    The static and time-dependent signature of ocean–continent and ocean–ocean subduction: the case studies of Sumatra and Mariana complexes

    SUMMARY The anomalous density structure at subduction zones, both in the wedge and in the upper mantle, is analysed to shed light on the processes that are responsible for the characteristic gravity fingerprints of two types of subduction: ocean–continent and ocean–ocean. Our modelling is then performed within the frame of the EIGEN-6C4 gravitational disturbance pattern of two subductions representative of the above two types, the Sumatra and Mariana complexes, finally enabling the different characteristics of the two patterns to be observed and understood on a physical basis, including some small-scale details. A 2-D viscous modelling perpendicular to the trench accounts for the effects on the gravity pattern caused by a wide range of parameters in terms of convergence velocity, subduction dip angle and lateral variability of the crustal thickness of the overriding plate, as well as compositional differentiation, phase changes and hydration of the mantle. Plate coupling, modelled within a new scheme where the relative velocity at the plate contact results self-consistently from the thermomechanical evolution of the system, is shown to have an important impact on the gravity signature. Beyond the already understood general bipolar fingerprint of subduction, perpendicular to the trench, we obtain the density and gravity signatures of the processes occurring within the wedge and mantle that are responsible for the two different gravity patterns. To be compliant with the geodetic EIGEN-6C4 gravitational disturbance and to compare our predictions with the gravity at Sumatra and Mariana, we define a model normal Earth. Although the peak-to-peak gravitational disturbance is comparable for the two types of subductions, approximately 250 mGal, from both observations and modelling, encompassing the highest positive maximum on the overriding plates and the negative minimum on the trench, the trough is wider for the ocean–ocean subduction: approximately 300 km compared to approximately 180 km for the ocean–continent subduction. Furthermore, the gravitational disturbance pattern is more symmetric for the ocean–ocean subduction compared to the ocean–continent subduction in terms of the amplitudes of the two positive maxima over the overriding and subducting plates. Their difference is, for the ocean–ocean type, approximately one half of the ocean–continent one. These different characteristics of the two types of subductions are exploited herein in terms of the different crustal thicknesses of the overriding plate and of the different dynamics in the wedge and in the mantle for the two types of subduction, in close agreement with the gravity data

    Abstract Solar photovoltaic systems installed on homes and commercial building rooftops are deemed central for a low-carbon future. As capital costs of photovoltaics continue to fall, its role towards making buildings more sustainable and environmentally-friendly will continue to grow. Capital costs of a photovoltaic system comprise the module and balance-of-system costs. The latter refers to everything-else needed to make the photovoltaic system functional including cables, mounts, labor, etc. While modules are priced internationally, the balance-of-system cost is country-specific. Price developments of modules, which have been thoroughly studied in literature, followed an 80% learning curve. Research on the balance-of-system learning curve however, has not been as extensive. In this paper, we estimate for the first time the learning curve of balance-of-system costs in photovoltaics for more than 20 countries via an extensive dataset. Our calculations yield a global learning curve for the balance-of-system of 89%, which corresponds to a progress ratio of 11% compared with 20% for modules. Understanding the rate at which capital costs of photovoltaics are falling with such detail will aid in more effective renewable energy policy planning and budgeting. Finally, some steps requiring no financial commitment but can bring down balance-of-system costs are discussed, which greatly contribute to a cleaner and more sustainable future

    Dual targeting of glutamine and serine metabolism in acute myeloid leukemia

    Acute myeloid leukemia (AML) is a heterogeneous hematological malignancy characterized by disrupted blood cell production and function. Recent investigations have highlighted the potential of targeting glutamine metabolism as a promising therapeutic approach for AML. Asparaginases, enzymes that deplete circulating glutamine and asparagine, are approved for the treatment of acute lymphoblastic leukemia, but are also under investigation in AML, with promising results. We previously reported an elevation in plasma serine levels following treatment with Erwinia-derived asparaginase (also called crisantaspase). This led us to hypothesize that AML cells initiate the de novo serine biosynthesis pathway in response to crisantaspase treatment and that inhibiting this pathway in combination with crisantaspase would enhance AML cell death. Here we report that in AML cell lines, treatment with the clinically available crisantaspase, Rylaze, upregulates the serine biosynthesis enzymes phosphoglycerate dehydrogenase (PHGDH) and phosphoserine aminotransferase (PSAT1) through activation of the Amino Acid Response (AAR) pathway, a cellular stress response mechanism that regulates amino acid metabolism and protein synthesis under conditions of nutrient limitation. Inhibition of serine biosynthesis through CRISPR-Cas9-mediated knockout of PHGDH resulted in a ~250-fold reduction in the half-maximal inhibitory concentration (IC50) for Rylaze, indicating heightened sensitivity to crisantaspase therapy. Treatment of AML cells with a combination of Rylaze and a small molecule inhibitor of PHGDH (BI4916) revealed synergistic anti-proliferative effects in both cell lines and primary AML patient samples. Rylaze-BI4916 treatment in AML cell lines led to the inhibition of cap-dependent mRNA translation and protein synthesis, as well as a marked decrease in intracellular glutathione levels, a critical cellular antioxidant. Collectively, our results highlight the clinical potential of targeting serine biosynthesis in combination with crisantaspase as a novel therapeutic strategy for AML

    Effects of Bariatric Surgery on COVID-19: a Multicentric Study from a High Incidence Area

    Introduction: The favorable effects of bariatric surgery (BS) on overall pulmonary function and obesity-related comorbidities could influence SARS-CoV-2 clinical expression. This has been investigated comparing COVID-19 incidence and clinical course between a cohort of patients submitted to BS and a cohort of candidates for BS during the spring outbreak in Italy. Materials and Methods: From April to August 2020, 594 patients from 6 major bariatric centers in Emilia-Romagna were administered an 87-item telephonic questionnaire. Demographics, COVID-19 incidence, suggestive symptoms, and clinical outcome parameters of operated patients and candidates to BS were compared. The incidence of symptomatic COVID-19 was assessed including the clinical definition of probable case, according to World Health Organization criteria. Results: Three hundred fifty-three operated patients (Op) and 169 candidates for BS (C) were finally included in the statistical analysis. While COVID-19 incidence confirmed by laboratory tests was similar in the two groups (5.7% vs 5.9%), lower incidence of most of COVID-19-related symptoms, such as anosmia (p: 0.046), dysgeusia (p: 0.049), fever with rapid onset (p: 0.046) were recorded among Op patients, resulting in a lower rate of probable cases (14.4% vs 23.7%; p: 0.009). Hospitalization was more frequent in C patients (2.4% vs 0.3%, p: 0.02). One death in each group was reported (0.3% vs 0.6%). Previous pneumonia and malignancies resulted to be associated with symptomatic COVID-19 at univariate and multivariate analysis. Conclusion: Patients submitted to BS seem to develop less severe SARS-CoV-2 infection than subjects suffering from obesity

    Non-CO2 generating energy shares in the world : cross-country differences and polarization.

    The aim of this paper is to examine the spatial distribution of non-CO2 generating energy sources in the world for the period 1990–2009, paying special attention to the evolution of cross-country disparities. To this end, after carrying out a classical convergence analysis, a more thorough investigation of the entire distribution is presented by examining its external shape, the intra-distribution dynamics and the long-run equilibrium distribution. This analysis reveals the existence of a weak, rather insignificant, convergence process and that large crosscountry differences are likely to persist in the long-run. Next, as polarization indicators are a proper way of appraising potential conflict in international environmental negotiations, we test whether, or not, the distribution dynamics concurs with the presence of polarization. Our results indicate that two poles can be clearly differentiated, one with high and other with low non-CO2 generating energy shares. In view of these findings, and to ensure a fair transition to a sustainable energy system, the paper calls for the development of an ambitious clean energy agenda, especially in countries with low non-CO2 generating energy shares

    Geopolymer/PEG Hybrid Materials Synthesis and Investigation of the Polymer Influence on Microstructure and Mechanical Behavior

    Geopolymers are aluminosilicate inorganic polymers, obtained from the alkali activation of powders containing SiO2+Al2O3>80wt%, mainly proposed as environmentally friendly building materials. In this work, metakaolin-based geopolymers have been prepared and a water-soluble polymer, polyethylene glycol (PEG), has been added in different percentages to obtain organic-inorganic hybrid geopolymers. The influence of both the polymer amount and aging time on the structure and the mechanical behavior of the materials were investigated. FTIR spectroscopy allowed us to follow the evolution of the aluminosilicate framework during the geopolymerization process. This analysis revealed that PEG leads to a network which is rich in Al-O-Si bonds and forms H-bonds with the inorganic phase. SEM microscope showed that the two phases are interpenetrated on micrometric scales. Traction and bending tests have been carried out on appropriate samples to investigate the mechanical behavior of the obtained hybrids, showing that both PEG content and aging time affect the material behavior

    Global disparities in surgeons’ workloads, academic engagement and rest periods: the on-calL shIft fOr geNEral SurgeonS (LIONESS) study

    : The workload of general surgeons is multifaceted, encompassing not only surgical procedures but also a myriad of other responsibilities. From April to May 2023, we conducted a CHERRIES-compliant internet-based survey analyzing clinical practice, academic engagement, and post-on-call rest. The questionnaire featured six sections with 35 questions. Statistical analysis used Chi-square tests, ANOVA, and logistic regression (SPSS® v. 28). The survey received a total of 1.046 responses (65.4%). Over 78.0% of responders came from Europe, 65.1% came from a general surgery unit; 92.8% of European and 87.5% of North American respondents were involved in research, compared to 71.7% in Africa. Europe led in publishing research studies (6.6 ± 8.6 yearly). Teaching involvement was high in North America (100%) and Africa (91.7%). Surgeons reported an average of 6.7 ± 4.9 on-call shifts per month, with European and North American surgeons experiencing 6.5 ± 4.9 and 7.8 ± 4.1 on-calls monthly, respectively. African surgeons had the highest on-call frequency (8.7 ± 6.1). Post-on-call, only 35.1% of respondents received a day off. Europeans were most likely (40%) to have a day off, while African surgeons were least likely (6.7%). On the adjusted multivariable analysis HDI (Human Development Index) (aOR 1.993) hospital capacity > 400 beds (aOR 2.423), working in a specialty surgery unit (aOR 2.087), and making the on-call in-house (aOR 5.446), significantly predicted the likelihood of having a day off after an on-call shift. Our study revealed critical insights into the disparities in workload, access to research, and professional opportunities for surgeons across different continents, underscored by the HDI

    BACKGROUND: Controlled and local drug delivery systems of anti-inflammatory agents are drawing increasing attention thanks to their possible pharmaceutical and biomedical applications. These systems have extended therapeutic effects and reduced side effects. METHODS: A single-step sol-gel process was used to prepare organic-inorganic hybrid materials based on silica (SiO2) and poly-ε-caprolactone, containing ketoprofen for controlled drug delivery applications. Fourier transform infrared spectroscopy analysis proved formation of H-bonds among the carbonyl groups of the polymer chains and Si-OH group of the inorganic matrix. X-ray diffraction analysis highlighted the amorphous nature of the synthesized materials. Scanning electron microscopy and atomic force microscope topography showed their homogeneous morphology and nanostructure nature. RESULTS: The bioactivity of the synthesized hybrid materials was shown by the formation of a layer of hydroxyapatite on the surface of samples soaked in a simulated body fluid (SBF). CONCLUSION: Release kinetics in SBF were subsequently investigated by means of UV-VIS spectroscopy. A large amount of drug release occurred during the first few hours, then a slower drug release supplied a maintenance dose until the end of the experiment
