    Settlement influences the distribution and abundance of many marine organisms, although the relative roles of abiotic and biotic factors influencing settlement are poorly understood. Species that aggregate often owe this characteristic to larval behaviour, and we investigated whether this predisposes ascidians to becoming invasive, by increasing their capacity to maintain their populations. We explored the interactive effects of larval phototaxis and geotaxis and conspecific adult extracts on settlement rates of a representative suite of 6 species of ascidians that form aggregations in the field, including 4 aliens with global distributions, and how they relate to adult habitat characteristics. In the laboratory, the larvae were (1) held in light or dark, (2) offered the choice of settling in the light or dark, or (3) held in the presence or absence of adult extract. When confined in either light or dark conditions, all species settled equally in dark and light. Four showed strong geotaxis, 3 settling preferentially on the bottom of experimental chambers, and one on the top. Offered a choice between dark and light, 2 species settled preferentially in the dark with no geotactic preferences and another 2 showed an interaction between light and geotaxis. For 4 of the species, the responses of settlers accorded with, and may contribute to, adult orientation patterns in the field. Adult extracts inhibited settlement of 3 species and failed to influence settlement of the other 3, arguing against conspecific attraction being a cause of aggregation and an explanation of the propensity of ascidians to become invasive

    Ultrasonographic Visualization of the Ovaries to Detect Ovarian Cancer According to Age, Menopausal Status and Body Type

    Because the effects of age, menopausal status, weight and body mass index (BMI) on ovarian detectability by transvaginal ultrasound (TVS) have not been established, we determined their contributions to TVS visualization of the ovaries. A total of 29,877 women that had both ovaries visualized on their first exam were followed over 202,639 prospective TVS exams. All images were reviewed by a physician. While visualization of both ovaries decreased with age, one or both ovaries could be visualized in two of every three women over 80 years of age. Around 93% of pre-menopausal women and ~69% of post-menopausal women had both ovaries visualized. Both ovaries were visualized in ~72% of women weighing over 300 lbs. and in ~70% of women with a BMI over 40. Conclusions: Age had the greatest influence on the visualization of the ovaries. The ovaries can be visualized well past the menopause. Body habitus was not limiting to TVS ovarian imaging, and TVS should be considered capable of imaging one or both ovaries in two of every three women over 80 years of age. Thus, older and obese patients remain good candidates for TVS exams

    Tanzania's reptile biodiversity : Distribution, threats and climate change vulnerability

    Assessments of biodiversity patterns and threats among African reptiles have lagged behind those of other vertebrate groups and regions. We report the first systematic assessment of the distribution, threat status, and climate change vulnerability for the reptiles of Tanzania. A total of 321 reptile species (including 90 Tanzanian endemics) were assessed using the global standard IUCN Red List methodology and 274 species were also assessed using the IUCN guidelines for climate change vulnerability. Patterns of species richness and threat assessment confirm the conservation importance of the Eastern Arc Mountains, as previously demonstrated for birds, mammals and amphibians. Lowland forests and savannah-woodland habitats also support important reptile assemblages. Protected area gap analysis shows that 116 species have less than 20% of their distribution ranges protected, among which 12 are unprotected, eight species are threatened and 54 are vulnerable to climate change. Tanzania's northern margins and drier central corridor support high numbers of climate vulnerable reptile species, together with the eastern African coastal forests and the region between Lake Victoria and Rwanda. This paper fills a major gap in our understanding of the distribution and threats facing Tanzania's reptiles, and demonstrates more broadly that the explicit integration of climate change vulnerability in Red Listing criteria may revise spatial priorities for conservation

    Atopic dermatitis is a common pruritic skin disease in which barrier dysfunction and cutaneous inflammation play a role in pathogenesis. Mechanisms underlying the associated inflammation are not fully understood, and while CD1a-expressing Langerhans cells are known to be enriched within lesions, their role in clinical disease pathogenesis has not been studied. Here we observed that house dust mite (HDM) generates neolipid antigens for presentation by CD1a to T cells in the blood and skin lesions of affected individuals. HDM-responsive CD1a-reactive T cells increased in frequency after birth and showed rapid effector function, consistent with antigen-driven maturation. To define the underlying mechanisms, we analyzed HDM-challenged human skin and observed allergen-derived phospholipase (PLA2) activity in vivo. CD1a-reactive T cell activation was dependent on HDM-derived PLA2 and such cells infiltrated the skin after allergen challenge. Filaggrin insufficiency is associated with atopic dermatitis, and we observed that filaggrin inhibits PLA2 activity and inhibits CD1a-reactive PLA2-generated neolipid-specific T cell activity from skin and blood. The most widely used classification schemes of hypersensitivity, such as Gell and Coombs are predicated on the idea that non-peptide stimulants of T cells act as haptens that modify peptides or proteins. However our results point to a broader model that does not posit haptenation, but instead shows that HDM proteins generate neolipid antigens which directly activate T cells. Specifically, the data identify a pathway of atopic skin inflammation, in which house dust mite-derived phospholipase A2 generates antigenic neolipids for presentation to CD1a-reactive T cells, and define PLA2 inhibition as a function of filaggrin, supporting PLA2 inhibition as a therapeutic approach

    Significance of Pelvic Fluid Observed during Ovarian Cancer Screening with Transvaginal Sonogram

    The primary objective was to examine the role of pelvic fluid observed during transvaginal ultrasonography (TVS) in identifying ovarian malignancy. A single-institution, observational study was conducted within the University of Kentucky Ovarian Cancer Screening trial from January 1987 to September 2019. We analyzed true-positive (TP), false-positive (FP), true-negative (TN), and false-negative (FN) groups for the presence of pelvic fluid during screening encounters. Measured outcomes were the presence and duration of fluid over successive screening encounters. Of the 48,925 women surveyed, 2001 (4.1%) had pelvic fluid present during a TVS exam. The odds ratio (OR) of detecting fluid in the comparison group (TN screen; OR = 1) significantly differed from that of the FP cases (benign pathology; OR: 13.4; 95% confidence interval (CI): 9.1–19.8), the TP cases with a low malignant potential (LMP; OR: 28; 95% CI: 26.5–29.5), TP ovarian cancer cases (OR: 50.4; 95% CI: 27.2–93.2), and FN ovarian cancer cases (OR: 59.3; 95% CI: 19.7–178.1). The mean duration that pelvic fluid was present for women with TN screens was 2.2 ± 0.05 encounters, lasting 38.7 ± 1.3 months. In an asymptomatic screening population, free fluid identified in TVS exams was more associated with ovarian malignancy than in the control group or benign ovarian tumors. While pelvic free fluid may not solely discriminate malignancy from non-malignancy, it appears to be clinically relevant and warrants thoughtful consideration
