    Newly identified LMO3-BORCS5 fusion oncogene in Ewing sarcoma at relapse is a driver of tumor progression

    International audienceRecently, we detected a new fusion transcript LMO3-BORCS5 in a patient with Ewing sarcoma within a cohort of relapsed pediatric cancers. LMO3-BORCS5 was as highly expressed as the characteristic fusion oncogene EWS/FLI1. However, the expression level of LMO3-BORCS5 at diagnosis was very low. Sanger sequencing depicted two LMO3-BORCS5 variants leading to loss of the functional domain LIM2 in LMO3 gene, and disruption of BORCS5. In vitro studies showed that LMO3-BORCS5 (i) increases proliferation, (ii) decreases expression of apoptosis-related genes and treatment sensitivity, and (iii) downregulates genes involved in differentiation and upregulates proliferative and extracellular matrix-related pathways. Remarkably, in vivo LMO3-BORCS5 demonstrated its high oncogenic potential by inducing tumors in mouse fibroblastic NIH-3T3 cell line. Moreover, BORCS5 probably acts, in vivo, as a tumor-suppressor gene. In conclusion, functional studies of fusion oncogenes at relapse are of great importance to define mechanisms involved in tumor progression and resistance to conventional treatments

    LITL at CLEF eHealth2016: recognizing entities in French biomedical documents

    International audienceThis paper describes the participation of master's students (LITL programme, university of Toulouse) and their teachers to the CLEF eHealth 2016 campaign. Two runs were submitted for task 2 (multilingual information extraction) which consisted in the recognition and categorization of medical entities in French biomedical documents. The system used consists of a CRF classier based on a number of dierent features (POS tagging, generic word lists and syntactic parsing). In addition , several patterns were used on the CRF's output in order to extract more complex entities. The best run achieved high precision (0.640.78) but lower recall (0.320.40), with an overall F1-measure of 0.430.53

    Rosmarinic Acid and Its Methyl Ester as Antimicrobial Components of the Hydromethanolic Extract of Hyptis atrorubens

    Primary biological examination of four extracts of the leaves and stems of Hyptis atrorubens Poit. (Lamiaceae), a plant species used as an antimicrobial agent in Guadeloupe, allowed us to select the hydromethanolic extract of the stems for further studies. It was tested against 46 microorganisms in vitro. It was active against 29 microorganisms. The best antibacterial activity was found against bacteria, mostly Gram-positive ones. Bioautography enabled the isolation and identification of four antibacterial compounds from this plant: rosmarinic acid, methyl rosmarinate, isoquercetin, and hyperoside. The MIC and MBC values of these compounds and their combinations were determined against eight pathogenic bacteria. The best inhibitory and bactericidal activity was found for methyl rosmarinate (0.3 mg/mL). Nevertheless, the bactericidal power of rosmarinic acid was much faster in the time kill study. Synergistic effects were found when combining the active compounds. Finally, the inhibitory effects of the compounds were evaluated on the bacterial growth phases at two different temperatures. Our study demonstrated for the first time antimicrobial activity of Hyptis atrorubens with identification of the active compounds. It supports its traditional use in French West Indies. Although its active compounds need to be further evaluated in vivo, this work emphasizes plants as potent sources of new antimicrobial agents when resistance to antibiotics increases dramatically

    Spatiotemporal changes in surface sediment characteristics and benthic macrofauna composition off the Rhône River in relation to its hydrological regime

    International audienceThe consequences of changes in the water flow of the Rhône River on surface sediment characteristics and benthic macrofauna composition were assessed within 3 distinct areas: (1) the delta front, (2) the prodelta, and (3) the distal zone. Five stations were sampled during or closely after: (1) an oceanic flood (April 2007), (2) a generalized flood (May 2008), (3) a Cevenol flood (December 2008), and (4) a dry period (July 2011). Measurements of sediment characteristics included granulometry (D0.5), bulk descriptors of sedimentary organics (OC, TN and THAA), descriptors of labile components of sedimentary organics (chloropigments, EHAA), and both descriptors of origin (Chl-b/Chl-a, C/N) and lability (Chl-a/(Chl-a+Phaeo-a), EHAA/THAA) of sedimentary organics. Sediment Profile Images were collected during April 2007, May 2008 and July 2011. Temporal changes in both sedimentary organics and benthic macrofauna were more important in the delta front and the prodelta than in the distal zone. Bulk characteristics of sedimentary organics presented decreasing inshore/offshore gradients during both April 2007 and July 2011 but not during May and December 2008. There were significant temporal changes in EHAA/THAA at all stations. Changes in benthic macrofauna composition differed between: (1) the delta front and the prodelta, and (2) the distal zone. In the former area, the dry period was associated with establishing a mature community characterized by high abundances and species richness. The best description of spatiotemporal changes in benthic macrofauna composition by surface sediment characteristics was obtained using D0.5, Chl-b/Chl-a, Chl-a/(Chl-a+Phaeo-a) and EHAA, which supports the role of the quality of sedimentary organics in controlling benthic macrofauna composition

    Systemic sclerosis (SSc) is an autoimmune connective tissue disorder, characterized by multisystem involvement, vasculopathy, and fibrosis. An increased risk of malignancy is observed in SSc (including breast and lung cancers), and in a subgroup of patients with specific autoantibodies (i.e., anti-RNA polymerase III and related autoantibodies), SSc could be a paraneoplastic syndrome and might be directly related to an immune response against cancer. Herein, we reviewed the literature, focusing on the most recent articles, and shed light onto the potential relationship between cancer and scleroderma regarding temporal and immunological dimensions

    Perfectionism, rumination, and gender are related to symptoms of eating disorders: A moderated mediation model

    Introduction Perfectionism and rumination both seem to be involved in the development and maintenance of eating disorders (ED), but the underlying studies have been performed almost exclusively in females. The aims of this study were to examine whether rumination is a potential mediator of the link between perfectionism and ED symptoms, and whether gender is a moderator of the link between perfectionism and ED symptoms and/or between perfectionism and brooding rumination. Method 390 participants (269 women, 121 men), aged between 18 and 25, completed three questionnaires: the Ruminative Response Scale for Eating Disorders, the Eating Attitudes Test, and the Frost Multidimensional Perfectionism Scale. Results Results showed that maladaptive evaluative-concerns perfectionism and positive strivings perfectionism both had indirect effects on ED symptoms via brooding rumination. Gender moderated only the direct effect between maladaptive evaluative-concerns perfectionism and ED symptoms. Conclusions These results highlighted gender differences, and support the literature on the importance of including rumination and perfectionism in the treatment and the prevention of eating disorders

    Effects of induced rumination on body dissatisfaction: Is there any difference between men and women?

    BACKGROUND AND OBJECTIVES: Rumination is a factor in the development and maintenance of body dissatisfaction. However, no study has yet investigated the impact of the type of rumination on body image. The first aim of this study was to examine whether the induction of analytic-abstract vs. concrete-experiential rumination affects body dissatisfaction following an induction of negative body image. The second objective was to examine gender differences in these effects. METHODS: Following induction of negative body image, 102 university undergraduates were randomly assigned to one of three experimental conditions-distraction, concrete rumination or abstract rumination. RESULTS: As expected, there were significant main effects of gender and condition, and a significant interaction between gender and condition on change in body dissatisfaction. In women abstract rumination predicted the highest increase in body dissatisfaction, whereas concrete rumination predicted the highest increase in body dissatisfaction in men. LIMITATIONS: Given that our sample consisted of undergraduate students, our findings cannot be generalized to clinical sample suffering from eating disorder. CONCLUSIONS: The different types of rumination seem to impact differentially body dissatisfaction in men and women

    C, N and P stoichiometric mismatch between resources and consumers influence the dynamics of a marine microbial food web model and its response to atmospheric N and P inputs

    International audienceResults from the DUNE experiments reported in this issue have shown that nutrient input from dust deposition in large mesocosms deployed in the western Mediterranean induced a response of the microbial food web, with an increase of primary production rates (PP), bacterial respiration rates (BR), as well as autotrophic and heterotrophic biomasses. Additionally, it was found that nutrient inputs strengthened the net heterotrophy of the system, with NPP : BR ratios < 1. In this study we used a simple microbial food web model, inspired from previous modelling studies, to explore how C, N and P stoichiometric mismatch between producers and consumers along the food chain can influence the dynamics and the trophic status of the ecosystem. Attention was paid to the mechanisms involved in the balance between net autotrophy vs. net heterotrophy. Although the model was kept simple, predicted changes in biomass and PP were qualitatively consistent with observations from DUNE experiments. Additionally, the model shed light on how ecological stoichiometric mismatch between producers and consumers can control food web dynamics and drive the system toward net heterotrophy. In the model, net heterotrophy was notably driven by the parameterisation of the production and excretion of extra DOC from phytoplankton under nutrient-limited conditions. This mechanism yielded to high C : P and C : N ratios of the DOM pool, and subsequent postabsorptive respiration of C by bacteria. The model also predicted that nutrient inputs from dust strengthened the net heterotrophy of the system; a pattern also observed during two of the three DUNE experiments (P and Q). However, the model was not able to account for the low NPP : BR ratios (down to 0.1) recorded during the DUNE experiments. Possible mechanisms involved in this discrepancy were discussed
