    An ELMO2-RhoG-ILK network modulates microtubule dynamics

    ELMO2 belongs to a family of scaffold proteins involved in phagocytosis and cell motility. ELMO2 can simultaneously bind integrin-linked kinase (ILK) and RhoG, forming tripartite ERI complexes. These complexes are involved in promoting β1 integrin-dependent directional migration in undifferentiated epidermal keratinocytes. ELMO2 and ILK have also separately been implicated in microtubule regulation at integrin-containing focal adhesions. During differentiation, epidermal keratinocytes cease to express integrins, but ERI complexes persist. Here we show an integrin-independent role of ERI complexes in modulation of microtubule dynamics in differentiated keratinocytes. Depletion of ERI complexes by inactivating the Ilk gene in these cells reduces microtubule growth and increases the frequency of catastrophe. Reciprocally, exogenous expression of ELMO2 or RhoG stabilizes microtubules, but only if ILK is also present. Mechanistically, activation of Rac1 downstream from ERI complexes mediates their effects on microtubule stability. In this pathway, Rac1 serves as a hub to modulate microtubule dynamics through two different routes: 1) phosphorylation and inactivation of the microtubule-destabilizing protein stathmin and 2) phosphorylation and inactivation of GSK-3β, which leads to the activation of CRMP2, promoting microtubule growth. At the cellular level, the absence of ERI species impairs Ca2+-mediated formation of adherens junctions, critical to maintaining mechanical integrity in the epidermis. Our findings support a key role for ERI species in integrin-independent stabilization of the microtubule network in differentiated keratinocytes

    Vector competence of Aedes aegypti, Culex tarsalis, and Culex quinquefasciatus from California for Zika virus.

    Zika virus (ZIKV) has emerged since 2013 as a significant global human health threat following outbreaks in the Pacific Islands and rapid spread throughout South and Central America. Severe congenital and neurological sequelae have been linked to ZIKV infections. Assessing the ability of common mosquito species to transmit ZIKV and characterizing variation in mosquito transmission of different ZIKV strains is important for estimating regional outbreak potential and for prioritizing local mosquito control strategies for Aedes and Culex species. In this study, we evaluated the laboratory vector competence of Aedes aegypti, Culex quinquefasciatus, and Culex tarsalis that originated in areas of California where ZIKV cases in travelers since 2015 were frequent. We compared infection, dissemination, and transmission rates by measuring ZIKV RNA levels in cohorts of mosquitoes that ingested blood meals from type I interferon-deficient mice infected with either a Puerto Rican ZIKV strain from 2015 (PR15), a Brazilian ZIKV strain from 2015 (BR15), or an ancestral Asian-lineage Malaysian ZIKV strain from 1966 (MA66). With PR15, Cx. quinquefasciatus was refractory to infection (0%, N = 42) and Cx. tarsalis was infected at 4% (N = 46). No ZIKV RNA was detected in saliva from either Culex species 14 or 21 days post feeding (dpf). In contrast, Ae. aegypti developed infection rates of 85% (PR15; N = 46), 90% (BR15; N = 20), and 81% (MA66; N = 85) 14 or 15 dpf. Although MA66-infected Ae. aegypti showed higher levels of ZIKV RNA in mosquito bodies and legs, transmission rates were not significantly different across virus strains (P = 0.13, Fisher's exact test). To confirm infectivity and measure the transmitted ZIKV dose, we enumerated infectious ZIKV in Ae. aegypti saliva using Vero cell plaque assays. The expectorated plaque forming units PFU varied by viral strain: MA66-infected expectorated 13±4 PFU (mean±SE, N = 13) compared to 29±6 PFU for PR15-infected (N = 13) and 35±8 PFU for BR15-infected (N = 6; ANOVA, df = 2, F = 3.8, P = 0.035). These laboratory vector competence results support an emerging consensus that Cx. tarsalis and Cx. quinquefasciatus are not vectors of ZIKV. These results also indicate that Ae. aegypti from California are efficient laboratory vectors of ancestral and contemporary Asian lineage ZIKV

The purpose of this study was to identify the underlying factors that contributed to adolescents’ perceptions of injury and the various pressures they experienced when returning to sport. Two themes emerged from the study: support and fear. Support was provided to participants through development, care, and the environment. Fear was the factor that affected the participant in their return to sport, which came in the form of worry and doubt

    Exact Eignstates for Trapped Weakly Interacting Bosons in Two Dimensions

    A system of N two-dimensional weakly interacting bosons in a harmonic trap is considered. When the two-particle potential is a delta function Smith and Wilkin have analytically proved that the elementary symmetric polynomials of particle coordinates measured from the center of mass are exact eigenstates. In this study, we point out that their proof works equally well for an arbitrary two-particle potential which possesses the translational and rotational symmetries. We find that the interaction energy associated with the eigenstate with angular momentum L is equal to aN(N-1)/2+(b-a)NL/2, where a and b are the interaction energies of two bosons in the lowest-energy one-particle state with zero and one unit of angular momentum, respectively. Additionally, we study briefly the case of attractive quartic interactions. We prove rigorously that the lowest-energy state is the one in which all angular momentum is carried by the center of mass motion.Comment: 4 pages, minor changes made, to appear in PRA Brie

    Quasi-continuous atom laser in the presence of gravity

    We analyse the extraction of a coherent atomic beam from a trapped Bose-Einstein condensate using a rf transition to a non-trapping state at T=0 K. Our quantum treatment fully takes gravity into account but neglects all interactions in the free falling beam. We obtain an analytical expression of the output rate and of the wave function of the extracted beam, i.e. the output mode of the ``atom laser''. Our model reproduces satisfactorily experimental data without any adjustable parameter.Comment: 4 pages, 2 figure

    A method for collective excitation of Bose-Einstein condensate

The proposed method is complementary to earlier suggestions.     Optical linewidth of a low density Fermi-Dirac gas

    We study propagation of light in a Fermi-Dirac gas at zero temperature. We analytically obtain the leading density correction to the optical linewidth. This correction is a direct consequence of the quantum statistical correlations of atomic positions that modify the optical interactions between the atoms at small interatomic separations.
