    A new wrinkle on liquid sheets: Turning the mechanism of viscous bubble collapse upside down

    Viscous bubbles are prevalent in both natural and industrial settings. Their rupture and collapse may be accompanied by features typically associated with elastic sheets, including the development of radial wrinkles. Previous investigators concluded that the film weight is responsible for both the film collapse and wrinkling instability. Conversely, we show here experimentally that gravity plays a negligible role: The same collapse and wrinkling arise independently of the bubble's orientation. We found that surface tension drives the collapse and initiates a dynamic buckling instability. Because the film weight is irrelevant, our results suggest that wrinkling may likewise accompany the breakup of relatively small-scale, curved viscous and viscoelastic films, including those in the respiratory tract responsible for aerosol production from exhalation events.Accepted manuscrip

    Introduction to focus issue on hydrodynamic quantum analogs

    Hydrodynamic quantum analogs is a nascent field initiated in 2005 by the discovery of a hydrodynamic pilot-wave system [Y. Couder, S. Protière, E. Fort, and A. Boudaoud, Nature 437, 208 (2005)]. The system consists of a millimetric droplet self-propeling along the surface of a vibrating bath through a resonant interaction with its own wave field [J. W. M. Bush, Annu. Rev. Fluid Mech. 47, 269-292 (2015)]. There are three critical ingredients for the quantum like-behavior. The first is “path memory” [A. Eddi, E. Sultan, J. Moukhtar, E. Fort, M. Rossi, and Y. Couder, J. Fluid Mech. 675, 433-463 (2011)], which renders the system non-Markovian: the instantaneous wave force acting on the droplet depends explicitly on its past. The second is the resonance condition between droplet and wave that ensures a highly structured monochromatic pilot wave field that imposes an effective potential on the walking droplet, resulting in preferred, quantized states. The third ingredient is chaos, which in several systems is characterized by unpredictable switching between unstable periodic orbits. This focus issue is devoted to recent studies of and relating to pilot-wave hydrodynamics, a field that attempts to answer the following simple but provocative question: Might deterministic chaotic pilot-wave dynamics underlie quantum statistics?</p

    Characterization and identification of dityrosine cross-linked peptides using tandem mass spectrometry

    The use of mass spectrometry coupled with chemical cross-linking of proteins has become a powerful tool for proteins structure and interactions studies. Unlike structural analysis of proteins using chemical reagents specific for lysine or cysteine residues, identification of gas-phase fragmentation patterns of endogenous dityrosine cross-linked peptides have not been investigated. Dityrosine cross-linking in proteins and peptides are clinical markers of oxidative stress, aging, and neurodegenerative diseases including Alzheimer’s disease and Parkinson’s disease. In this study, we investigated and characterized the fragmentation pattern of a synthetically prepared dityrosine cross-linked dimer of Aβ(1–16) using ESI tandem mass spectrometry. We then detailed the fragmentation pattern of dityrosine cross-linked Aβ(1–16), using collision induced dissociation (CID), higher-energy collision induced dissociation (HCD), electron transfer dissociation (ETD), and electron capture dissociation (ECD). Application of these generic fragmentation rules of dityrosine cross-linked peptides allowed for the identification of dityrosine cross-links in peptides of Aβ and α-synuclein generated in vitro by enzymatic peroxidation. We report, for the first time, the dityrosine cross-linked residues in human hemoglobin and α-synuclein under oxidative conditions. Together these tools open up the potential for automated analysis of this naturally occurring post-translation modification in neurodegenerative diseases as well as other pathological conditions

    Epithelial IL-6 trans-signaling defines a new asthma phenotype with increased airway inflammation

    Background: Although several studies link high levels of IL-6 and soluble IL-6 receptor (sIL-6R) to asthma severity and decreased lung function, the role of IL-6 trans-signaling (IL-6TS) in asthmatic patients is unclear. Objective: We sought to explore the association between epithelial IL-6TS pathway activation and molecular and clinical phenotypes in asthmatic patients. Methods: An IL-6TS gene signature obtained from air-liquid interface cultures of human bronchial epithelial cells stimulated with IL-6 and sIL-6R was used to stratify lung epithelial transcriptomic data (Unbiased Biomarkers in Prediction of Respiratory Disease Outcomes [U-BIOPRED] cohorts) by means of hierarchical clustering. IL-6TS-specific protein markers were used to stratify sputum biomarker data (Wessex cohort). Molecular phenotyping was based on transcriptional profiling of epithelial brushings, pathway analysis, and immunohistochemical analysis of bronchial biopsy specimens. Results: Activation of IL-6TS in air-liquid interface cultures reduced epithelial integrity and induced a specific gene signature enriched in genes associated with airway remodeling. The IL-6TS signature identified a subset of patients with IL-6TS-high asthma with increased epithelial expression of IL-6TS-inducible genes in the absence of systemic inflammation. The IL-6TS-high subset had an overrepresentation of frequent exacerbators, blood eosinophilia, and submucosal infiltration of T cells and macrophages. In bronchial brushings Toll-like receptor pathway genes were upregulated, whereas expression of cell junction genes was reduced. Sputum sIL-6R and IL-6 levels correlated with sputum markers of remodeling and innate immune activation, in particular YKL-40, matrix metalloproteinase 3, macrophage inflammatory protein 1 beta, IL-8, and IL-1 beta. Conclusions: Local lung epithelial IL-6TS activation in the absence of type 2 airway inflammation defines a novel subset of asthmatic patients and might drive airway inflammation and epithelial dysfunction in these patients.Peer reviewe

    New insights into the genetic etiology of Alzheimer's disease and related dementias.

    Characterization of the genetic landscape of Alzheimer's disease (AD) and related dementias (ADD) provides a unique opportunity for a better understanding of the associated pathophysiological processes. We performed a two-stage genome-wide association study totaling 111,326 clinically diagnosed/'proxy' AD cases and 677,663 controls. We found 75 risk loci, of which 42 were new at the time of analysis. Pathway enrichment analyses confirmed the involvement of amyloid/tau pathways and highlighted microglia implication. Gene prioritization in the new loci identified 31 genes that were suggestive of new genetically associated processes, including the tumor necrosis factor alpha pathway through the linear ubiquitin chain assembly complex. We also built a new genetic risk score associated with the risk of future AD/dementia or progression from mild cognitive impairment to AD/dementia. The improvement in prediction led to a 1.6- to 1.9-fold increase in AD risk from the lowest to the highest decile, in addition to effects of age and the APOE ε4 allele

    Parâmetros hematológicos e alterações histopatológicas em bijupirá (Rachycentron canadum Linnaeus, 1766) com amyloodiniose

    O objetivo do trabalho foi descrever os parâmetros hematológicos e as alterações histopatológicas em bijupirás infectados por Amyloodinium ocellatum. Um grupo de 27 peixes foi anestesiado para coleta de amostras de sangue e eutanasiados para coleta de muco e fragmentos de tecido cutâneo e branquial. Foram avaliadas a prevalência e a intensidade parasitária da infecção, assim como os valores de parâmetros hematológicos e alterações histopatológicas. A prevalência parasitária nas brânquias foi de 100% e no muco foi de 80,8% e as intensidades parasitárias médias foram de 683,5 nas brânquias, e 67,1 no muco cutâneo. Os valores médios dos parâmetros hematológicos foram: eritrócitos 4,3x10(6)µL; VG 26%; VGM 64,2fL; proteína plasmática 5,8mg/dL; trombócitos 5,2 x10³/µL e leucócitos 3,6 x10³/µL. Além disso, foram verificadas hiperplasia do epitélio respiratório acompanhada de fusão lamelar, descolamento do epitélio, dilatação do seio venoso, formação de aneurisma, ruptura do epitélio lamelar, hemorragia, necrose, reação inflamatória linfocítica. O parasito foi observado nas lamelas branquiais, o VMA variou do grau discreto ao severo e o IAH foi de 76,8. A pesquisa assume importância por se tratar dos primeiros estudos em Rachycentron canadum, um peixe que se destaca com potencial ao cultivo

    Droplets bouncing on a wet, inclined substrate

    We present the results of an experimental investigation of fluid drops impacting an inclined rigid surface covered with a thin layer of high viscosity fluid. We deduce the conditions under which droplet bouncing, splitting, and merger arise.     Classical pilot-wave dynamics: the free particle

    We present the results of a theoretical investigation into the dynamics of a vibrating particle propelled by its self-induced wave field. Inspired by the hydrodynamic pilot-wave system discovered by Yves Couder and Emmanuel Fort, the idealized pilot-wave system considered here consists of a particle guided by the slope of its quasi-monochromatic “pilot” wave, which encodes the history of the particle motion. We characterize this idealized pilot-wave system in terms of two dimensionless groups that prescribe the relative importance of particle inertia, drag and wave forcing. Prior work has delineated regimes in which self-propulsion of the free particle leads to steady or oscillatory rectilinear motion; it has further revealed parameter regimes in which the particle executes a stable circular orbit, confined by its pilot wave. We here report a number of new dynamical states in which the free particle executes self-induced wobbling and precessing orbital motion. We also explore the statistics of the chaotic regime arising when the time scale of the wave decay is long relative to that of particle motion and characterize the diffusive and rotational nature of the resultant particle dynamics. We thus present a detailed characterization of free-particle motion in this rich two-parameter family of dynamical systems
