Cell-free prediction of protein expression costs for growing cells
Translating heterologous proteins places significant burden on host cells, consuming expression resources leading to slower cell growth and productivity. Yet predicting the cost of protein production for any given gene is a major challenge, as multiple processes and factors combine to determine translation efficiency. To enable prediction of the cost of gene expression in bacteria, we describe here a standard cell-free lysate assay that provides a relative measure of resource consumption when a protein coding sequence is expressed. These lysate measurements can then be used with a computational model of translation to predict the in vivo burden placed on growing E. coli cells for a variety of proteins of different functions and lengths. Using this approach, we can predict the burden of expressing multigene operons of different designs and differentiate between the fraction of burden related to gene expression compared to action of a metabolic pathway
Controlled Production of Sub-Radiant States of a Diatomic Molecule in an Optical Lattice
We report successful production of sub-radiant states of a two-atom system in
a three-dimensional optical lattice starting from doubly occupied sites in a
Mott insulator phase of a quantum gas of atomic ytterbium. We can selectively
produce either sub-radiant 1g state or super-radiant 0u state by choosing the
excitation laser frequency. The inherent weak excitation rate for the
sub-radiant 1g state is overcome by the increased atomic density due to the
tight-confinement in a three-dimensional optical lattice. Our experimental
measurements of binding energies, linewidth, and Zeeman shift confirm
observation of sub-radiant levels of the 1g state of the Yb_2 molecule.Comment: To be published in Phys. Rev. Let
Local density of states induced by anisotropic impurity scattering in a d-wave superconductor
We study a single impurity effect on the local density of states in a d-wave
superconductor accounting for the momentum-dependent impurity potential. We
show that the anisotropy of the scattering potential can alter significantly
the spatial dependence of the quasiparticle density of states in the vicinity

Direct measurement of the charge distribution along a biased carbon nanotube bundle using electron holography
Nanowires and nanotubes can be examined in the transmission electron microscope under an applied bias. Here we introduce a model-independent method, which allows the charge distribution along a nanowire or nanotube to be measured directly from the Laplacian of an electron holographic phase image. We present results from a biased bundle of carbon nanotubes, in which we show that the charge density increases linearly with distance from its base, reaching a value of similar to 0.8 electrons/nm near its tip. Oxylipins in triglyceride-rich lipoproteins of dyslipidemic subjects promote endothelial inflammation following a high fat meal.
Elevated triglyceride-rich lipoproteins (TGRL) in circulation is a risk factor for atherosclerosis. TGRL from subjects consuming a high saturated fat test meal elicited a variable inflammatory response in TNFα-stimulated endothelial cells (EC) that correlated strongly with the polyunsaturated fatty acid (PUFA) content. This study investigates how the relative abundance of oxygenated metabolites of PUFA, oxylipins, is altered in TGRL postprandially, and how these changes promote endothelial inflammation. Human aortic EC were stimulated with TNFα and treated with TGRL, isolated from subjects' plasma at fasting and 3.5 hrs postprandial to a test meal high in saturated fat. Endothelial VCAM-1 surface expression stimulated by TNFα provided a readout for atherogenic inflammation. Concentrations of esterified and non-esterified fatty acids and oxylipins in TGRL were quantified by mass spectrometry. Dyslipidemic subjects produced TGRL that increased endothelial VCAM-1 expression by ≥35%, and exhibited impaired fasting lipogenesis activity and a shift in soluble epoxide hydrolase and lipoxygenase activity. Pro-atherogenic TGRL were enriched in eicosapentaenoic acid metabolites and depleted in esterified C18-PUFA-derived diols. Abundance of these metabolites was strongly predictive of VCAM-1 expression. We conclude the altered metabolism in dyslipidemic subjects produces TGRL with a unique oxylipin signature that promotes a pro-atherogenic endothelial phenotype
Strategy instruction and attributional beliefs in West Germany and the United States: Do teachers foster metacognitive development?
Previous research has shown German children to be more strategic on sort-recall memory tasks than their American age-mates, and to show fewer effort-related attributions. We conducted this study to determine if those differences are due to systematic differences in the strategy instruction and attributional beliefs of German and U.S. teachers, and to explore metacognitive instructional practices in the two countries. Teachers responded to a questionnaire that inquired about their use of strategy instructions, fostering of reflective thinking in pupils, sources of children’s learning problems, and modeling of metacognitive skills such as monitoring. The second part of the questionnaire asked about the reasons underlying children’s academic successes and failures. German teachers reported more instruction of task-specific strategies, while American teachers showed more effort-related attributions. The types of strategies instructed and types of learning problems most frequently described varied across the two countries, and also according to how many years the teachers had taught. Results were discussed regarding their implications for metacognitive developmental theory, particularly regarding culture and other environmental influences on the development of controlled processing
Strategy acquisition and transfer among American and German children: Environmental influences on metacognitive development
This study explored the differential effects of strategy training on German and American elementaryschool children and assessed the role of parents in the development of their children's strategic behavior and metacognition. 184 German and 161 American children were pretested on memory and metamemory tasks. Children were then assigned to either an organizational strategy training condition or a control condition. All children were tested on the maintenance and far-transfer of the strategy and task-related metamemory 1 week following training. Parents completed questionnaires about strategy instruction in the home. Strategy maintenance and metacognition were reassessed 6 months following training. German children were more strategic than American children. Instructed children performed better than control children. German parents reported more instruction of strategies in the home. These data suggest that formal education is responsible for aspects of cognitive development that have sometimes been viewed as a function of age
XMM-Newton evidence of shocked ISM in SN 1006: indications of hadronic acceleration
Shock fronts in young supernova remnants are the best candidates for being
sites of cosmic ray acceleration up to a few PeV, though conclusive
experimental evidence is still lacking. Hadron acceleration is expected to
increase the shock compression ratio, providing higher postshock densities, but
X-ray emission from shocked ambient medium has not firmly been detected yet in
remnants where particle acceleration is at work. We exploited the deep
observations of the XMM-Newton Large Program on SN 1006 to verify this
prediction. We performed spatially resolved spectral analysis of a set of
regions covering the southeastern rim of SN 1006. We studied the spatial
distribution of the thermodynamic properties of the ambient medium and
carefully verified the robustness of the result with respect to the analysis
method. We detected the contribution of the shocked ambient medium. We also
found that the postshock density of the interstellar medium significantly
increases in regions where particle acceleration is efficient. Under the
assumption of uniform preshock density, we found that the shock compression
ratio reaches a value of ~6 in regions near the nonthermal limbs. Our results
support the predictions of shock modification theory and indicate that effects
of acceleration of cosmic ray hadrons on the postshock plasma can be observed

Searching for the pulsar in G18.95-1.1: Discovery of an X-ray point source and associated synchrotron nebula with Chandra
Using the Chandra X-ray Observatory, we have pinpointed the location of a
faint X-ray point source (CXOUJ182913.1-125113) and an associated diffuse
nebula in the composite supernova remnant G18.95-1.1. These objects appear to
be the long-sought pulsar and its wind nebula. The X-ray spectrum of the point
source is best described by an absorbed powerlaw model with Gamma=1.6 and an
N_H of ~1x10^(22) cm^(-2). This model predicts a relatively low unabsorbed
X-ray luminosity of about L_X (0.5-8.0keV) = 4.1x10^(31)D_2^2 erg s^(-1), where
D_2 is the distance in units of 2kpc. The best-fitted model of the diffuse
nebula is a combination of thermal (kT = 0.48keV) and non-thermal (1.4 < Gamma
< 1.9) emission. The unabsorbed X-ray luminosity of L_X = 5.4x10^(33)D_2^2 erg
s^(-1) in the 0.5-8keV energy band seems to be largely dominated by the thermal
component from the SNR, providing 87% of L_X in this band. No radio or X-ray
pulsations have been reported for CXOUJ182913.1-125113. If we assume an age of
~5300yr for G18.95-1.1 and use the X-ray luminosity for the pulsar and the wind
nebula together with the relationship between spin-down luminosity (via
magnetic dipole radiation) and period, we estimate the pulsar's period to be P
= 0.4s. Compared to other rotation-powered pulsars, a magnetic field of
2.2x10^(13)G is implied by its location in the P-Pdot diagram, a value which is
