    This paper examines the economy produced by modernism as the site for developing an aesthetic grounded in opulence and consumption. While early modern architects aimed to break with tradition and create a new language of architectural forms, the call for new architecture has exploded into sites of what Glen Hill calls “aesthetic waste” in his article, “Aesthetics of Architectural Consumption” (2011). In Los Angeles, this aesthetic obsolescence results in developments being demolished at an alarming rate. As the idea of beauty is valued by its proximity to cleanliness or novelty, massive homes are turned to “junk” in pursuit of the ‘new’

    EMA-amplicon-based sequencing informs risk assessment analysis of water treatment systems

    Illumina amplicon-based sequencing was coupled with ethidium monoazide bromide (EMA) pre-treatment to monitor the total viable bacterial community and subsequently identify and prioritise the target organisms for the health risk assessment of the untreated rainwater and rainwater treated using large-volume batch solar reactor prototypes installed in an informal settlement and rural farming community. Taxonomic assignments indicated that Legionella and Pseudomonas were the most frequently detected genera containing opportunistic bacterial pathogens in the untreated and treated rainwater at both sites. Additionally, Mycobacterium, Clostridium sensu stricto and Escherichia/Shigella displayed high (≥80%) detection frequencies in the untreated and/or treated rainwater samples at one or both sites. Numerous exposure scenarios (e.g. drinking, cleaning) were subsequently investigated and the health risk of using untreated and solar reactor treated rainwater in developing countries was quantified based on the presence of L. pneumophila, P. aeruginosa and E. coli. The solar reactor prototypes were able to reduce the health risk associated with E. coli and P. aeruginosa to below the 1 × 10−4 annual benchmark limit for all the non-potable uses of rainwater within the target communities (exception of showering for E. coli). However, the risk associated with intentional drinking of untreated or treated rainwater exceeded the benchmark limit (E. coli and P. aeruginosa). Additionally, while the solar reactor treatment reduced the risk associated with garden hosing and showering based on the presence of L. pneumophila, the risk estimates for both activities still exceeded the annual benchmark limit. The large-volume batch solar reactor prototypes were thus able to reduce the risk posed by the target bacteria for non-potable activities rainwater is commonly used for in water scarce regions of sub-Saharan Africa. This study highlights the need to assess water treatment systems in field trials using QMRA

    Rare-disease registries can be useful for studying the associations between environmental exposures and disease severity, but often require the addition of a healthy comparison control group. Defining a surrogate control group, matched and balanced on potentially confounding variables, would allow for the comparison of exposure distributions with cases from a registry. In the present study, we assess whether controls selected externally, using stratification score (SS) matching, can serve as effective proxies for internal controls. In addition, we use methyl paraben (MEPB) to compare the estimated associations between an externally matched sample and case–control frequencies in a cohort with internally matched controls. We started with 225 eligible cases of autism spectrum disorder (ASD) from Childhood Autism Risks from Genetics and the Environment (CHARGE), 241 internal controls from CHARGE, and 265 external controls from the National Health and Nutrition Examination Survey (NHANES) cycles 2005–2016. We calculated the SSs using demographic covariates and matched 1:1 using a caliper method without a replacement. The distribution of the covariates and the mean squared error of the paired differences (MSEpaired) in the SSs between the internal and external group were similar (MSEpaired = 0.007 and 0.011, respectively). The association between MEPB and ASD compared to the controls was similar between the externally matched SS pairs and the original frequency matched cohort. Controls selected externally, via SS matching, can provide a comparable bias reduction to that provided by the internal controls, and therefore may be a useful strategy in situations when the internal controls are not available

    Quantitative Microbial Risk Analysis for Various Bacterial Exposure Scenarios Involving Greywater Reuse for Irrigation

    Greywater reuse can significantly reduce domestic water consumption. While the benefits are promising, risks are still under debate. Using a quantitative microbial risk-assessment model, we assessed the health risks associated with greywater reuse. The pathogens Salmonella enterica, Shigella spp., and Staphylococcus aureus were evaluated due to their possible prevalence in greywater and limited information regarding their potential risk with relation to greywater reuse for irrigation. Various exposure scenarios were investigated. Monte Carlo simulation was used and results were compared to the maximum “acceptable” limit of 10−6 disability-adjusted life years (DALY) set by the World Health Organization. Safe reuse was met for all worst-case exposure scenarios for Staphylococcus aureus, Salmonella enterica and Shigella spp. If their concentrations were kept below 10,000, 50 and 5 cfu/100 mL, respectively. For the best-practice (more realistic) scenarios, safe reuse was met for Staphylococcus aureus if its concentration was kept below 106 cfu/100 mL. Salmonella enterica met the safe reuse requirements if a maximum concentration of 500 cfu/100 mL was maintained and Shigella spp. if a maximum concentration was lower than 5 cfu/100 mL. Based on reported concentrations of these bacteria in greywater, proper treatment and disinfection are recommended

    Maternal Plasma Metabolic Profile Demarcates a Role for Neuroinflammation in Non-Typical Development of Children.

    Maternal and cord plasma metabolomics were used to elucidate biological pathways associated with increased diagnosis risk for autism spectrum disorders (ASD). Metabolome-wide associations were assessed in both maternal and umbilical cord plasma in relation to diagnoses of ASD and other non-typical development (Non-TD) compared to typical development (TD) in the Markers of Autism risk in Babies: Learning Early Signs (MARBLES) cohort study of children born to mothers who already have at least one child with ASD. Analyses were stratified by sample matrix type, machine mode, and annotation confidence level. Dimensionality reduction techniques were used [i.e, principal component analysis (PCA) and random subset weighted quantile sum regression (WQSRS)] to minimize the high multiple comparison burden. With WQSRS, a metabolite mixture obtained from the negative mode of maternal plasma decreased the odds of Non-TD compared to TD. These metabolites, all related to the prostaglandin pathway, underscored the relevance of neuroinflammation status. No other significant findings were observed. Dimensionality reduction strategies provided confirming evidence that a set of maternal plasma metabolites are important in distinguishing Non-TD compared to TD diagnosis. A lower risk for Non-TD was linked to anti-inflammatory elements, thereby linking neuroinflammation to detrimental brain function consistent with studies ranging from neurodevelopment to neurodegeneration
