    Neuroprotective effect of Canthium parviflorum on streptozotocin induced diabetic neuropathy in rats

    45-51In traditional system to cure diabetes and related complications, Canthium parviflorum Lam plant leaves are used. Ethanolic extract of leaves were considered to investigate its neuroprotective property in streptozotocin elicited diabetic neuropathy rat model. Streptozotocin (60 mg/kg body weight) given by intraperitoneal injection to induce diabetes, with no treatment given, was intended to develop diabetic neuropathy. Fourteen days after induction, animals were given the extract (100 and 200 mg/kg) once daily for thirty five days. Hypersensitivity to temperature in diabetic neuropathy was assessed by tail immersion and eddy’s hot plate tests. Nerve growth factor, blood glucose, and pro-inflammatory cytokines involved in pathological pain were measured in sciatic nerve. Neuropathy which is also associated with oxidative stress was assessed by measuring bio-markers in sciatic tissue. Diabetic induced neuropathic animals exhibited significant decrease in the paw and tail withdraw time thus signifying diabetic neuropathy. Extract treatment prevented diabetic neuropathy in animals thus showing normalized response to hot and cold. These actions in extract treatment groups are supported evidently by normal blood glucose, significant low levels of pro-inflammatory cytokines, oxidative stress bio-markers, and high nerve growth factor in sciatic nerve tissue.     HyFlex pedagogy: six strategies supported by design-based research

    Purpose–This study investigates the following research question: What pedagogical strategies are necessary for the success of the project? The findings to this question are based in new media literacies and help to further pedagogy in an emerging HyFlex model while also grounding in needed theorization. Design/methodology/approach–This study uses design-based research(DBR) across two iterations and four doctoral, higher education courses, using mixed methods of data collection and analysis. Findings–Six pedagogical strategies influential for HyFlex research are presented, each grounded in a new media literacy skill. Originality/value–These six pedagogical strategies help practitioners grappling with the HyFlex or blended learning model merge traditional pedagogy with how this might be tailored for students entrenched in a participatory culture

    Strengths and Weaknesses for Climate Change: Adaptation in Water Governance: A Comparison Across Six European Regions

    This chapter comparatively analyses the policy and governance contexts of six European regions that are affected by different hydrological impacts of climate change. The results demonstrate that a major governance strength across regions lies in the organization of management capacities to deal with existing water-related risks. For example, the Dutch context focuses on water safety, Cyprus has a clear policy framework to deal with water scarcity and in the Norwegian city of Bergen, wastewater is well managed. As a consequence of this focus on present-day risks, climate adaptation governance also focuses on historical risks. New or exacerbated risks posed by climate change remain largely untreated, and responsibilities for dealing with climate-related risks remain unspecified, as also becomes clear in the German and Spanish cases. A high degree of governmental fragmentation is identified as another point of weakness. Identified most clearly in the Portuguese case but recognizable in all regional contexts, different subdomains of water management are dealt with under separate policies and are governed by different responsible agencies. Consequently, information about current performance of the water system is scattered and coordinative efforts, which are key to developing adaptation strategies, are hampered

    The feasibility of milkfish (Chanos chanos) aquaculture in Solomon Islands

    Fish is crucial to food and nutrition security in Solomon Islands, and demand is expected to increase due to a growing population. However, it is projected that current capture fisheries production will not meet this growing demand. Aquaculture has the potential to mitigate the capture fishery shortfall, and the Government of Solomon Islands is prioritizing aquaculture as a solution to meet future food and income needs. Aquaculture in Solomon Islands is still in early development. Mozambique tilapia (Oreochromis mossambicus) is farmed for household consumption, but its prolific reproductive rate and resulting slow growth limit its potential as a commercial aquaculture species. More productive fish species that are not indigenous to Solomon Islands but are successfully farmed overseas could be introduced; however, such a decision needs to take into account the potential ecological or social impacts. For land-based pond aquaculture, the only indigenous species that has been farmed extensively elsewhere is milkfish (Chanos chanos). This report presents a feasibility assessment for milkfish farming in Solomon Islands. It synthesizes the current knowledge about milkfish farming and presents results of a 4-year study on the potential for milkfish aquaculture in Solomon Islands

    Sequential action of ATPase, ATP, ADP, Pi and dsDNA in procapsid-free system to enlighten mechanism in viral dsDNA packaging

    Many cells and double-stranded DNA (dsDNA) viruses contain an AAA+ ATPase that assembles into oligomers, often hexamers, with a central channel. The dsDNA packaging motor of bacteriophage phi29 also contains an ATPase to translocate dsDNA through a dodecameric channel. The motor ATPase has been investigated substantially in the context of the entire procapsid. Here, we report the sequential action between the ATPase and additional motor components. It is suggested that the contact of ATPase to ATP resulted in its conformational change to a higher binding affinity toward dsDNA. It was found that ATP hydrolysis led to the departure of dsDNA from the ATPase/dsDNA complex, an action that is speculated to push dsDNA to pass the connector channel. Our results suggest that dsDNA packaging goes through a combined effort of both the gp16 ATPase for pushing and the channel as a one-way valve to control the dsDNA translocation direction. Many packaging models have previously been proposed, and the packaging mechanism has been contingent upon the number of nucleotides packaged per ATP relative to the 10.5 bp per helical turn for B-type dsDNA. Both 2 and 2.5 bp per ATP have been used to argue for four, five or six discrete steps of dsDNA translocation. Combination of the two distinct roles of gp16 and connector renews the perception of previous dsDNA packaging energy calculations and provides insight into the discrepancy between 2 and 2.5 bp per ATP

    Fermented wheat germ extract - nutritional supplement or anticancer drug?

    <p>Abstract</p> <p>Background</p> <p>Fermented wheat germ extract (FWGE) is a multisubstance composition and, besides others, contains 2-methoxy benzoquinone and 2, 6-dimethoxy benzoquinone which are likely to exert some of its biological effects. FWGE interferes with anaerobic glycolysis, pentose cycle and ribonucleotide reductase. It has significant antiproliferative effects and kills tumor cells by the induction of apoptosis via the caspase-poly [ADP-ribose] polymerase-pathway. FWGE interacts synergistically with a variety of different anticancer drugs and exerted antimetastatic properties in mouse models. In addition, FWGE modulates immune response by downregulation of MHC-I complex and the induction of TNF-α and various interleukins. Data in the F-344 rat model provide evidence for a colon cancer preventing effect of FWGE.</p> <p>Clinical data from a randomized phase II trial in melanoma patients indicate a significant benefit for patients treated with dacarbazine in combination with FWGE in terms of progression free survival (PFS) and overall survival (OS). Similarly, data from studies in colorectal cancer suggested a benefit of FWGE treatment. Besides extension of OS and PFS, FWGE improved the quality of life in several studies.</p> <p>Conclusion</p> <p>In conclusion, available data so far, justify the use of FWGE as a non-prescription medical nutriment for cancer patients. Further randomized, controlled and large scale clinical studies are mandatory, to further clarify the value of FWGE as a drug component of future chemotherapy regimens.</p

    EMT transcription factors snail and slug directly contribute to cisplatin resistance in ovarian cancer

    <p>Abstract</p> <p>Background</p> <p>The epithelial to mesenchymal transition (EMT) is a molecular process through which an epithelial cell undergoes transdifferentiation into a mesenchymal phenotype. The role of EMT in embryogenesis is well-characterized and increasing evidence suggests that elements of the transition may be important in other processes, including metastasis and drug resistance in various different cancers.</p> <p>Methods</p> <p>Agilent 4 × 44 K whole human genome arrays and selected reaction monitoring mass spectrometry were used to investigate mRNA and protein expression in A2780 cisplatin sensitive and resistant cell lines. Invasion and migration were assessed using Boyden chamber assays. Gene knockdown of <it>snail </it>and <it>slug </it>was done using targeted siRNA. Clinical relevance of the EMT pathway was assessed in a cohort of primary ovarian tumours using data from Affymetrix GeneChip Human Genome U133 plus 2.0 arrays.</p> <p>Results</p> <p>Morphological and phenotypic hallmarks of EMT were identified in the chemoresistant cells. Subsequent gene expression profiling revealed upregulation of EMT-related transcription factors including <it>snail, slug, twist2 </it>and <it>zeb2</it>. Proteomic analysis demonstrated up regulation of Snail and Slug as well as the mesenchymal marker Vimentin, and down regulation of E-cadherin, an epithelial marker. By reducing expression of <it>snail </it>and <it>slug</it>, the mesenchymal phenotype was largely reversed and cells were resensitized to cisplatin. Finally, gene expression data from primary tumours mirrored the finding that an EMT-like pathway is activated in resistant tumours relative to sensitive tumours, suggesting that the involvement of this transition may not be limited to <it>in vitro </it>drug effects.</p> <p>Conclusions</p> <p>This work strongly suggests that genes associated with EMT may play a significant role in cisplatin resistance in ovarian cancer, therefore potentially leading to the development of predictive biomarkers of drug response or novel therapeutic strategies for overcoming drug resistance.</p

    Grambank reveals the importance of genealogical constraints on linguistic diversity and highlights the impact of language loss

    While global patterns of human genetic diversity are increasingly well characterized, the diversity of human languages remains less systematically described. Here we outline the Grambank database. With over 400,000 data points and 2,400 languages, Grambank is the largest comparative grammatical database available. The comprehensiveness of Grambank allows us to quantify the relative effects of genealogical inheritance and geographic proximity on the structural diversity of the world's languages, evaluate constraints on linguistic diversity, and identify the world's most unusual languages. An analysis of the consequences of language loss reveals that the reduction in diversity will be strikingly uneven across the major linguistic regions of the world.
