    Using relative sea-level data to constrain the deglacial and Holocene history of southern Greenland

    This paper presents new Holocene relative sea-level (RSL) data collected from isolation basins close to the town of Paamiut in south west Greenland. The data shows a rapid fall from a marine limit of c. 52 m asl at c. 10.9 cal. ka BP to close to present by c. 9.5 cal. ka BP, at rates of up to c. 32 mm/yr, falling below present for the majority of the Holocene before rising to present in the last 2000 years. The elevation of the RSL lowstand is not well constrained, but was at least below −3 m. This pattern of rapid RSL fall during the early Holocene matches the pattern seen at other southern Greenland locations suggesting rapid, largely simultaneous ice retreat from the area surrounding the Qassimiut Lobe at the start of the Holocene, occurring c. 2000 years after the initial deglaciation of the extreme southern tip of Greenland. The RSL histories from this and other southern Greenland locations are distinct to those recorded further north along the west coast, and are in broad agreement with a pattern of vertical land motion and RSL predicted by the Huy2 model (Simpson et al., 2009), which predicts an 80 m drop in the contribution of vertical land motion to RSL at 10 cal. ka BP between Sisimiut and Paamiut on the west coast. Despite this broad-scale spatial agreement between the RSL data and the Huy2 model, it fails to satisfactorily predict the Holocene RSL histories at Paamiut and other southern Greenland locations. Sensitivity tests indicate that the data-model misfits are most likely due to an over-estimate of the forcing during the Holocene Thermal Maximum (or the response to this forcing) in southern Greenland and error in the North American ice sheets component of the background deglaciation model. Our new data suggests that much of the southern part of the ice sheet acted differently to the area further north. However RSL changes at Paamiut are also largely impacted by regional and larger-scale processes including a bulls-eye of uplift centred on the west, the impact of the Holocene Thermal Maximum and the influence of the collapse of the North American ice sheets

    Expressions of climate perturbations in western Ugandan crater lake sediment records during the last 1000 yr

    Equatorial East Africa has a complex, regional patchwork of climate regimes, with multiple interacting drivers. Recent studies have focussed on large lakes and reveal signals that are smoothed in both space and time, and, whilst useful at a continental scale, are of less relevance when understanding short-term, abrupt or immediate impacts of climate and environmental changes. Smaller-scale studies have highlighted spatial complexity and regional heterogeneity of tropical palaeoenvironments in terms of responses to climatic forcing (e.g. the Little Ice Age [LIA]) and questions remain over the spatial extent and synchroneity of climatic changes seen in East African records. Sediment cores from paired crater lakes in western Uganda were examined to assess ecosystem response to long-term climate and environmental change as well as testing responses to multiple drivers using redundancy analysis. These archives provide annual to sub-decadal records of environmental change. The records from the two lakes demonstrate an individualistic response to external (e.g. climatic) drivers, however, some of the broader patterns observed across East Africa suggest that the lakes are indeed sensitive to climatic perturbations such as a dry Mediaeval Climate Anomaly (MCA; 1000–1200 AD) and a relatively drier climate during the main phase of the LIA (1500–1800 AD); though lake levels in western Uganda do fluctuate. The relationship of Ugandan lakes to regional climate drivers breaks down c. 1800 AD, when major changes in the ecosystems appear to be a response to sediment and nutrient influxes as a result of increasing cultural impacts within the lake catchments. The data highlight the complexity of individual lake response to climate forcing, indicating shifting drivers through time. This research also highlights the importance of using multi-lake studies within a landscape to allow for rigorous testing of climate reconstructions, forcing and ecosystem response

    Together, yet still not equal? Sex integration in equestrian sport

    Sex segregation is a core organising principle of most modern sports and is a key element in the marginalisation and subordination of girls and women in sport and beyond. In this article I explore the only Olympic-level sport which is not organised around sex segregation – equestrian sport – in order to consider the implications of sex integration for female participants. I draw on a study conducted on elite riders that found that although sex integration in equestrian sport does not lead to female participants being excluded from high-level competition, men continue to perform disproportionately well. This suggests that although sex integration may be an important step towards breaking down gender hierarchies in sport, without accompanying wider changes in gender norms and expectations, sex integration alone will not be enough to achieve greater gender equality in equestrian sport

    Evidence for a bi-partition of the Younger Dryas Stadial in East Asia associated with inversed climate characteristics compared to Europe

    The Younger Dryas Stadial (YDS) was an episode of northern hemispheric cooling which occurred within the Last Glacial Interglacial Transition (LGIT). A major driver for the YDS climate was a weakening of the Atlantic Meridional Overturning Circulation (AMOC). It has been inferred that the AMOC began to strengthen mid-YDS, producing a bipartite structure of the YDS in records from continental Europe. These records imply that the polar front and westerlies shifted northward, producing a warmer second phase of the YDS in Europe. Here we present multi-proxy data from the sediments of Lake Suigetsu (Japan), as evidence that a related bi-partition of the YDS also occurred in East Asia. Besides showing for the first time that the bi-partition was not limited to the North Atlantic/European region, the data also imply a climatic dipole between Europe and East Asia since the cold-warm characteristics are reversed at Lake Suigetsu. We suggest that changes in eastward moisture transport from the North Atlantic are the primary mechanism by which the teleconnection can be explained

    Event layers in the Japanese Lake Suigetsu 'SG06' sediment core:Description, interpretation and climatic implications

    Event layers in lake sediments are indicators of past extreme events, mostly the results of floods or earthquakes. Detailed characterisation of the layers allows the discrimination of the sedimentation processes involved, such as surface runoff, landslides or subaqueous slope failures. These processes can then be interpreted in terms of their triggering mechanisms. Here we present a 40 ka event layer chronology from Lake Suigetsu, Japan. The event layers were characterised using a multi-proxy approach, employing light microscopy and ΌXRF for microfacies analysis. The vast majority of event layers in Lake Suigetsu was produced by flood events (362 out of 369), allowing the construction of the first long-term, quantitative (with respect to recurrence) and well dated flood chronology from the region. The flood layer frequency shows a high variability over the last 40 ka, and it appears that extreme precipitation events were decoupled from the average long-term precipitation. For instance, the flood layer frequency is highest in the Glacial at around 25 ka BP, at which time Japan was experiencing a generally cold and dry climate. Other cold episodes, such as Heinrich Event 1 or the Late Glacial stadial, show a low flood layer frequency. Both observations together exclude a simple, straightforward relationship with average precipitation and temperature. We argue that, especially during Glacial times, changes in typhoon genesis/typhoon tracks are the most likely control on the flood layer frequency, rather than changes in the monsoon front or snow melts. Spectral analysis of the flood chronology revealed periodic variations on centennial and millennial time scales, with 220 yr, 450 yr and a 2000 yr cyclicity most pronounced. However, the flood layer frequency appears to have not only been influenced by climate changes, but also by changes in erosion rates due to, for instance, earthquakes

    Review article: the future of microbiome-based therapeutics

    Published online 24 May 2022Background: From consumption of fermented foods and probiotics to emerging applications of faecal microbiota transplantation, the health benefit of manipulating the human microbiota has been exploited for millennia. Despite this history, recent technological advances are unlocking the capacity for targeted microbial manipulation as a novel therapeutic.Aim: This review summarises the current developments in microbiome- based medicines and provides insight into the next steps required for therapeutic development.Methods: Here we review current and emerging approaches and assess the capabilities and weaknesses of these technologies to provide safe and effective clinical inter-ventions. Key literature was identified through Pubmed searches with the following key words, ‘microbiome’, ‘microbiome biomarkers’, ‘probiotics’, ‘prebiotics’, ‘synbiotics’, ‘faecal microbiota transplant’, ‘live biotherapeutics’, ‘microbiome mimetics’ and ‘postbiotics’.Results: Improved understanding of the human microbiome and recent technological advances provide an opportunity to develop a new generation of therapies. These therapies will range from dietary interventions, prebiotic supplementations, single probiotic bacterial strains, human donor-derived faecal microbiota transplants, ra-tionally selected combinations of bacterial strains as live biotherapeutics, and the beneficial products or effects produced by bacterial strains, termed microbiome mimetics.Conclusions: Although methods to identify and refine these therapeutics are continually advancing, the rapid emergence of these new approaches necessitates accepted technological and ethical frameworks for measurement, testing, laboratory practices and clinical translation.Emily L.     Increased EZH2 expression in prostate cancer is associated with metastatic recurrence following external beam radiotherapy

    Background Enhancer of zeste 2 (EZH2) promotes prostate cancer progression. We hypothesized that increased EZH2 expression is associated with postradiotherapy metastatic disease recurrence, and may promote radioresistance. Methods EZH2 expression was investigated using immunohistochemistry in diagnostic prostate biopsies of 113 prostate cancer patients treated with radiotherapy with curative intent. Associations between EZH2 expression in malignant and benign tissue in prostate biopsy cores and outcomes were investigated using univariate and multivariate Cox regression analyses. LNCaP and PC3 cell radiosensitivity was investigated using colony formation and γH2AX assays following UNC1999 chemical probe‐mediated EZH2 inhibition. Results While there was no significant association between EZH2 expression and biochemical recurrence following radiotherapy, univariate analysis revealed that prostate cancer cytoplasmic and total EZH2 expression were significantly associated with metastasis development postradiotherapy (P = 0.034 and P = 0.003, respectively). On multivariate analysis, the prostate cancer total EZH2 expression score remained statistically significant (P = 0.003), while cytoplasmic EZH2 expression did not reach statistical significance (P = 0.053). No association was observed between normal adjacent prostate EZH2 expression and biochemical recurrence or metastasis. LNCaP and PC3 cell treatment with UNC1999 reduced histone H3 lysine 27 tri‐methylation levels. Irradiation of LNCaP or PC3 cells with a single 2 Gy fraction with UNC1999‐mediated EZH2 inhibition resulted in a statistically significant, though modest, reduction in cell colony number for both cell lines. Increased γH2AX foci were observed 24 hours after ionizing irradiation in LNCaP cells, but not in PC3, following UNC1999‐mediated EZH2 inhibition vs controls. Conclusions Taken together, these results reveal that high pretreatment EZH2 expression in prostate cancer in diagnostic biopsies is associated with an increased risk of postradiotherapy metastatic disease recurrence, but EZH2 function may only at most play a modest role in promoting prostate cancer cell radioresistance

    Broadband Quantum Enhancement of the LIGO Detectors with Frequency-Dependent Squeezing

    Quantum noise imposes a fundamental limitation on the sensitivity of interferometric gravitational-wave detectors like LIGO, manifesting as shot noise and quantum radiation pressure noise. Here, we present the first realization of frequency-dependent squeezing in full-scale gravitational-wave detectors, resulting in the reduction of both shot noise and quantum radiation pressure noise, with broadband detector enhancement from tens of hertz to several kilohertz. In the LIGO Hanford detector, squeezing reduced the detector noise amplitude by a factor of 1.6 (4.0 dB) near 1 kHz; in the Livingston detector, the noise reduction was a factor of 1.9 (5.8 dB). These improvements directly impact LIGO's scientific output for high-frequency sources (e.g., binary neutron star postmerger physics). The improved low-frequency sensitivity, which boosted the detector range by 15%-18% with respect to no squeezing, corresponds to an increase in the astrophysical detection rate of up to 65%. Frequency-dependent squeezing was enabled by the addition of a 300-meter-long filter cavity to each detector as part of the LIGO A+ upgrade
