    Effects of an Ionic Bracelet on Physical, Cognitive, and Integrative Tasks

    Abstract: One approach in complementary/alternative medicine posits that manipulations of undocumented body energy patterns (biofields) can improve health and function. Various clinician-delivered interventions focus on biofields, but devices that purportedly correct these are also marketed and claim immediate benefits in a variety of domains. OBJECTIVE: To determine acute effects of ionic bracelet use on physical, cognitive and integrative sports-related tasks. METHODS: 52 healthy young adults participated (9 men and 43 women; 20.2±3.4 years of age; 65.4±11.3 kg; 165±6.9 cm). None reported prior or current use of the product. Each completed 2 series of 6 tasks wearing the ionic and an inert bracelet (counter-balanced presentation order). Bracelets were applied by the investigators behind a shield and covered by a sleeve in a single-blind design. Center of pressure excursion in single leg stance on a dynamic surface assessed posture control; anticipation timing absolute error represented perceptuomotor performance; time difference between 2 Stroop task variants tested concentration; peak vertical jump velocity (m/s) served to test power; maximum force in an isometric dead-lift was used to test strength; and time able to hold a weight (20% of body weight) at chin level served to test endurance. Effects of test order and bracelet type were analyzed independently via paired t-tests (α =.05). RESULTS: An order effect for strength was observed, attributable to a learning effect or an artifact of the number of t-tests applied; no other significant contrast was revealed. CONCLUSION: This ionic bracelet demonstrated no immediate benefits in any of the domains examined

    Specificity protein 1 induces the expression of angiomotin in response to IL-6/STAT3 activation to mediate YAP-dependent growth of breast cancer cells

    Indiana University-Purdue University Indianapolis (IUPUI)Chronic inflammation is a major driver of tumor progression in over fifty percent of breast cancers. Tumors activate inflammatory processes by secreting factors that recruit and trigger inflammatory cells to release cytokines such as Interleukin 6 (IL-6). IL-6 stimulates the activity of signal transducers and activators of transcription 3 (STAT3), a transcription factor that has been extensively studied for its role in promoting breast cancer. Recently, downregulated HIPPO signaling was shown to drive the pro-growth effects of IL 6. Reduced HIPPO signaling allows for the nuclear translocation of transcriptional co-activator yes associated protein (YAP), implicating IL-6 in the co-activation of several transcription factors such as the TEADs that trigger pro growth programs. While IL-6/STAT3 stimulation has been shown to increase YAP activity, the mechanism driving this remains undocumented. The Angiomotins (Amots) are adapters of the HIPPO pathway that directly bind and regulate YAP activity. Molecular characterization of Amot transcriptional regulation unexpectedly revealed a single promoter controlling the expression of its two major isoforms: Amot 130 and Amot 80. Through immunofluorescent analysis, this study found that total Amot levels were elevated across multiple breast tumor subtypes and highest in samples with increased presence of stromal inflammatory cells. Further, the induction of total Amot expression by IL 6 was found to be essential for YAP dependent growth of breast cancer cells. The activation of Amot transcription by IL-6 was found to be through Specificity Protein 1 (Sp1), a transcription factor that is activated by STAT3. This work connects the activation of YAP1 by IL-6/STAT3 through the elevation of Amot expression by Sp1. Taken together, this explains a new avenue whereby breast cancer cells acquire enhanced oncogenic properties in response to inflammatory signaling

    Cost-effectiveness of alternative methods of surgical repair of inguinal hernia

    Objectives: To assess the relative cost-effectiveness of laparoscopic methods of inguinal hernia repair compared with open flat mesh and open non-mesh repair. Methods: Data on the effectiveness of these alternatives came from three systematic reviews comparing: (i) laparoscopic methods with open flat mesh or non-mesh methods; (ii) open flat mesh with open non-mesh repair; and (iii) methods that used synthetic mesh to repair the hernia defect with those that did not. Data on costs were obtained from the authors of economic evaluations previously conducted alongside trials included in the reviews. A Markov model was used to model cost-effectiveness for a five-year period after the initial operation. The outcomes of the model were presented using a balance sheet approach and as cost per hernia recurrence avoided and cost per extra day at usual activities. Results: Open flat mesh was the most cost-effective method of preventing recurrences. Laparoscopic repair provided a shorter period of convalescence and less long-term pain compared with open flat mesh but was more costly. The mean incremental cost per additional day back at usual activities compared with open flat mesh was €38 and €80 for totally extraperitoneal and transabdominal preperitoneal repair, respectively. Conclusions: Laparoscopic repair is not cost-effective compared with open flat mesh repair in terms of cost per recurrence avoided. Decisions about the use of laparoscopic repair depend on whether the benefits (reduced pain and earlier return to usual activities) outweigh the extra costs and intraoperative risks.     Retroviral Expression of Transforming Growth Factor-Alpha Does Not Transform Fibroblasts or Keratinocytes

    Transforming growth factor α (TGFα) is a peptide so named because it helps to impart anchorage-independent growth to normal rat kidney (NRK) cells in vitro and is secreted by many rodent and human tumor cells. To directly investigate the transforming properties of this factor, we constructed a replication-defective murine retrovirus that expresses the human sequence coding for TGFα. infection of NIH/3T3 cells with the TGFα retrovirus led to the integration of a transcriptionally active provirus and overexpression of biologically active TGFα, but failed to induce morphologic transformation. Similarly, the TGFα retrovirus failed to induce morphologic transformation of five other types of rodent fibroblasts.We also investigated the effect of TGFα expression on the growth of BALB/MK mouse keratinocytes, which require epidermal growth factor (EGF) for proliferation. We show that exogenously added TGFα is an extremely potent mitogen for BALB/MK cells. However, retroviral expression of TGFa in BALB/MK cells failed to relieve dependence on exogenously added EGF (or TGFα) for cell growth. These results suggest that overexpression of TGFα does not, by itself, transform rodent fibroblasts or keratinocytes

    Learning Behavioral Representations of Human Mobility

    In this paper, we investigate the suitability of state-of-the-art representation learning methods to the analysis of behavioral similarity of moving individuals, based on CDR trajectories. The core of the contribution is a novel methodological framework, mob2vec, centered on the combined use of a recent symbolic trajectory segmentation method for the removal of noise, a novel trajectory generalization method incorporating behavioral information, and an unsupervised technique for the learning of vector representations from sequential data. Mob2vec is the result of an empirical study conducted on real CDR data through an extensive experimentation.     Comments on Non-holomorphic Modular Forms and Non-compact Superconformal Field Theories

    We extend our previous work arXiv:1012.5721 [hep-th] on the non-compact N=2 SCFT_2 defined as the supersymmetric SL(2,R)/U(1)-gauged WZW model. Starting from path-integral calculations of torus partition functions of both the axial-type (`cigar') and the vector-type (`trumpet') models, we study general models of the Z_M-orbifolds and M-fold covers with an arbitrary integer M. We then extract contributions of the degenerate representations (`discrete characters') in such a way that good modular properties are preserved. The `modular completion' of the extended discrete characters introduced in arXiv:1012.5721 [hep-th] are found to play a central role as suitable building blocks in every model of orbifolds or covering spaces. We further examine a large M-limit (the `continuum limit'), which `deconstructs' the spectral flow orbits while keeping a suitable modular behavior. The discrete part of partition function as well as the elliptic genus is then expanded by the modular completions of irreducible discrete characters, which are parameterized by both continuous and discrete quantum numbers modular transformed in a mixed way. This limit is naturally identified with the universal cover of trumpet model.
