    "From womb to tomb; we're bound to others": Microbiome in forensic science

    Microbiome is a new field of interest in clinical medicine with high potential in forensic medicine. It could be used in several applications, such as post-mortem interval (PMI) estimation, personal identification, differential diagnosis of cause of death and toxicology. Regarding PMI, during the decomposition of a corpse, the passage of time involves changing in microbial population both outside and inside the corpse but also in surrounding soil (cadaver decomposition island). These variations could be hypothetically used as PMI indicators (microbial clock), even thanks to the development of machine learning approach. Another potential use of skin and saliva microbiome is personal identification thanks to its inter-individual variability and tendency to remain unvarying over time. It may also be helpful to link a person to a specific object that has been touched (microbial fingerprint). Furthermore, we could infer some information about health state of human subjects, comparing post-mortem and ante-mortem microbiome, but this field of research is quite new and needs further studies. Moreover, we have to consider the influence of microbiome metabolism in post-mortem toxicological evaluation; microbes could alter substances concentrations - for example of ethanol, gamma-hydroxybutyric acid (GHB) and nitrobenzodiazepines - due to enzymatic degradation and individual microbial metabolism. Finally, integration of microbiome and human being's transcriptomic analysis may be helpful to depict their complex interactions even after death

    Heterocycle-containing tranylcypromine derivatives endowed with high anti-LSD1 activity

    As regioisomers/bioisosteres of 1a, a 4-phenylbenzamide tranylcypromine (TCP) derivative previously disclosed by us, we report here the synthesis and biological evaluation of some (hetero)arylbenzoylamino TCP derivatives 1b-6, in which the 4-phenyl moiety of 1a was shifted at the benzamide C3 position or replaced by 2- or 3-furyl, 2- or 3-thienyl, or 4-pyridyl group, all at the benzamide C4 or C3 position. In anti-LSD1-CoREST assay, all the meta derivatives were more effective than the para analogues, with the meta thienyl analogs 4b and 5b being the most potent (IC50 values Π0.015 and 0.005 lM) and the most selective over MAO-B (selectivity indexes: 24.4 and 164). When tested in U937 AML and prostate cancer LNCaP cells, selected compounds 1a,b, 2b, 3b, 4b, and 5a,b displayed cell growth arrest mainly in LNCaP cells. Western blot analyses showed increased levels of H3K4me2 and/or H3K9me2 confirming the involvement of LSD1 inhibition in these assays

    Cats’ and dogs’ welfare: text mining and topics modeling analysis of the scientific literature

    Animal welfare is a field with increasing significance and has been raising huge concerns of the public and the political stage. Cats and dogs possess an important role in human life, but their welfare is not always secured from a legal aspect. This review aimed to describe the evolution and geographical distribution of “cats and dogs” and “puppies and kittens” welfare literature over the last 40 years, distinguish the main research topics studied and highlight gaps in knowledge. A search using Scopus¼ was performed with different search strings and predetermined filters as time range, language, and subject area. A total of 2,725 scientific literature records were retrieved but only the ones that referred to cats and dogs’ welfare aspects were retained. The final 1,775 records were processed through descriptive statistics, and text mining and topic analysis procedures were performed on their titles and abstracts. The results showed that the number of studies has been increasing, especially in Europe and North America. “Shelter” was the most frequent word, followed by “behavior,” “owner” and “adopt.” The nine topics that emerged from the analysis were breeding, stress and housing conditions, welfare and pain assessment, public health, shelter management and euthanasia, behavioral problems, health issues and management, human-animal interaction, and owners’ and veterinarians’ perceptions. While stress and housing conditions, public health, and owners’ and veterinarians’ perceptions were the most studied topics, human-animal interaction was the least studied. This review confirmed the increasing research and interest in cats’ and dogs’ welfare and showed gaps in knowledge where further studies are needed

    Staging and storing data sprint-based research results: a communication design approach

    Data sprints are multidisciplinary, time-limited, practice-oriented, and group activities that explore complex issues through data gathering, visualization, and analysis. Data sprints usually conclude with final presentations where participants showcase the research process they followed and the achieved results. Although this activity is now widespread in some research centers, there does not appear to be a data-sprint tailored dissemination format that can support the organization, archive, and online diffusion of results. Starting from the experience of a data sprint on Digital Methods, the paper describes the design process of a digital format which condenses and reinterprets good practices already in use in current supports for disseminating data sprints results. The design process of the format is structured in two steps: (i) designing and (ii) staging the format. The former aims to dissect and recompose data sprint results in hierarchical block structures filled with variable contents; the latter describes outcomes that emerged during a preliminary evaluation phase with experts and a stress-test activity. The format is a container that participants can fill at the end of a data sprint to organize, archive, and present research results. Evaluation activities confirmed the efficiency of the format, which is distinguished by its nested structure and flexibility with different content. Future research opportunities concern the ultimate implementation of the format as a public Content Management System to be used even outside data sprint contexts, such as disseminating more thorough research and teaching students how to structure Digital Methods research

    Punctual and kinetic MRD analysis from the Fondazione Italiana Linfomi MCL0208 phase III trial in mantle cell lymphoma

    Minimal residual disease (MRD) analysis is a known predictive tool in mantle cell lymphoma (MCL). We describe MRD results from the Fondazione Italiana Linfomi phase III MCL0208 prospective clinical trial assessing lenalidomide maintenance vs observation after autologous transplantation (ASCT), in the first prospective comprehensive analysis of different techniques, molecular markers, and tissues (peripheral blood, PB, and bone marrow, BM), taken at well-defined timepoints. Among the 300 patients enrolled, a molecular marker was identified in 250 (83%), allowing us to analyze 234 patients and 4351 analytical findings from 10 timepoints. ASCT induced high rates of molecular remission (91% in PB and 83% in BM, by quantitative real-time PCR [RQ-PCR]). Nevertheless, the number of patients with persistent clinical and molecular remission decreased over time in both arms (up to 30% after 36 months). MRD predicted early progression and long-term outcome, particularly from 6 months after ASCT (6-month TTP HR 3.83, p<0.001). In single-timepoint analysis, BM outperformed PB, and RQ-PCR was more reliable, while nested PCR appeared applicable to a larger number of patients (234 vs 176). To improve MRD performance we developed a time-varying kinetic model, based on regularly updated MRD results and the Mantle Cell Lymphoma International Prognostic Index, showing an area under the ROC curve (AUROC) of up to 0.87 using BM. Most notably, PB reached an AUROC of up to 0.81: with kinetic analysis it was comparable to BM in performance. MRD is a powerful predictor over the entire natural history of MCL and suitable for models with continuous adaptation of patient risk.     Patients undergoing allogeneic hematopoietic stem cell transplant (HSCT) are exposed to a number of neurological complications that may be related to drugs, infections, metabolic alterations, cerebrovascular events and immune-\uadmediated disorders including myositis, myasthenia gravis, Guillain-\uadBarr\ue8-\uadlike demyelinating polyneuropathy and central nervous system (CNS) manifestations of graft versus host disease (GVHD). The multifactorial etiology of neurological complications in HSCT patients makes diagnosis difficult. However a timely and rigorous characterization of such complications should be obtained in the attempt to avoid fatal outcomes or long-\uadterm effects. Data regarding neurological complications in HSCT patients derives from small series and varies largely in respect to incidence and severity. Aim of this study is to describe incidence, characteristics and outcome of neurological complications in a large series of consecutive HSCT patients

    Predicting needlestick and sharps injuries in nursing students: Development of the SNNIP scale

Aim: To develop an instrument to investigate knowledge and predictive factors of needlestick and sharps injuries (NSIs) in nursing students during clinical placements. Design: Instrument development and cross-sectional study for psychometric testing. Methods: A self-administered instrument including demographic data, injury epidemiology and predictive factors of NSIs was developed between October 2018–January 2019. Content validity was assessed by a panel of experts. The instrument's factor structure and discriminant validity were explored using principal components analysis. The STROBE guidelines were followed. Results: Evidence of content validity was found (S-CVI 0.75; I-CVI 0.50–1.00). A three-factor structure was shown by exploratory factor analysis. Of the 238 participants, 39% had been injured at least once, of which 67.3% in the second year. Higher perceptions of “personal exposure” (4.06, SD 3.78) were reported by third-year students. Higher scores for “perceived benefits” of preventive behaviours (13.6, SD 1.46) were reported by second-year students
