    Intestinal microbiota, probiotics and mental health: from Metchnikoff to modern advances: Part II – contemporary contextual research

    In recent years there has been a renewed interest concerning the ways in which the gastrointestinal tract – its functional integrity and microbial residents – might influence human mood (e.g. depression) and behavioral disorders. Once a hotbed of scientific interest in the early 20th century, this area lay dormant for decades, in part due to its association with the controversial term ‘autointoxication’. Here we review contemporary findings related to intestinal permeability, small intestinal bacterial overgrowth, lipopolysaccharide endotoxin (LPS) exposure, D-lactic acid, propionic acid, and discuss their relevance to microbiota and mental health. In addition, we include the context of modern dietary habits as they relate to depression, anxiety and their potential interaction with intestinal microbiota

    Mental health disorders, depression in particular, have been described as a global epidemic. Research suggests that a variety of lifestyle and environmental changes may be driving at least some portion of the increased prevalence. One area of flourishing research involves the relationship between the intestinal microbiota (as well as the related functional integrity of the gastrointestinal tract) and mental health. In order to appreciate the recent scientific gains in this area, and its potential future directions, it is critical to review the history of the topic. Probiotic administration (e.g. Lactobacillus) and fecal microbiota transfer for conditions associated with depression and anxiety is not a new concept. Here, in the first of a 3-part series, we begin by reviewing the origins of the contemporary research, providing a critical appraisal of what has become a revisionist history of the controversial term ‘autointoxication’. We argue that legitimate interests in the gut-brain-microbiota connection were obscured for decades by its association with a narrow historical legacy. Historical perspectives provide a very meaningful context to the current state of the contemporary research as outlined in parts II and III

    Rapid scientific and technological advances have allowed for a more detailed understanding of the relevance of intestinal microbiota, and the entire body-wide microbiome, to human health and well-being. Rodent studies have provided suggestive evidence that probiotics (e.g. lactobacillus and bifidobacteria) can influence behavior. More importantly, emerging clinical studies indicate that the administration of beneficial microbes, via supplementation and/or fecal microbial transplant (FMT), can influence end-points related to mood state (glycemic control, oxidative status, uremic toxins), brain function (functional magnetic resonance imaging fMRI), and mental outlook (depression, anxiety). However, despite the advances in the area of gastro-biological psychiatry, it becomes clear that there remains an urgent need to explore the value of beneficial microbes in controlled clinical investigations. With the history explored in this series, it is fair to ask if we are now on the cusp of major clinical breakthroughs, or are we merely in the quicksand of Autointoxication II

    Fermented foods, microbiota, and mental health: ancient practice meets nutritional psychiatry

    The purposeful application of fermentation in food and beverage preparation, as a means to provide palatability, nutritional value, preservative, and medicinal properties, is an ancient practice. Fermented foods and beverages continue to make a significant contribution to the overall patterns of traditional dietary practices. As our knowledge of the human microbiome increases, including its connection to mental health (for example, anxiety and depression), it is becoming increasingly clear that there are untold connections between our resident microbes and many aspects of physiology. Of relevance to this research are new findings concerning the ways in which fermentation alters dietary items pre-consumption, and in turn, the ways in which fermentation-enriched chemicals (for example, lactoferrin, bioactive peptides) and newly formed phytochemicals (for example, unique flavonoids) may act upon our own intestinal microbiota profile. Here, we argue that the consumption of fermented foods may be particularly relevant to the emerging research linking traditional dietary practices and positive mental health. The extent to which traditional dietary items may mitigate inflammation and oxidative stress may be controlled, at least to some degree, by microbiota. It is our contention that properly controlled fermentation may often amplify the specific nutrient and phytochemical content of foods, the ultimate value of which may associated with mental health; furthermore, we also argue that the microbes (for example, Lactobacillus and Bifidobacteria species) associated with fermented foods may also influence brain health via direct and indirect pathways

    Cytokine responses to exercise and activity in patients with chronic fatigue syndrome: Case control study

    Chronic fatigue syndrome (CFS) is characterized by fatigue after exertion. A systematic review suggested that transforming growth factor beta (TGF-β) concentrations are often elevated in cases of CFS when compared to healthy controls. This study attempted to replicate this finding, and investigate whether post-exertional symptoms were associated with altered cytokine protein concentrations and their RNA in CFS patients. Twenty-four patients fulfilling Centers for Disease Control criteria for CFS, but with no comorbid psychiatric disorders, were recruited from two CFS clinics in London, UK. Twenty-one healthy, sedentary controls were matched by gender, age, and other variables. Circulating proteins and RNA were measured for TGF-β, TNF, IL-8, IL-6 and IL-1β. We measured six further cytokine protein concentrations (IL-2, IL-4, IL-5, IL-10, IL-12p70, and IFN-γ). Measures were taken at rest, and before and after both commuting and aerobic exercise. CFS cases had higher TGF-β protein levels compared to controls at rest (median (quartiles) = 43.9 (19.2, 61.8) versus 18.9 (16.1, 30.0) ng/ml) (p = 0.003), and consistently so over a nine-day period. However, this was a spurious finding due to variation between different assay batches. There were no differences between groups in changes to TGF-β protein concentrations after either commuting or exercise. All other cytokine protein and RNA levels were similar between cases and controls. Post-exertional symptoms and perceived effort were not associated with any increased cytokines. We were unable to replicate previously found elevations in circulating cytokine concentrations, suggesting that elevated circulating cytokines are not important in the pathophysiology of CFS

    Combining unassisted and robot-guided practice benefits motor learning for a golf putting task

    Robotic guidance has been employed with limited effectiveness in neurologically intact and patient populations. For example, our lab has effectively used robotic guidance to acutely improve movement smoothness of a discrete trajectory without influencing movement endpoint distributions. The purpose of the current study was to investigate the efficacy of combining robotic guidance and unassisted trials in the learning of a golf putting task. Participants completed a pre-test, an acquisition phase, and an immediate and delayed (24-hour) post-test. During the pre-test, kinematic data from the putter was converted into highly accurate, consistent, and smooth trajectories delivered by a robot arm. During acquisition, three groups performed putts towards three different targets with robotic guidance on either 0%, 50%, or 100% of acquisition trials. Only the 50% guidance group statistically reduced both the ball endpoint distance and variability between the pre-test and the immediate or 24-hr post-test. The results of the 50% guidance group yielded seminal evidence that combining both unassisted and robotic guidance trials (i.e., mixed practice) could facilitate at least short-term motor learning for a golf putting task. Such work is relevant to incorporating robotic guidance in sport skills and other practical areas (e.g., rehabilitation)

    What’s your number? The effects of trial order on the one-target advantage

    When moving our upper-limb towards a single target, movement times are typically shorter than when movement to a second target is required. This is known as the one-target advantage. Most studies that have demonstrated the one-target advantage have employed separate trial blocks for the one- and two-segment movements. To test if the presence of the one-target advantage depends on advance knowledge of the number of segments, the present study investigated whether the one-target advantage would emerge under different trial orders/sequences. One- and two-segment responses were organized in blocked (i.e., 1-1-1, 2-2-2), alternating (i.e., 1-2-1-2-1-2), and random (i.e., 1-1-2-1-2-2) trial sequences. Similar to previous studies, where only blocked schedules have typically been utilized, the one-target advantage emerged during the blocked and alternate conditions, but not in the random condition. This finding indicates that the one-target advantage is contingent on participants knowing the number of movement segments prior to stimulus onset

    Humans versus robots: Converting golf putter trajectories for robotic guidance

    Robotic devices are used to provide physical guidance when teaching different movements. To advance our knowledge of robotic guidance in training complex movements, this investigation tested different kinematic data filtering methods of individual’s golf putts to convert them into trajectories to be employed by a robot arm. The purpose of the current study was to identify a simple filtering method to aptly replicate participants’ individual golf putter trajectories which could be used by the robot to execute them with greater consistency and accuracy than their human counterpart. Participants putted towards 3 targets where three-dimensional data of the putter’s head was filtered and then fitted by using one- or two-dimensions of the participant’s putter head trajectories. As expected, both filtering methods employed with the robot outperformed the human participants in ball endpoint accuracy and consistency. Further, after comparing the filtered to the human participants trajectories, the two-dimensional method best replicated the kinematic features of human participants natural putter trajectory, while the one-dimensional method failed to replicate participant’s backstroke position. This investigation indicates that a two-dimensional filtering method, using Y-forward and Z-vertical position data, can be used to create accurate, consistent, and smooth trajectories delivered by a robot arm

    Train less and still similarly improve? Maturational growth is more influential than training engagement on performance indices development in volleyball

    Identifying factors which more or less account for performance improvement during developmental stages is essential for sports science knowledge and coaching practice. Accordingly, this study examined the longitudinal, changing, influences of Volleyball-specific Training Engagement (VTE) and Maturity Status on physical test performance development. Participants were N = 139 Swiss female competitive volleyball players, aged 10–14 years at baseline (M = 12.93, SD = 1.15 years). Annually for 3 years, participants completed the 9-3-6-3-9 Agility Sprint; Standing Long Jump [SLJ] and Jump & Reach test. Linear Mixed Models (LMMs) examined longitudinal independent and interactive relationships between VTE (hours/week) and Maturity Status (YPHV) with test performance indices. LMMs identified both interactive and independent relationships on test indices. Interactions highlighted the influence of VTE generally reduced during peak-post maturational stages (− 0.5–2.5 YPHV), while maturational growth was predominantly more influential on performance development. Findings identified that lowered weekly VTE during maturational peak-post growth periods led to equivalent performance development. With maturational growth more influential, findings highlight the potential to misattribute longitudinal performance development toward training engagement and question the benefit of heightened physiological-focused engagement circa-post PHV stages. However, such questioning may not necessarily apply to skill acquisition or technical-focused training
