective improvements for MS patients during stance and gait. The CoEs lasted at least 1 month. A Parametric Study of Erupting Flux Rope Rotation. Modeling the "Cartwheel CME" on 9 April 2008
The rotation of erupting filaments in the solar corona is addressed through a
parametric simulation study of unstable, rotating flux ropes in bipolar
force-free initial equilibrium. The Lorentz force due to the external shear
field component and the relaxation of tension in the twisted field are the
major contributors to the rotation in this model, while reconnection with the
ambient field is of minor importance. Both major mechanisms writhe the flux
rope axis, converting part of the initial twist helicity, and produce rotation
profiles which, to a large part, are very similar in a range of shear-twist
combinations. A difference lies in the tendency of twist-driven rotation to
saturate at lower heights than shear-driven rotation. For parameters
characteristic of the source regions of erupting filaments and coronal mass
ejections, the shear field is found to be the dominant origin of rotations in
the corona and to be required if the rotation reaches angles of order 90
degrees and higher; it dominates even if the twist exceeds the threshold of the
helical kink instability. The contributions by shear and twist to the total
rotation can be disentangled in the analysis of observations if the rotation
and rise profiles are simultaneously compared with model calculations. The
resulting twist estimate allows one to judge whether the helical kink
instability occurred. This is demonstrated for the erupting prominence in the
"Cartwheel CME" on 9 April 2008, which has shown a rotation of \approx 115
degrees up to a height of 1.5 R_sun above the photosphere. Out of a range of
initial equilibria which include strongly kink-unstable (twist Phi=5pi), weakly
kink-unstable (Phi=3.5pi), and kink-stable (Phi=2.5pi) configurations, only the
evolution of the weakly kink-unstable flux rope matches the observations in
