Foglio di via e altri versi di Franco Fortini. Edizione critica e commentata

    Get PDF

In-Vivo Real-Time Control of Protein Expression from Endogenous and Synthetic Gene Networks

We describe an innovative experimental and computational approach to control the expression of a protein in a population of yeast cells. We designed a simple control algorithm to automatically regulate the administration of inducer molecules to the cells by comparing the actual protein expression level in the cell population with the desired expression level. We then built an automated platform based on a microfluidic device, a time-lapse microscopy apparatus, and a set of motorized syringes, all controlled by a computer. We tested the platform to force yeast cells to express a desired fixed, or time-varying, amount of a reporter protein over thousands of minutes. The computer automatically switched the type of sugar administered to the cells, its concentration and its duration, according to the control algorithm. Our approach can be used to control expression of any protein, fused to a fluorescent reporter, provided that an external molecule known to (indirectly) affect its promoter activity is available

The Correlation between Retinal and Choroidal Thickness with Age-Related White Matter Hyperintensities in Progressive Supranuclear Palsy

Choroid; Optical coherence tomography; RetinaCoroide; Tomografia de coherència òptica; RetinaCoroides; Tomografía de coherencia óptica; RetinaProgressive supranuclear palsy (PSP) is a rare neurodegenerative disease. Recently, several retinal layers in PSP compared to healthy controls. were found to be thinner. However, no studies evaluating the correlation between retinal layers and cerebral white matter changes, nor eventual choroidal changes in PSP, have been conducted so far. The goals of the present study were to explore potential differences in choroidal structure between PSP and healthy controls, and to describe the relationship between retinal layers’ thickness and volume, using spectral-domain optical coherence tomography (SD-OCT) and age-related white matter change scores (ARWMC) using magnetic resonance imaging (MRI) of the brain. Choroidal structures of 26 PSP patients and 26 healthy controls using standard SD-OCT with an enhanced depth imaging (EDI) approach were analyzed; then, retinal the structures of 16 of these PSP patients using standard SD-OCT were examined; finally, the same patients underwent brain MRI, and their cerebral white matter changes were calculated. Non-statistically significant differences between PSP patients’ and healthy controls’ choroidal structure were found. On the contrary, PSP patients’ inner retinal layers (INR), retinal pigmented epithelium (RPE) and all retinal layers’ thicknesses in the macular region were found to be significantly correlated with ARWMC, independently from age and axial length (AL). PSP patients’ neurological alterations go hand in hand with retinal ones, independently from age and axial length. Our results suggest a mutual relationship between cerebral and retinal structure pathological alterations. On the other hand, no significant differences in the choroidal evaluation compared to healthy controls have been found

Background: The TIMI RISK (TR) score for ST-Evaluation Myocardial Infarction (STEMI) was originally describes by Morrow et al. (2000), basead on 8 variables. It was used in the In TIME study as 30-day mortality predictor and several studies successfully relate TR as prognosis index for STEMI, although few reports evaluate lete evolution of these patients and the relation whith the score. Objective: Evaluating the evolution of the STEMI interned patients in University-Based Hospitals after 6 months and the relation with the TR stratification. Methods and Results: 89 patients were clinicallyfollowed up in University-Based Hospital for 6 months after the initial determination of the TR score realized during hospital admission. The following parameters were evaluated: death, readmission for cardiovascular event, and revascularization necessity. They were classified in 3 groups according the score: Group I (0, 1, 2); Group II (3, 4, 5) and Group III (>5), were submit to statistic analysis by chisquaretest. The results of death, readmission in coronary-care unit and revascularization were respectively: GI: 4,1%, 25,0%, 20,8%; GII: 10,0%, 35,0%, 30,0%; GIII: 52,0%, 60,0%, 48,0%. Conclusion: TR score maintained significant events increase on Groups II and III comparing to Group I; presenting the possible score applicability also at long term as predictor (6 moths)

Nowadays, cardiovascular diseases are the principal cause of morbimortality, specially the coronary artery disease. New physiopathological acknowledgement indicates new mechanisms for myocardial injury, still not quite studied. The aim of this study is to analyze the effect of N2-Mercaptopropionilglicine (N2-MPG) as a myocardial protection agent during ischemya/reperfusion based on the preservationof the necrosis area. Sixteen dogs where divided into two groups of eight dogs each one: Group I (N2-MPG) and Group II (control). After the experimental acute myocardialinfarction reproduction, the hearts where colored (TTC/Blue Evans dye) and weighed for the myocardial preservation analysis. The amount of necrosis area were: Group I(73,14%) and Group II (49,98%) through the Mann-Whitney method. The necrosis area reduction showed an average reduction of 23,16%, concluding that N2-MPG has aprotection effect during myocardial ischemya/reperfusion.

    Get PDF
New technologies, namely eHealth platforms, are being used more than ever before. These platforms enable older people to have a more independent lifestyle, enhance their participation, and improve their well-being. Information and communication technologies are expected to be linked to the triad of aging, social inclusion, and active participation, which is in line with the implementation of Smart Healthy and Age-Friendly Environments. This scoping review aimed to map eHealth platforms designed to promote autonomous life and active aging. The Joanna Briggs Institute methodology and the PRISMA-ScR checklist were used. A search was conducted on MEDLINE (via PubMed), CINAHL Complete (via EBSCOhost), Scopus, Cochrane Database of Systematic Reviews (via EBSCOhost), SciELO, DART-Europe, CAPES, and MedNar databases. Fourteen studies were included. This scoping review synthesized information on eHealth platforms designed to promote active living, their domains of intervention, and the outcomes assessed in those studies that have implemented and evaluated these eHealth platforms
