    Gastric adenomas: relationship between clinicopathological findings, Helicobacter pylori infection, APC mutations and COX-2 expression.

    Gastric adenomas are rare neoplastic growths characterized by localized polypoid proliferations of dysplastic epithelium that tend to progress to infiltrating adenocarcinoma. Therefore, the identification of molecular markers that could reliably recognize adenomas at risk of progression is advocated in the clinical management. In this study we investigated, in a series of gastric adenoma specimens from an area at high risk of gastric cancer, the relationship between clinicopathological characteristics of adenoma and Helicobacter pylori infection, APC mutational status, and COX-2 and the down-stream enzyme mPGES1 expression. Helicobacter pylori infection, detected in 24%, and 33% by histology and PCR analyses, respectively, did not show any relationship with growth pattern, localization, size, dysplasia grade and presence of synchronous cancer. Pathogenetic mutations of MCR region (codons 1269-1589) of the APC gene were detected only in one case corresponding to a single, small size, low grade, H. pylori-negative adenoma. The expression of COX-2 largely matched that of mPGES(1). Both were overexpressed in 79% of cases showing a relationship with high-grade dysplasia, size >10 mm and presence of a synchronous carcinoma. In conclusion, COX-2 may play a key role in the development and progression of gastric adenoma and could be an attractive target in the management of gastric adenoma at major risk of cancer development

    SARS-CoV-2 seroprevalence on the north coast of Peru: A cross-sectional study after the first wave

    During the first wave, COVID-19 had devastating effects on developing countries like Peru, which reported more than 900,000 cases of the disease and more than 36,000 confirmed deaths from it. The informal settlements are counted in COVID-19 regional statistics, but they are very different from the rest of the region in terms of sanitation, access to water and sewage, housing conditions, and others. To make good decisions about public health, it is important to understand how the disease is spread in informal settlements. We did an observational, cross-sectional study in Puerto Pizarro, a remote port village in Tumbes, from November 11, 2020, to November 30, 2020, to assess the seroprevalence of COVID-19 after the first wave, as well as the sociodemographic factors and symptoms linked to a positive COVID-19 antibody lateral flow test. 1391 people older than 2 years old were given census and symptom questionnaires. They were also tested for the presence of anti-SARS-CoV-2 antibodies (IgG and IgM) in their blood. This study shows an adjusted seroprevalence of 24.82 percent (95% CI 22.49–27.25) posterior to the first wave of COVID-19 in Tumbes. Women had higher adjusted seroprevalence (28.03% vs. 21.11% [95% CI 24.83–31.41], p = 0.002). Extrapolating our adjusted prevalence of 24.82 per 100 persons to Tumbes (N = 251,541), 62,432 people were infected during the first wave, which is too high for adequate primary care in health institutions. Puerto Pizarro’s causespecific mortality rate was 198.49 per 100,000 inhabitants (deaths documented between May and December 2020), which was higher than Tumbes’ up to December 2020 (137.6 per 100,000 inhabitants). The presence of mostly symptomatic patients at health care facilities, the limited use of social networks, the scarce availability of eHealth technologies in government institutions, unused emergency telephone numbers, and the fear of dying if referred to the hospital may have led to underreporting cases. Participation of the community in epidemiological surveillance strategies is critical to assisting the Ministry of Health in the future success of the establishment of an eHealth surveillance monitoring program

    Moving Toward a Strategy for Addressing Climate Displacement of Marine Resources: A Proof-of-Concept

    Realistic predictions of climate change effects on natural resources are central to adaptation policies that try to reduce these impacts. However, most current forecasting approaches do not incorporate species-specific, process-based biological information, which limits their ability to inform actionable strategies. Mechanistic approaches, incorporating quantitative information on functional traits, can potentially predict species- and population-specific responses that result from the cumulative impacts of small-scale processes acting at the organismal level, and can be used to infer population-level dynamics and inform natural resources management. Here we present a proof-of-concept study using the European anchovy as a model species that shows how a trait-based, mechanistic species distribution model can be used to explore the vulnerability of marine species to environmental changes, producing quantitative outputs useful for informing fisheries management. We crossed scenarios of temperature and food to generate quantitative maps of selected mechanistic model outcomes (e.g., Maximum Length and Total Reproductive Output). These results highlight changing patterns of source and sink spawning areas as well as the incidence of reproductive failure. This study demonstrates that model predictions based on functional traits can reduce the degree of uncertainty when forecasting future trends of fish stocks. However, to be effective they must be based on high spatial- and temporal resolution environmental data. Such a sensitive and spatially explicit predictive approach may be used to inform more effective adaptive management strategies of resources in novel climatic conditions

    New Italian data on the distribution of the Annex I Habitats 1510*, 2130*, 2250*, 3180*, 3260, 5230*, 6410, 7140, 7220*, 9320 are reported in this contribution. Specifically, 14 new occurrences in Natura 2000 sites are presented and 20 new cells are added in the EEA 10 km × 10 km reference grid. The new data refer to the Italian administrative regions of Abruzzo, Apulia, Friuli Venezia Giulia, Liguria, Marche, Molise, Sardinia, Sicily, Tuscany and Umbria

    Gravitational Waves from Gravitational Collapse

    Gravitational wave emission from the gravitational collapse of massive stars has been studied for more than three decades. Current state of the art numerical investigations of collapse include those that use progenitors with realistic angular momentum profiles, properly treat microphysics issues, account for general relativity, and examine non--axisymmetric effects in three dimensions.     Essential versus accessory aspects of cell death: recommendations of the NCCD 2015

    Cells exposed to extreme physicochemical or mechanical stimuli die in an uncontrollable manner, as a result of their immediate structural breakdown. Such an unavoidable variant of cellular demise is generally referred to as ‘accidental cell death’ (ACD). In most settings, however, cell death is initiated by a genetically encoded apparatus, correlating with the fact that its course can be altered by pharmacologic or genetic interventions. ‘Regulated cell death’ (RCD) can occur as part of physiologic programs or can be activated once adaptive responses to perturbations of the extracellular or intracellular microenvironment fail. The biochemical phenomena that accompany RCD may be harnessed to classify it into a few subtypes, which often (but not always) exhibit stereotyped morphologic features. Nonetheless, efficiently inhibiting the processes that are commonly thought to cause RCD, such as the activation of executioner caspases in the course of apoptosis, does not exert true cytoprotective effects in the mammalian system, but simply alters the kinetics of cellular demise as it shifts its morphologic and biochemical correlates. Conversely, bona fide cytoprotection can be achieved by inhibiting the transduction of lethal signals in the early phases of the process, when adaptive responses are still operational. Thus, the mechanisms that truly execute RCD may be less understood, less inhibitable and perhaps more homogeneous than previously thought. Here, the Nomenclature Committee on Cell Death formulates a set of recommendations to help scientists and researchers to discriminate between essential and accessory aspects of cell death

    Overexpressed vs mutated Kras in murine fibroblasts: a molecular phenotyping study

    Ras acts in signalling pathways regulating the activity of multiple cellular functions including cell proliferation, differentiation, and apoptosis. Amino-acid exchanges at position 12, 13, or 61 of the Kras gene convert the proto-oncogene into an activated oncogene. Until now, a direct comparison of genome-wide expression profiling studies of Kras overexpression and different Kras mutant forms in a single assay system has not been carried out. In our study, we focused on the direct comparison of global gene expression effects caused by mutations in codon 12 or 13 of the Kras gene and Kras overexpression in murine fibroblasts. We determined Kras cellular mRNA, Ras protein and activated Ras protein levels. Further, we compared our data to the proteome analysis of the same transfected cell lines. Both overexpression and mutations of Kras lead to common altered gene expression patterns. Only two genes, Lox and Col1a1, were reversely regulated in the Kras transfectants. They may contribute to the higher aggressiveness of the Kras codon 12 mutation in tumour progression. The functional annotation of differentially expressed genes revealed a high frequency of proteins involved in tumour growth and angiogenesis. These data further support the important role of these genes in tumour-associated angiogenesis

    Differential Ligand Binding to a Human Cytomegalovirus Chemokine Receptor Determines Cell Type–Specific Motility

    While most chemokine receptors fail to cross the chemokine class boundary with respect to the ligands that they bind, the human cytomegalovirus (HCMV)-encoded chemokine receptor US28 binds multiple CC-chemokines and the CX3C-chemokine Fractalkine. US28 binding to CC-chemokines is both necessary and sufficient to induce vascular smooth muscle cell (SMC) migration in response to HCMV infection. However, the function of Fractalkine binding to US28 is unknown. In this report, we demonstrate that Fractalkine binding to US28 not only induces migration of macrophages but also acts to inhibit RANTES-mediated SMC migration. Similarly, RANTES inhibits Fractalkine-mediated US28 migration in macrophages. While US28 binding of both RANTES and Fractalkine activate FAK and ERK-1/2, RANTES signals through Gα12 and Fractalkine through Gαq. These findings represent the first example of differential chemotactic signaling via a multiple chemokine family binding receptor that results in migration of two different cell types. Additionally, the demonstration that US28-mediated chemotaxis is both ligand-specific and cell type–specific has important implications in the role of US28 in HCMV pathogenesis
