    Studies of encephalitozoonosis in vervet monkeys (Cercopithecus pygerythrus) orally inoculated with spores of Encephalitozoon cuniculi isolated from dogs (Canis familiaris)

    Encephalitozoonosis was induced in 35 of 38 vervet monkeys (Cercopithecus pygerythrus). They were either directly (orally) inoculated with Encephlitozoon cuniculi or indirectly exposed to this protozoan parasite. Cell-culture-grown spores of E. cuniculi, isolated from the kidneys of dogs with natural, fatal disease, were administered orally to 29 of these monkeys. Another 5 were exposed in utero by orally infecting pregnant females, and 3 were exposed to horizontal infection by nursing infected infants. Only one was given an intravenous inoculation of spores. The disease was induced in non-gravid and late-pregnant adults, immunocompetent infants, and in infants that were immunologically compromised by parenteral steroid administration, as well as in one infant that was immunologically immature because of its premature birth. The effects of age, dosage, post-inoculation (PI) interval, passage level of the parasite in cell culture and immunological status of the host were correlated with macroscopical and microscopical lesions. The experimentally induced infection was confirmed either by reisolation of the parasite in cell culture or by observation of spores in tissue sections. Both confirmatory methods were supported by serological examination. Reisolation of the organism in primary cell culture prepared from kidneys usually resulted in more frequent isolates and larger yields of spores from infants than from adult vervets. Infection with E. cuniculi invariably induced subclinical disease. Based on histology, lesions were minimal to moderately severe, depending on age, PI interval, and immunological status of the host. Alimentary tract infections were seen histologically as early as three days Pl. Subsequently, infections resulted in detectable lesions most consistently in the liver, kidneys and brain. Lesions in these organs were generally granulomatous and were similar to those found in canine encephalitozoonosis. In addition, multifocal interstitial pneumonitis and myocarditis as well as vasculitis and perivasculitis were seen in other tissues and organs. Infants had more severe and more widespread lesions than adults. Although lesions and spores were still present in the brain of one immunocompetent infant 36 weeks after initial infection, the disease in immunocompetent infants and adults is thought to be self-limiting. However, infection may persist. Immunological depression favoured increased growth and multiplication of the organism, and resulted in detection of more spores within inflammatory lesions as well as more intracellular colonies of the organism that were free of inflammatory reaction. Long-term growth of E. cuniculi in cell culture did not appear to alter its pathogenicity, nor did varying the numbers of spores (2 x 10⁶-3 x 10¹º) in the inoculum appear to influence the severity of the disease. Horizontal infection was readily established, and vertical transmission was demonstrated. Results of this study suggest that spores of E. cuniculi shed by dogs may become agents of latent infection and disease in vervets, and probably other nonhuman primates as well.     Considering the importance of the interrelationship between health and economic development, a review was made of selected papers on the subject published in the last 20 years, both in Brazil and abroad. The review shows a tendency towards an objective analysis of the subject by the use of refined statistical analysis and econometric methods. Due to the relative lack of this kind of study in Brazil, more cooperation is recommended among economists, planners and physicians, in an effort to fultil this gap by researching together.Tendo em vista a importância das relações entre saúde e desenvolvimento econômico, foi feita uma revisão do tema destacando trabalhos publicados nas duas últimas décadas, tanto na literatura médica estrangeira como na nacional. A revisão revela uma tendência à análise objetiva do problema, mediante o uso de análise estatística sofisticada e métodos econométricos. Diante da carência deste tipo de estudos no Brasil, recomenda-se maior colaboração entre médicos, planejadores e economistas para realizar pesquisas que preencham esta lacuna

    Antemortem volume loss mirrors TDP-43 staging in older adults with non-frontotemporal lobar degeneration

    Over the past decade, the transactive response DNA-binding protein of 43 kDa (TDP-43) has been recognized as a major protein in normal and pathological ageing, increasing the risk of cognitive impairment and dementia. In conditions distinct from the frontotemporal lobar degenerations, TDP-43 appears to progress in a stereotypical pattern. In the present study, we aimed at providing a better understanding of the effects of TDP-43 and other age-related neuropathologies on cross-sectional grey matter volume in a cohort of non-FTLD subjects. We included 407 individuals with an antemortem MRI and post-mortem brain tissue from the Mayo Clinic Alzheimer's Disease Research Center, Mayo Clinic Alzheimer's Disease Patient Registry, or the Mayo Clinic Study of Aging. All individuals were assigned pathological stages for TDP-43, tau, amyloid-\u3b2, Lewy bodies, argyrophilic grain disease and vascular pathologies. Robust regressions were performed in regions of interest and voxel-wise to explore the relationships between TDP-43 stages and grey matter volume while controlling for other pathologies. Grey matter volumes adjusted for pathological and demographic variables were also computed for each TDP-43-positive case to further characterize the sequential involvement of brain structures associated with TDP-43, irrespective of the TDP-43 staging scheme. Robust regressions showed that the extent of TDP-43 pathology was associated with the extent of grey matter atrophy. Specifically, we found that the volume in medial temporal regions (i.e. amygdala, entorhinal cortex, hippocampus) decreased progressively with advancing TDP-43 stages. Importantly, these effects were of similar magnitude to those related to tau stages. Additional analyses using adjusted grey matter volume demonstrated a sequential pattern of volume loss associated with TDP-43, starting within the medial temporal lobe, followed by early involvement of the temporal pole, and eventually encompassing additional temporal and frontal regions. Altogether, this study demonstrates the major and independent contribution of TDP-43 pathology on neurodegeneration and provides further insight into the regional distribution of TDP-43 in non-FTLD subjects.     First report of Triassic vertebrate assemblages from the Villány Hills (Southern Hungary)

    Abstract Remains of Triassic vertebrates discovered in the Villány Hills (SW Hungary) are described here. After the well-documented Late Cretaceous Iharkút locality, this material represents the second systematically collected assemblage of Mesozoic vertebrates from Hungary. Fossils were collected from both the classical abandoned road-cut at Templom Hill (Templom-hegy) and a newly discovered site at a construction zone located 200 meters west of the road-cut. Macrofossils of the construction site are mainly isolated bones and teeth of nothosaurs from the Templomhegy Dolomite, including a fragmentary mandible referred to as Nothosaurus sp. and placodont teeth tentatively assigned here to cf. Cyamodus sp. Affinities of these fossils suggest a Middle Triassic (Ladinian) age of these shallow marine deposits. New palynological data prove for the first time a Late Triassic (Carnian) age of the lower part of the Mészhegy Sandstone Formation. Vertebrate remains discovered in this formation clearly represent a typical Late Triassic shallow-marine fauna including both chondrichthyan (Lissodus, Palaeobates, Hybodus) and osteichthyan (cf. Saurichthys, ?Sphaerodus sp.) fish fossils. The presence of reworked nothosaur and placodont tooth fragments as well as of possible archosauriform teeth, suggest an increase of terrestrial influence and the erosion of underlying Triassic deposits during the Late Triassic. A belemnite rostrum collected from the lowermost beds of the Somssichhegy Limestone Formation proves that this Lower Jurassic (Pliensbachian) layer was deposited in a marine environment. Most of the vertebrate remains (nothosaurs, placodonts, hybodont shark teeth, perhaps Palaeobates, Lissodus) recovered from these beds are also reworked Triassic elements strongly supporting an erosive, nearshore depositional environment
