    Morphology and ultrastructure of the pro- and postpharyngeal glands in workers of Pachycondyla sennaarensis

    The pro- and postpharyngeal glands of Brachyponera sennaarensis both appear as globular formations at a general anatomical level. However, only for the propharyngeal gland do these formations correspond with spherical secretory cells with diameters of 30-40 µm. For the lobed postpharyngeal gland, in contrast, this globular appearance is caused by the bulbous protrusions of the epithelial cells. This lobed appearance and globular cell shape also occur in the postpharyngeal glands of other Ponerinae and thus may represent a phylogenetic character. At the ultrastructural level, the propharyngeal gland cells are characterized by a well-developed granular endoplasmic reticulum, which is in agreement with its presumed production of digestive enzymes. The postpharyngeal gland cells contain a well-developed smooth endoplasmic reticulum, which allows the production of a non-proteinaceous secretion

    Pleiotropic and Novel Phenotypes in The \u3cem\u3eDrosophila\u3c/em\u3e Gut Caused by Mutation of \u3cem\u3eDrop-Dead\u3c/em\u3e

    Normal gut function is vital for animal survival, and deviations from such function can contribute to malnutrition, inflammation, increased susceptibility to pathogens, diabetes, neurodegenerative diseases, and cancer. In the fruit fly Drosophila melanogaster, mutation of the gene drop-dead (drd) results in defective gut function, as measured by enlargement of the crop and reduced food movement through the gut, and drd mutation also causes the unrelated phenotypes of neurodegeneration, early adult lethality and female sterility. In the current work, adult drd mutant flies are also shown to lack the peritrophic matrix (PM), an extracellular barrier that lines the lumen of the midgut and is found in many insects including flies, mosquitos and termites. The use of a drd-gal4 construct to drive a GFP reporter in late pupae and adults revealed drd expression in the anterior cardia, which is the site of PM synthesis in Drosophila. Moreover, the ability of drd knockdown or rescue with several gal4 drivers to recapitulate or rescue the gut phenotypes (lack of a PM, reduced defecation, and reduced adult survival 10–40 days post-eclosion) was correlated to the level of expression of each driver in the anterior cardia. Surprisingly, however, knocking down drd expression only in adult flies, which has previously been shown not to affect survival, eliminated the PM without reducing defecation rate. These results demonstrate that drd mutant flies have a novel phenotype, the absence of a PM, which is functionally separable from the previously described gut dysfunction observed in these flies. As the first mutant Drosophila strain reported to lack a PM, drd mutants will be a useful tool for studying the synthesis of this structure

    Metformin promotes lifespan through mitohormesis via the peroxiredoxin PRDX-2

    The antiglycemic drug metformin, widely prescribed as first-line treatment of type II diabetes mellitus, has lifespan-extending properties. Precisely how this is achieved remains unclear. Via a quantitative proteomics approach using the model organism Caenorhabditis elegans, we gained molecular understanding of the physiological changes elicited by metformin exposure, including changes in branched-chain amino acid catabolism and cuticle maintenance. We show that metformin extends lifespan through the process of mitohormesis and propose a signaling cascade in which metformin-induced production of reactive oxygen species increases overall life expectancy. We further address an important issue in aging research, wherein so far, the key molecular link that translates the reactive oxygen species signal into a prolongevity cue remained elusive. We show that this beneficial signal of the mitohormetic pathway is propagated by the peroxiredoxin PRDX-2. Because of its evolutionary conservation, peroxiredoxin signaling might underlie a general principle of prolongevity signaling

    Differences in Climbing Ability of Cimex lectularius and Cimex hemipterus (Hemiptera: Cimicidae)

    The climbing abilities of two bed bug species, Cimex lectularius L. and Cimex hemipterus (F.), were determined by evaluating their escape rates from smooth surface pitfall traps using four commercial bed bug monitors (Verifi Bed Bug Detector, ClimbUp Insect Interceptor, BlackOut Bed Bug Detector, and SenSci Volcano Bed Bug Detector). All detectors were used in the absence of lures or attractants. Unlike C. lectularius, adult C. hemipterus were able to escape from all traps. On the other hand, no or a low number nymphs of both species escaped, depending on the evaluated traps. Examination of the vertical friction force of adults of both species revealed a higher vertical friction force in C. hemipterus than in C. lectularius. Scanning electron microscope micrograph observation on the tibial pad of adult bed bugs of C. hemipterus showed the presence of a greater number of tenent hairs on the tibial pad than on that of adult C. lectularius. No tibial pad was found on the fourth and fifth instars of both species. Near the base of the hollow tenent hairs is a glandular epithelium that is better developed in adult C. hemipterus than in adult C. lectularius. This study highlights significant morphological differences between C. lectularius and C. hemipterus, which may have implications in the monitoring and management of bed bug infestations

    An Overlooked Mandibular-Rubbing Behavior Used during Recruitment by the African Weaver Ant, Oecophylla longinoda

    In Oecophylla, an ant genus comprising two territorially dominant arboreal species, workers are known to (1) use anal spots to mark their territories, (2) drag their gaster along the substrate to deposit short-range recruitment trails, and (3) drag the extruded rectal gland along the substrate to deposit the trails used in long-range recruitment. Here we study an overlooked but important marking behavior in which O. longinoda workers first rub the underside of their mandibles onto the substrate, and then—in a surprising posture—tilt their head and also rub the upper side of their mandibles. We demonstrate that this behavior is used to recruit nestmates. Its frequency varies with the rate at which a new territory, a sugary food source, a prey item, or an alien ant are discovered. Microscopy analyses showed that both the upper side and the underside of the mandibles possess pores linked to secretory glands. So, by rubbing their mandibles onto the substrate, the workers probably spread a secretion from these glands that is involved in nestmate recruitment

    Worker policing in the German wasp Vespula germanica

    In some ants, bees, and wasps, workers kill or "police” male eggs laid by other workers in order to maintain the reproductive primacy of the queen. Kin selection theory predicts that multiple mating by the queen is one factor that can selectively favor worker policing. This is because when the queen is mated to multiple males, workers are more closely related to the queen's sons than to the sons of other workers. Earlier work has suggested that reproductive patterns in the German wasp Vespula germanica may contradict this theory, because in some colonies a large fraction of the adult males were inferred to be the workers' sons, despite the effective queen mating frequency being greater than 2 (2.4). In the present study, we reexamine the V. germanica case and show that it does support the theory. First, genetic analysis confirms that the effective queen mating frequency is high, 2.9, resulting in workers being more related to the queen's sons than to other workers' sons. Second, behavioral assays show that worker-laid eggs are effectively killed by other workers, despite worker-laid eggs having the same intrinsic viability as queen-laid ones. Finally, we estimate that approximately 58.4% of the male eggs but only 0.44% of the adult males are worker derived in queenright colonies, consistent with worker reproduction being effectively police
