    Mental wellbeing, air pollution and the ecological state

    The ecological State puts environmental considerations at the centre of its actions. To explores its role in the association between air pollution and mental wellbeing, this work employs a hierarchical three-level analysis on the third wave (2011-2012) of the European Quality of Life Survey (N-citizens = 25007, N-regions = 216, N-countries = 20). It uses a classification of Environmental Governance Regimes, subjective and objective indicators of air pollution, and the WHO-5 index of mental wellbeing. The findings show that the perception of major air pollution problems and worse mental wellbeing go hand in hand only in partial and established environmental States

    Frondoside A suppressive effects on lung cancer survival, tumor growth, angiogenesis, invasion, and metastasis

    A major challenge for oncologists and pharmacologists is to develop less toxic drugs that will improve the survival of lung cancer patients. Frondoside A is a triterpenoid glycoside isolated from the sea cucumber, Cucumaria frondosa and was shown to be a highly safe compound. We investigated the impact of Frondoside A on survival, migration and invasion in vitro, and on tumor growth, metastasis and angiogenesis in vivo alone and in combination with cisplatin. Frondoside A caused concentration-dependent reduction in viability of LNM35, A549, NCI-H460-Luc2, MDA-MB-435, MCF-7, and HepG2 over 24 hours through a caspase 3/7-dependent cell death pathway. The IC50 concentrations (producing half-maximal inhibition) at 24 h were between 1.7 and 2.5 mu M of Frondoside A. In addition, Frondoside A induced a time- and concentration-dependent inhibition of cell migration, invasion and angiogenesis in vitro. Frondoside A (0.01 and 1 mg/kg/day i.p. for 25 days) significantly decreased the growth, the angiogenesis and lymph node metastasis of LNM35 tumor xenografts in athymic mice, without obvious toxic side-effects. Frondoside A (0.1-0.5 mu M) also significantly prevented basal and bFGF induced angiogenesis in the CAM angiogenesis assay. Moreover, Frondoside A enhanced the inhibition of lung tumor growth induced by the chemotherapeutic agent cisplatin. These findings identify Frondoside A as a promising novel therapeutic agent for lung cancer

    Activation of the E-cadherin/catenin complex in human MCF-7 breast cancer cells by all-trans-retinoic acid.

    All-trans-retinoic acid (RA), like insulin-like growth factor I (IGF-I) and tamoxifen, inhibit invasion of human MCF-7/6 mammary cancer cells in vitro. For tamoxifen and for IGF-I, activation of the invasion-suppressor function of the E-cadherin/catenin complex was shown to be the most probable mechanism of the anti-invasive action. We did a series of experiments to determine whether the anti-invasive effect of RA also implicated the invasion-suppressor E-cadherin/catenin complex. Human MCF-7/6 mammary and HCT-8/R1 colon cancer cells, both with a dysfunctional E-cadherin/catenin complex, were treated with RA and the function of the complex was evaluated through Ca(2+)-dependent fast aggregation. Fast aggregation of both MCF-7/6 and HCT-8/R1 cells was induced by 1 microM RA. This effect was abolished by antibodies against E-cadherin. RA-induced fast aggregation was not sensitive to cycloheximide, tyrosine kinase inhibitors or antibodies against IGF-I or against the IGF-I receptor. RA did not stimulate IGF-I receptor phosphorylation or alter the E-cadherin/catenin complex, as evidenced by immunoprecipitation. RA up-regulates the function of the invasion-suppressor complex E-cadherin/catenin. Its action mechanism is different from that of IGF-I. RA may act as an anti-invasive agent with unique mechanisms of action

    Differences in Tumor-Associated Protein Levels among Middle-Age Flemish Women in Association with Area of Residence and Exposure to Pollutants

    We measured tumor-associated proteins (TAPs) and pollutants in blood, serum, and urine of 200 nonsmoking women 50–65 years of age, residing in the rural municipality of Peer or in Hoboken or Wilrijk, industrial suburbs of Antwerp, Belgium. Persons with occupational exposures or commuting to other towns were excluded. Residents from Hoboken had significantly higher levels of blood lead and serum zinc and polychlorinated biphenyls. Surprisingly, residents of Peer had significantly higher levels of serum cadmium, dioxin-like activity in blood fat, and urinary 1-hydroxypyrene. For 5 of the 12 TAPs assessed in this study, we observed significant differences in serum levels among residents of the three municipalities after adjusting for personal or lifestyle parameters. Although we found levels of internal exposure to pollutants to be quite homogeneous in Flanders, we found significantly higher levels of TAPs only in the industrial suburbs. In multiple regression with all 29 available personal, lifestyle, and internal exposure parameters, blood lead levels showed a positive association with serum levels of anti-p53, carcino-embryonic antigen (CEA), and tissue polypeptide–specific antigen (TPS) and with an index for mean TAP level (I(tap)); dioxin-like activity in serum and serum copper showed a positive association with serum CA 125 (cancer antigen 125); and serum zinc showed a positive association with serum levels of c-erbB-2 ectodomain and TPS. An index of internal exposure showed a positive association with serum levels of both CEA and anti-p53 and with I(tap). This study provides some evidence that levels of internal exposure such as those present in Flanders, in particular concerning lead, are indeed associated with biologic effects

    The New RF Control System for the CERN SPS Accelerator

    The old SPS RF control system designed in 1972 has been replaced completely, i.e. both hardware and software. The new system has to control both RF equipment conceived during the last 23 years and future (modern) equipment. Using information analysis methods, we derived a model of an RF command and designed a data base accordingly (ORACLE®). Information from this data base is used for command generation and processing and also for archiving settings. The advantage is purely generic software, i.e. the same computer code is used for switching on an RF amplifier, as for setting a frequency synthesizer. New equipment is added very simply by entering new records in the data base. Additional features include a reservation scheme whereby a user can take private control of any piece of equipment, a reporting facility notifying the user of the simultaneous control activity by other users on RF equipment, and a capability scheme assigning a level of expertise to each user restricting action on the equipment

    Thermo-mechanical behavior of a granodiorite from the Liquiñe fractured geothermal system (39°S) in the Southern Volcanic Zone of the Andes

    Fractures and faults in granitic rocks play an important role in geothermal systems because they permit the circulation of hot fluids. However, the thermo-hydro-mechanical behavior of granitic rocks has predominantly been studied at temperatures exceeding 300 °C but many geothermal systems experience temperatures much lower than this. The aim of this study was to evaluate how the depth, temperature, and amount and rate of mechanical loading associated conditions, that are realistic in low temperature geothermal system, influence the physical properties of geothermal reservoir hosting rock. We carried out both room temperature and low temperature thermo-mechanical tests on a granodiorite sample from the Liquiñe area, Chile, and performed post-experimental X-ray microtomography analysis to numerically estimate the permeability of the generated fractures. The results showed that both rock strength and rock stiffness decreased with increments of temperature treatment related to the development of thermal crack damage at temperatures > 150 °C and through the development of sub-critical cracking at constant temperatures between 50–75 °C. Slowest deformed samples also exhibited lower strengths, attributed to the development of sub-critical cracking. The cyclic triaxial loading test indicated that significant mechanical fracture damage was only initiated above 80% of the peak stress regardless of the number of repeated loading cycles at lower stresses. Low-temperature treatment appears to be a conditioning factor, but not the dominant factor in controlling the physical properties of reservoir hosting rocks. Our findings indicate that thermal crack damage is likely important for developing microfracture related permeability at depths between around 2–6 km where the temperature is sufficiently high to induce thermal cracking. At shallower depths, such was previously estimated the reservoir of Liquiñe, thermal crack damage is only generated adjacent to fractures that remain open and circulate the hot fluids but sub-critical cracking over time reduces the strength of rocks in lower temperature regimes. These processes combined to produce a geothermal reservoir in Liquiñe which likely first required the presence of a highly fractured fault zone
