    Indole 3-acetic acid, indoxyl sulfate and paracresyl-sulfate do not influence anemia parameters in hemodialysis patients

    International audienceBackground: The main reason for anemia in renal failure patients is the insufficient erythropoietin production by the kidneys. Beside erythropoietin deficiency, in vitro studies have incriminated uremic toxins in the pathophysiology of anemia but clinical data are sparse. In order to assess if indole 3-acetic acid (IAA), indoxyl sulfate (IS), and paracresyl sulfate (PCS)-three protein bound uremic toxins-are clinically implicated in end-stage renal disease anemia we studied the correlation between IAA, IS and PCS plasmatic concentrations with hemoglobin and Erythropoietin Stimulating Agents (ESA) use in hemodialysis patients. Methods: Between June and July 2014, we conducted an observational cross sectional study in two hemodialysis center. Three statistical approaches were conducted. First, we compared patients treated with ESA and those not treated. Second, we performed linear regression models between IAA, IS, and PCS plasma concentrations and hemoglobin, the ESA dose over hemoglobin ratio (ESA/Hemoglobin) or the ESA resistance index (ERI). Third, we used a polytomous logistic regression model to compare groups of patients with no/low/high ESA dose and low/high hemoglobin statuses

    FEZ2 Has Acquired Additional Protein Interaction Partners Relative to FEZ1: Functional and Evolutionary Implications

    BACKGROUND: The FEZ (fasciculation and elongation protein zeta) family designation was purposed by Bloom and Horvitz by genetic analysis of C. elegans unc-76. Similar human sequences were identified in the expressed sequence tag database as FEZ1 and FEZ2. The unc-76 function is necessary for normal axon fasciculation and is required for axon-axon interactions. Indeed, the loss of UNC-76 function results in defects in axonal transport. The human FEZ1 protein has been shown to rescue defects caused by unc-76 mutations in nematodes, indicating that both UNC-76 and FEZ1 are evolutionarily conserved in their function. Until today, little is known about FEZ2 protein function. METHODOLOGY/PRINCIPAL FINDINGS: Using the yeast two-hybrid system we demonstrate here conserved evolutionary features among orthologs and non-conserved features between paralogs of the FEZ family of proteins, by comparing the interactome profiles of the C-terminals of human FEZ1, FEZ2 and UNC-76 from C. elegans. Furthermore, we correlate our data with an analysis of the molecular evolution of the FEZ protein family in the animal kingdom. CONCLUSIONS/SIGNIFICANCE: We found that FEZ2 interacted with 59 proteins and that of these only 40 interacted with FEZ1. Of the 40 FEZ1 interacting proteins, 36 (90%), also interacted with UNC-76 and none of the 19 FEZ2 specific proteins interacted with FEZ1 or UNC-76. This together with the duplication of unc-76 gene in the ancestral line of chordates suggests that FEZ2 is in the process of acquiring new additional functions. The results provide also an explanation for the dramatic difference between C. elegans and D. melanogaster unc-76 mutants on one hand, which cause serious defects in the nervous system, and the mouse FEZ1 -/- knockout mice on the other, which show no morphological and no strong behavioural phenotype. Likely, the ubiquitously expressed FEZ2 can completely compensate the lack of neuronal FEZ1, since it can interact with all FEZ1 interacting proteins and additional 19 proteins

    The Digital Twin at the service of storage zone optimisation and assembly line simulation

    International audienceThis paper presents a complete optimisation methodology to improve costs linked to storage zones in assembly lines. It consists of coupled combinatorial optimisation and simulation techniques, relying on a Digital Twin UML model. The methodology is applied to a use-case provided by Groupe PSA, an automobile manufacturer. Asynchronisms necessarily appear in assembly lines because, generally, different types of products are produced on the same line, and because of hazards which result in a temporary stop of part of the line. The role of storage zones is to improve the ow of the line despite these desynchronisations. They however represent a financial cost, so a goal for industrialists is to minimise the size of these zones while still insuring the good performance of the line. At present, this optimisation process is done by experts who explore solutions by hand with a flow simulation tool in a trial and error approach. In this paper, a three step solution is proposed. A modelling framework is defined to create a Digital Twin of the factory, which serves as input to an optimisation process. A first combinatorial optimisation tool reduces the possible number of configurations, and then a simulation tool chooses the best configuration insuring the performance of the line

    Cet article reprend les intentions de départ des journées d'étude qui entendaient explorer, dans une perspective comparative France-Québec, la pluralité des récits impossibles dans divers champs d'études et de pratiques (intervenants sociaux, formateurs, juristes, psychologues, traducteurs-interprètes). Elles avaient pour ambition de décloisonner les expertises et faire dialoguer les différents acteurs pour mieux comprendre et analyser les expériences et phénomènes en contexte migratoire. Ceci nous a amenés progressivement sur le terrain de l’interculturalité. Nous pensons que les perspectives théoriques et pratiques entourant le recherche et l’intervention en contexte interculturel au XXIe siècle pourraient être actualisées et enrichir ainsi les regards sur les phénomènes sociaux et migratoires. C’est à partir d’une recension des écrits et une reconnaissance des tensions interculturelles, que nous proposons de développer une dimension critique qui permet de tenir compte des rapports de pouvoir, des dimensions structurelles et systémiques qui régissent les rapports sociétaux. Ainsi, nos propos invitent l’univers du politique, de la pratique et la recherche à mieux définir leurs contours pour s’adapter aux enjeux contemporains et promouvoir une perspective interculturelle ancrée dans les réalités et expériences vécues par les premiers concernés, les acteurs eux-mêmes, les citoyens et la société civile
