    Identification of two new intimin types in atypical enteropathogenic Escherichia coli

    Stool specimens of patients with diarrhea or other gastrointestinal alterations who were admitted to Xeral-Calde Hospital (Lugo, Spain) were analyzed for the prevalence of typical and atypical enteropathogenic Escherichia coli (EPEC). Atypical EPEC strains (eae+ bfp–) were detected in 105 (5.2%) of 2015 patients, whereas typical EPEC strains (eae+ bfp+) were identified in only five (0.2%) patients. Atypical EPEC strains were (after Salmonella) the second most frequently recovered enteropathogenic bacteria. In this study, 110 EPEC strains were characterized. The strains belonged to 43 O serogroups and 69 O:H serotypes, including 44 new serotypes not previously reported among human EPEC. However, 29% were of one of three serogroups (O26, O51, and O145) and 33% belonged to eight serotypes (O10:H–, O26:H11, O26:H–, O51:H49, O123:H19, O128:H2, O145:H28, and O145:H–). Only 14 (13%) could be assigned to classical EPEC serotypes. Fifteen intimin types, namely, α1 (6 strains), α2 (4 strains), β1 (34 strains), ξR/β2 (6 strains), γ1 (13 strains), γ2/θ (16 strains), δ/k (5 strains), ε1 (9 strains), νR/ε2 (5 strains), ζ (6 strains), ι1 (1 strain), μR/ι2 (1 strain), νB (1 strain), ξB (1 strain), and ο (2 strains), were detected among the 110 EPEC strains, but none of the strains was positive for intimin types μ1, μ2, λ, or μB. In addition, in atypical EPEC strains of serotypes O10:H–, O84:H–, and O129:H–, two new intimin genes (eae-νB and eae-ο) were identified. These genes showed less than 95% nucleotide sequence identity with existing intimin types. Phylogenetic analysis revealed six groups of closely related intimin genes: (i) α1, α2, ζ, νB, and ο; (ii) ι1 and μR/ι2; (iii) β1, ξR/β2B, δ/β2O, and κ; (iv) ε1, ξB, η1,η2, and νR/ε2; (v) γ1, μB, γ2, and θ; and (vi) λ. These results indicate that atypical EPEC strains belonging to large number of serotypes and with different intimin types might be frequently isolated from human clinical stool samples in Spain.     Drug development in Parkinson's disease: From emerging molecules to innovative drug delivery systems

    Current treatments for Parkinson’s disease (PD) are aimed at addressing motor symptoms but there is no therapy focused on modifying the course of the disease. Successful treatment strategies have been so far limited and brain drug delivery remains a major challenge that restricts its treatment. This review provides an overview of the most promising emerging agents in the field of PD drug discovery, discussing improvements that have been made in brain drug delivery for PD. It will be shown that new approaches able to extend the length of the treatment, to release the drug in a continuous manner or to cross the blood brain barrier and target a specific region are still needed. Overall, the results reviewed here show that there is an urgent need to develop both symptomatic and disease-modifying treatments, giving priority to neuroprotective treatments. Promising perspectives are being provided in this field by rasagiline and by neurotrophic factors like glial cell line-derived neurotrophic factor. The identification of disease-relevant genes has also encouraged the search for disease-modifying therapies that function by identifying molecularly-targeted drugs. The advent of new molecular and cellular targets like α-synuclein, leucine-rich repeat serine/threonine protein kinase 2 or parkin, among others, will require innovative delivery therapies. In this regard, drug delivery systems (DDS) have shown great potential for improving the efficacy of conventional and new PD therapy and reducing its side effects. The new DDS discussed here, which include microparticles, nanoparticles and hydrogels among others, will probably open up possibilities that extend beyond symptomatic relief. However, further work needs to be done before DDS become a therapeutic option for PD patients

    Palinología del Cretácico Inferior de la sección de Montoria-La Mina (Cuenca Vasco-Cantábrica, España): polen de angiospermas primitivas

    La sección del Cretácico Inferior de Montoria-La Mina se encuentra en el margen sur de la Cuenca Vasco-Cantábrica (España). Sus materiales están integrados por una sucesión de lutitas y arcillas con niveles de lignitos y areniscas que se depositaron en un cinturón deltáico, pudiéndose atribuir a la Formación Escucha. Se han obtenido asociaciones palinológicas bien conservadas con un número significativo de palinomorfos que ha permitido realizar estudios de tipo cuantitativo en ocho de los doce niveles analizados. Se ha puesto de manifiesto la existencia de una flora diversa, integrada por cuarenta y cuatro tipos diferentes de criptógamas vasculares y veintiocho de fanerógamas de las divisiones Bryophyta, Lycophyta, Pteridophyta, Pteridospermophyta, Coniferophyta, Cycadophyta/Ginkgophyta, Gnetophyta y Magnoliophyta. Las asociaciones palinológicas se encuentran dominadas por granos de polen anemófilo de Corollina obidosensis e Inaperturopollenites dubius y por esporas psiladas de Cyathidites australis, que indican la existencia de climas cálidos y secos. Es relevante el registro de polen de angiospermas primitivas en las asociaciones estudiadas, correspondiendo a formas de tamaño pequeño, reticuladas y, por lo general, columeladas. Se describen formas inaperturadas, operculadas, monosulcadas y tricotomosulcadas que se han atribuido a los grupos Pre-Afropollis, Clavatipollenites, Liliacidites, Retimonocolpites y Retimonoporites. La aparición de estos grupos polínicos permite relacionar las asociaciones registradas con las de la parte inferior de la Zona I del Grupo Potomac (Maryland, USA) y con las del Cretácico Inferior de Israel. [ABSTRACT] The Lower Cretaceous Montoria-La Mina section is located in the extreme southern part of the Basque–Cantabrian Basin (Spain). The succession might be included into the Escucha Formation and contains siltstones and clays with interbedded deltaic lignite and sandstone rocks. The palynological analysis of these rocks shows well preserved miospore assemblages with significant numbers of specimens in eight of the twelve analysed horizons. A diverse flora composed of forty four vascular cryptogams and twenty eight fanerogams are identified, included in the divisions Bryophyta, Lycophyta, Pteridophyta, Pteridospermophyta, Coniferophyta, Cycadophyta/Ginkgophyta, Gnetophyta and Magnoliophyta. The palynological assemblages are dominated by anemophilous pollen grains of Corollina obidosensis and Inaperturopollenites dubius, as well as psilate spores of Cyathidites australis, which suggest warm and dry climatic conditions. The occurrence of pollen grains of ancient angiosperms is significant in the studied assemblages. They were identified as small-sized, reticulate and generally collumelate miospores. Inaperturate, operculate, monosulcate and trichotomosulcate forms attributed to the Pre-Afropollis, Clavatipollenites, Liliacidites, Retimonocolpites and Retimonoporites pollen groups are described. The appearance of these palynological groups are compared to those in the lower part of Zone I of Potomac Group (Maryland, USA) and in the Lower Cretaceous of Israel

    In this article the network configuration for fulfillment and distribution of online orders of two British retailers is analyzed and compared. For this purpose, it is proposed a conceptual framework that consists of the key following aspects: network configuration, transportation management and location of demand. As a result is not obvious to determine the ideal centralization degree in each case. Finally, it is suggested the future development of an analytic tool that helps to choose the most appropriate model

    Direct and indirect approaches based on paper analysis by Py-GC/MS for estimating the age of documents

    The age of a relatively old document is one of the pending issues to be resolved in the field of forensic documentary examination. Although nowadays there are a variety of analytical methodologies focused in the analysis of inks for dating documents, the paper analysis has attained little attention. This work aims to develop two complementary approaches for estimating the age of documents based on paper analysis employing the pyrolysis technique coupled to gas chromatography with detection by mass spectrometry (Py-GC/MS): (i) a direct approach using the pyrolytic fingerprints and multivariate regression with artificially aged samples, and (ii) an indirect approach based on the identification of compounds characteristic of the document period. The direct approach has successfully allowed the age estimation of relatively old documents under police custody (up to 30 years of age) and the determination of a relation between the natural and the accelerated aging of paper under the used conditions. This approach is applicable to papers that have the same (or similar) composition and have been stored under comparable storage conditions. Additionally, the indirect approach is presented as an interesting perspective to ratify valuable information of the document age

    Characteristics of the Shiga-toxin-producing enteroaggregative Escherichia coli O104:H4 German outbreak strain and of STEC strains isolated in Spain

    A Shiga-toxin-producing Escherichia coli (STEC) strain belonging to serotype O104:H4, phylogenetic group B1 and sequence type ST678, with virulence features common to the enteroaggregative E. coli (EAEC) pathotype, was reported as the cause of the recent 2011 outbreak in Germany. The outbreak strain was determined to carry several virulence factors of extraintestinal pathogenic E. coli (ExPEC) and to be resistant to a wide range of antibiotics. There are only a few reports of serotype O104:H4, which is very rare in humans and has never been detected in animals or food. Several research groups obtained the complete genome sequence of isolates of the German outbreak strain as well as the genome sequences of EAEC of serotype O104:H4 strains from Africa. Those findings suggested that horizontal genetic transfer allowed the emergence of the highly virulent Shiga-toxin-producing enteroaggregative E. coli (STEAEC) O104:H4 strain responsible for theoutbreak in Germany. Epidemiologic investigations supported a linkage between the outbreaks in Germany and France and traced their origin to fenugreek seeds imported from Africa. However, there has been no isolation of the causative strain O104:H4 from any of the samples of fenugreek seeds analyzed. Following the German outbreak, we conducted a large sampling to analyze the presence of STEC, EAEC, and other types of diarrheagenic E. coli strains in Spanish vegetables. During June and July 2011, 200 vegetable samples from different origins were analyzed. All were negative for the virulent serotype O104:H4 and only one lettuce sample (0.6%) was positive for a STEC strain of serotype O146:H21 (stx1, stx2), considered of low virulence. Despite the single positive case, the hygienic and sanitary quality of Spanish vegetables proved to be quite good. In 195 of the 200 samples (98%), <10 colony-forming units (cfu) of E. coli per gram were detected, and the microbiological levels of all samples were satisfactory (<100 cfu/g). The samples were also negative for other pathotypes of diarrheagenic E. coli (EAEC, ETEC, tEPEC, and EIEC). Consistent with data from other countries, STEC belonging to serotypeO157:H7 and other serotypes have been isolated from beef, milk, cheese, and domestic (cattle, sheep, goats) and wild (deer, boar, fox) animals in Spain.     Expresión y purificación de GDNF para su microencapsulación y aplicación en la enfermedad de Parkinson

    La enfermedad de Parkinosn (EP) es un proceso neurodegenerativo del sistema nervioso central que afecta a las neuronas de dopamina de la sustancia negra, núcleo mesoencefálico del control motor. La perdida en el cerebro de este neurotransmisor vital causa los síntomas de la enfermedad. La EP afecta actualmente a 200 de cada 100.000 personas y a 2 de cada 100 entre los mayores de 60 años. En España hay unos 110.000 enfermos. Además, hoy por hoy no se conoce nada que pueda prevenir o curar la enfermedad, ni existe ninguna prueba de laboratorio que permita diagnosticarla. Recentiemente se ha demostrado que el GDNF, factor neurotrófico derivado de las células gliales, es capaz de proteger las neuronas dopaminérgicas e incluso inducir la regeneración del tejido dopaminérgico dañado in vivo. El objetivo del trabajo fue diseñar y desarrolar un método de expresión y purificación de GDNF bioactivo para su posterior microencapsulación y aplicación en la enfermedad de Parkison. El sistema escogido para expresar el GDNF fue el sistema de células eucariotas de mamífero. El vector utilizado para la producción del GDNF en células eucariotas fue el pDEST26 (Tecnología Gateway de Invitrogen). Como sistema de expresión de GDNF se utilizaron las líneas celulares eucariota BHK, 293 y COS 7. Estas células fueron cultivadas en medio D-MEM (Invitrogen) complementado con un 10% de suero fetal bovino (FBS) y Penicilina/Streptomicina (100u/ml) (Invitrogen). La transfección se realizó con Lipofectamine Plus (Invitrogen). Se analizó la expresión de GDNF a nivel de mRNA mediante PCR y a nivel de proteína mediante Western Blot del medio condicionado. Los clones positivos se crecieron en botellas de cultivo de 850 cm2 (Corning) y se realizaron ciclos de recolección del medio. Cada ciclo fue analizado por SDS-PAGE y Western Blot. Para evaluar la actividad de la proteína se ha desarrollado un ensayo de actividad en el que se demuestra la diferenciación morfológica de células PC-12 inducida por GDNF. La presencia de los receptores GFRa1 y RET, necesarios para que el GDNF ejerza su acción, fue determinada por PCR. Las conclusiones obtenidas de este estudio son la obtención de GDNF recombinante a partir de un sistema de expresión en células eucariotas, el desarrollo de un protocolo para su posterior purificación y la obtención de GDNF recombinante biológicamente activo

    Arsenic in Water Resources of the Southern Pampa Plains, Argentina

    Confronted with the need for accessible sources of good quality water and in view of the fact that the threat to public health posed by arsenic occurs mainly through the ingestion of contaminated drinking water, the presence and distribution of arsenic was evaluated in the southern Pampa Plains of Bahía Blanca district in Argentina. The findings show variable concentrations of arsenic in a complex distribution pattern. Complementary information is provided on the behavior of the groundwater resource and its salinity in terms of dissolved ions. Groundwater is the most severely affected, 97% of the samples exceeding the guideline value for arsenic in drinking water as recommended by the WHO (Guidelines for Drinking Water Quality, 2004). and showing maximum concentrations of up to 0.30 mg/L. Informing those responsible for preventive medicine and alerting the community at large will facilitate measures to mitigate exposure and ensure the safety of drinking water

    An empirical examination of the role of emotional knowledge in the teaching profession

    Modern society has plunged us into a maelstrom of telematics information with little control. Teachers from any field must be trained in order to pass on the confidence and values to withstand this media pressure to their students. To this end, one of the areas of knowledge on which greater emphasis should be placed is that related to emotional intelligence. The study reported on here focused on this type of knowledge among professional music studies teachers in conservatories, asking what the perception of this group on the aforementioned area of knowledge is and to what extent it has been received during their initial training. For this investigation we used a quantitative methodological approach, and via descriptive analysis, a non-parametric Wilcoxon signed-rank test, and inferential analysis, we found significant differences regarding the two scales considered: level of agreement and emotional intelligence training received. Furthermore, regarding the descriptive variables, significant differences were also found through the application of the Mann-Whitney U and Kruskal-Wallis test.Faculty of Education, Economy and Technology of Ceut
