    Effect of Saprotrophic Soil Fungi on Toxocara canis Eggs

    The purpose of this work was to assess the ovicidal activity of Chrysosporium merdarium, Trichoderma harzianum, Fusarium oxysporum, F. moniliforme and F. sulphureum isolated from public areas in the city of La Plata, Argentina, on Toxocara canis eggs in vitro. Each species were cultured on water agar 2% with a suspension of immature-stage T. canis eggs. At 4, 7, 14, 21 and 28 days post-culture, they were observed by light and scanning electron microscopy. One hundred eggs were evaluated and scored according to Lỳsek’s ovicidal effect classification. These procedures were repeated three times which each fungal species. Chrysosporium merdarium and F. oxysporum showed very high ovicidal activity, F. sulphureum high ovicidal activity, F. moniliforme intermediate ovicidal activity and T. harzianum did not affect the viability of T. canis eggs. Taking into account the effects on human and animal health and the environment, the species with better prospects for studying its potential use as biological control was F. sulphureum

    Human Immunodeficiency Virus type 1 in seronegative infants born to HIV-1-infected mothers

    BACKGROUND: Some individuals repeatedly exposed to Human Immunodeficiency Virus do not seroconvert and are resistant to HIV infection. Here, in a pediatric cohort of HIV seronegative infants born of HIV-infected mothers, we have studied eight non-breastfed children in whom viral DNA was detected in their PBMC. Our objective was to assess whether silent infection in these children can be explained by the presence of integrated viral DNA. METHODS: The presence of viral DNA was corroborated by nested PCR with primers for gag and the nef/LTR regions of HIV-1. Integration of HIV DNA into the host genome was assessed by an Alu-LTR PCR. Amplicons were sequenced and phylogenetic analyzes were done. RESULTS: HIV-1 DNA was detected in the earliest available PBMC sample from all eight infants, and two of them tested positive for HIV DNA at 2 years of age. Nested PCR resulted in the amplification of gag, nef/LTR and Alu-LTR fragments, which demostrated that HIV-1 DNA was integrated in the host cell genome. Each individual has a characteristic sequence pattern and is different from the LTR sequence of HXB2 prototype virus and other Mexican isolates. CONCLUSION: HIV-1 DNA was observed in PBMC from HIV exposed seronegative children in this pediatric cohort

    Parasitosis intestinales y factores ambientales en una población rural de Argentina, 2002-2003

    We evaluate the prevalence of intestinal parasites in 504 people and the degree of association between environmental variables and parasites found in population, soil and water in a rural area of Argentina during 2002-2003. A structured survey was used to evaluate the environmental variables and fecal-human, soil and water samples were analyzed. The prevalence of parasites was 45.4%. Most prevalent protozoa were Blastocystis hominis (27.2%) and Giardia lamblia (6.9%), while the most prevalent helminth was Ascaris lumbricoides (3.8%). The analyzed environmental variables showing association (p < 0.05) with presence of parasites in population were: cardboard-tin or wooden house, dirt floor, home or communal water pump, faucet outside the house or public faucet and cesspool or latrine. Parasite forms were found in 82.3% of the soil samples and in 84.2% of the water samples. In both samples we found parasites that were also found in people. In this study we have found deficient sanitary conditions associated with presence of parasites in population and we have evidenced that contaminated soil and water were the source of these parasites.Hemos evaluado la prevalencia de parásitos intestinales en 504 personas y el grado de asociación entre las variables ambientales y los parásitos intestinales hallados en la población, el suelo y el agua de una zona rural de Argentina durante 2002-2003. Se utilizó una encuesta estructurada para relevar las variables ambientales y se examinaron muestras fecales humanas, de agua y de suelo. La prevalencia parasitaria fue de 45,4%. Los protozoos prevalentes fueron Blastocystis hominis (27,2%) y Giardia lamblia (6,9%), mientras que el helminto mas prevalente fue Ascaris lumbricoides (3,8%). Las variables ambientales analizadas que mostraron asociación (p < 0,05) con la presencia de parásitos en la población fueron: casa de cartón-chapa o de madera con piso de tierra, bomba de agua domiciliaria o comunitaria, canilla fuera de la casa o pública y pozo ciego o letrina. Elementos parasitarios fueron hallados en el 82,3% de las muestras de suelo y en el 84,2% de las muestras de agua. En ambas muestras fueron hallados parásitos que también se encontraron en las personas. En este trabajo hemos hallado deficientes condiciones sanitarias asociadas con la presencia de parásitos en las personas y hemos evidenciado que el suelo y el agua contaminada fueron la fuente de esos parasitos

    Related factors to human toxocariasis in a rural community of Argentina

    The objective of this study was to evaluate the relationship between toxocariasis frequency and demographic, environmental, sanitary variables, eosinophylia, and other intestinal parasites in a rural population of Argentina. Serological examination of 100 individuals was carried out by using ELISA technique for the detection of antitoxocara antibodies. Eosinophiles in peripheral blood, presence of intestinal parasites, and demographic, environmental, and socio-cultural data were evaluated. Eighty-one feces samples of dogs belonging to the studied people were analyzed to detect eggs of Toxocara canis. Thirty of them were from 30 dogs and 51 were pools from dog feces. Samples of dirt from around the homes (n: 47) and from public park (n: 4) were taken. To determine the associations, the X2 and Fisher tests were used. The seroprevalence was 23%. Eosinophilia in peripheral blood was detected in 86.95% seropositive individuals and in 37.66% seronegative individuals (p < 0.001, OR = 11.03). Of the 23 people with positive serology, 69.56% had at least one intestinal parasite. All individuals with positive serology had dogs in their homes. Among the dog owners there was a significant association between the presence of anti-toxocara antibodies and home flooding. Eggs of T. canis were detected in the feces of 5/81 dogs and three of these dogs belonged to individuals with positive serology. Eggs of Toxocara spp. were found in 41.17% of the dirt samples, eight of which came from the area surrounding the homes of individuals with positive serology (p = 0.032; OR = 4.36). Taking into account all the variables influencing the frequency of toxocariasis in this population, the implementation of Public Health programs specifically focused on anti-parasitic treatment of dogs is recommended

    Chronic obstructive pulmonary disease subtypes. transitions over time

    Background Although subtypes of chronic obstructive pulmonary disease are recognized, it is unknown what happens to these subtypes over time. Our objectives were to assess the stability of cluster-based subtypes in patients with stable disease and explore changes in clusters over 1 year. Methods Multiple correspondence and cluster analysis were used to evaluate data collected from 543 stable patients included consecutively from 5 respiratory outpatient clinics. Results Four subtypes were identified. Three of them, A, B, and C, had marked respiratory profiles with a continuum in severity of several variables, while the fourth, subtype D, had a more systemic profile with intermediate respiratory disease severity. Subtype A was associated with less dyspnea, better health-related quality of life and lower Charlson comorbidity scores, and subtype C with the most severe dyspnea, and poorer pulmonary function and quality of life, while subtype B was between subtypes A and C. Subtype D had higher rates of hospitalization the previous year, and comorbidities. After 1 year, all clusters remained stable. Generally, patients continued in the same subtype but 28% migrated to another cluster. Together with movement across clusters, patients showed changes in certain characteristics (especially exercise capacity, some variables of pulmonary function and physical activity) and changes in outcomes (quality of life, hospitalization and mortality) depending on the new cluster they belonged to Conclusions Chronic obstructive pulmonary disease clusters remained stable over 1 year. Most patients stayed in their initial subtype cluster, but some moved to another subtype and accordingly had different outcomes

    High prevalences of infection with Giardia intestinalis genotype B among children in urban and rural areas of Argentina

    The protozoan parasite most frequently associated with diarrhoea worldwide is Giardia intestinalis. In 2005, a study was initiated to identify the genotypes of this parasite infecting children in the Argentinian provinces of Buenos Aires, Mendoza and Chaco, and to explore the associations between the genotype detected in a child, the characteristics of the child’s household and the child’s clinical presentation. Overall, 998 children (504 boys and 494 girls) aged between 2–14 years, with or without symptoms, were enrolled. The G. intestinalis in 94 of the 117 stool samples found positive for the parasite by microscopy were successfully genotyped by PCR. Seventy-seven of the children were found to be infected with genotype B only and 14 with genotype AII only, three children being found to have mixed (AII and B) infections. Only genotype B was detected in children from rural areas (P&lt;0·05) and most Giardia detected in children from households with a piped water supply were also of this genotype (P&lt;0·05). The other household characteristics investigated (quality of building, history of flooding, type of sanitation, level of overcrowding, and presence/absence of pet dogs) had no significant effect on the genotype distribution. Children infected with genotype AII were significantly younger than those infected with genotype B (P&lt;0·05) and there was a significant positive association between infection with genotype B and abdominal pain (P&lt;0·05). Diarrhoea was not, however, found to be significantly associated with genotype-AII or genotype-B infection. This is the first published report on the Giardia genotypes circulating in the provinces of Mendoza and Chaco. The results indicate the importance of asymptomatic children in the transmission of Giardia among the young.Facultad de Ciencias Médica

    Atlas Floresta Americana. Bonpland. 1850: La identificación de las plantas de la Materia Médica Misionera de Pedro de Montenegro (SJ)

    The manuscript bequeathed by Amado Bonpland, Atlas Floresta Americana 1850, obtained from the Library of the Casa de la Cultura Benjamín Carrión (Quito, Ecuador) after efforts made by various entities, allows us to have material of great historical-scientific value and that contributes to the study of the plants identification described in the Materia Medica Missionera of Pedro de Montenegro. In this sense, it constitutes a documentary collection that integrates valuable data pertaining to the Jesuit period, as well as the period in which Bonpland remained in the River Plate basin. The manuscript was analyzed with an interdisciplinary approach. The objectives of the present study are to present the unpublished document, characterize it historically, transcribe it, translate it into Spanish, update the phytonyms to the current Guaraní, update the identifications of the plants of the Jesuit work Pedro de Montenegro, and compare these identifications with others made on the same work. The characteristics of the manuscript and the historical context of its production are described. The botanical affiliation of the plants identified by Bonpland from the Materia Medica Misionera is presented. The importance of this unpublished document as heritage is highlighted, as well as its possible contributions to Jesuit, botanical and pharmacological studies in general.El manuscrito legado por Amado Bonpland, Atlas Floresta Americana 1850, obtenido de la Biblioteca de la Casa de la Cultura Benjamín Carrión (Quito, Ecuador) luego de gestiones realizadas por diversas entidades, nos permite contar con un material de gran valor históricocientífico y que aporta al estudio de la identificación de las plantas descriptas en la Materia Médica Misionera de Pedro de Montenegro. En tal sentido constituye un fondo documental que integra datos valiosos pertenecientes al período jesuítico, como también del período en que Bonpland permaneció en la cuenca rioplatense. El manuscrito fue analizado desde un enfoque interdisciplinario. Los objetivos del presente trabajo son: presentar el documento inédito, caracterizarlo históricamente, transcribirlo, traducirlo al español, actualizar los fitónimos al guaraní actual, actualizar las identificaciones de las plantas de la obra del jesuita Pedro de Montenegro, y comparar dichas identificaciones con otras realizadas sobre la misma obra. Se describen las características del manuscrito y el contexto histórico de su producción. Se presenta la afiliación botánica de las plantas identificadas por Bonpland a partir de la Materia Médica Misionera. Se resalta la importancia de este documento inédito como patrimonio, como también sus posibles aportes a los estudios jesuíticos, botánicos y farmacológicos en general. INFORMACION COMPLEMENTARIA 

    Antibacterial properties of tualang honey and its effect in burn wound management: a comparative study

    <p>Abstract</p> <p>Background</p> <p>The use of honey as a natural product of <it>Apis </it>spp. for burn treatment has been widely applied for centuries. Tualang honey has been reported to have antibacterial properties against various microorganisms, including those from burn-related diagnoses, and is cheaper and easier to be absorbed by Aquacel dressing. The aim of this study is to evaluate the potential antibacterial properties of tualang honey dressing and to determine its effectiveness as a partial thickness burn wound dressing.</p> <p>Methods</p> <p>In order to quantitate the bioburden of the swabs, pour plates were performed to obtain the colony count (CFU/ml). Swabs obtained from burn wounds were streaked on blood agar and MacConkey agar for bacterial isolation and identification. Later, antibacterial activity of Aquacel-tualang honey, Aquacel-Manuka honey, Aquacel-Ag and Aquacel- plain dressings against bacteria isolated from patients were tested (<it>in-vitro</it>) to see the effectiveness of those dressings by zone of inhibition assays.</p> <p>Results</p> <p>Seven organisms were isolated. Four types of Gram-negative bacteria, namely <it>Enterobacter cloacae</it>, <it>Klebsiella pneumoniae</it>, <it>Pseudomonas </it>spp. and <it>Acinetobacter </it>spp., and three Gram-positive bacteria, namely <it>Staphylococcus aureus</it>, coagulase-negative <it>Staphylococcus aureus </it>(CONS) and <it>Streptococcus </it>spp., were isolated. Total bacterial count decreased on day 6 and onwards. In the <it>in-vitro </it>antibacterial study, Aquacel-Ag and Aquacel-Manuka honey dressings gave better zone of inhibition for Gram positive bacteria compared to Aquacel-Tualang honey dressing. However, comparable results were obtained against Gram negative bacteria tested with Aquacel-Manuka honey and Aquacel-Tualang honey dressing.</p> <p>Conclusions</p> <p>Tualang honey has a bactericidal as well as bacteriostatic effect. It is useful as a dressing, as it is easier to apply and is less sticky compared to Manuka honey. However, for Gram positive bacteria, tualang honey is not as effective as usual care products such as silver-based dressing or medical grade honey dressing.</p
