    APES: Approximate Posterior Ensemble Sampler

    This paper proposes a novel approach to generate samples from target distributions that are difficult to sample from using Markov Chain Monte Carlo (MCMC) methods. Traditional MCMC algorithms often face slow convergence due to the difficulty in finding proposals that suit the problem at hand. To address this issue, the paper introduces the Approximate Posterior Ensemble Sampler (APES) algorithm, which employs kernel density estimation and radial basis interpolation to create an adaptive proposal, leading to fast convergence of the chains. The APES algorithm's scalability to higher dimensions makes it a practical solution for complex problems. The proposed method generates an approximate posterior probability that closely approximates the desired distribution and is easy to sample from, resulting in smaller autocorrelation times and a higher probability of acceptance by the chain. In this work, we compare the performance of the APES algorithm with the affine invariance ensemble sampler with the stretch move in various contexts, demonstrating the efficiency of the proposed method. For instance, on the Rosenbrock function, the APES presented an autocorrelation time 140 times smaller than the affine invariance ensemble sampler. The comparison showcases the effectiveness of the APES algorithm in generating samples from challenging distributions. This paper presents a practical solution to generating samples from complex distributions while addressing the challenge of finding suitable proposals. With new cosmological surveys set to deal with many new systematics, which will require many new nuisance parameters in the models, this method offers a practical solution for the upcoming era of cosmological analyses.Comment: 15 pages, 6 figures, 7 table

    Phosphodiesterase Type 5 Inhibitors and Visual Side Effects: A Narrative Review

    Phosphodiesterase type 5 inhibitors such as sildenafil citrate and tadalafil are well known for the treatment of erectile dysfunction. However, their use in the presence of pulmonary hypertension can cause ophthalmologic side effects, including non-arteritic optic ischemic neuropathy, chorioretinopathy, glaucoma, and optic atrophy. The present review aimed to identify these visual side effects and provide recommendations. We identified articles published from January 2000 to March 2019 on diseases arising from the management of sexual dysfunction in urology or pulmonary hypertension in pneumonia that could cause pathologic alterations in eye structure based on a literature search of the MEDLINE electronic database using keywords for the most common adverse effects and different kinds of phosphodiesterase 5 inhibitors. After applying the exclusion criteria, we selected 36 of the 77 articles initially identified to write the narrative review and added 20 additional articles to completely describe the pathological entities. Phosphodiesterase type 5 inhibitors can cause side effects in the eye including ocular surface abnormalities, increased intraocular pressure and glaucoma, uveitis, non-arteritic ischemic neuropathy, chorioretinopathy, retinal occlusion, and visual field changes. There is an increased need for well-performed studies to better understand these side effects, which are common due to the wide use of sildenafil

    Estudio comparado del Budismo Mahayana, el Budismo Vajrayana y el Yungdrung Bon

    El objetivo del presente proyecto es generar una contribución al conocimiento comparativo de las diferentes ramas del budismo, tomando como eje tres escuelas que se consideran exponentes importantes dentro de esa religión/filosofía. El budismo Mahayana, imperante en China, Corea y Japón, el budismo Vajrayana o Tántrico, imperante en el Tíbet, Nepal, Mongolia y en menor medida en Japón, y el Yungdrun Bön, la tradición originaria del Tíbet, basada en las enseñanzas de su fundador, Sherab Miwoche, considerado como un Buda, pero que es anterior a la llegada del budismo desde la India. Para este estudio nos centramos en el área de la disciplina moral (skt. Sila), área desarrollada por las tres ramas del budismo tratadas. Realizaremos una comparación de tipo conceptual-cualitativo que permita una clara definición de conceptos básicos. Es nuestra intención generar un mapa conceptual que sirva de marco taxonómico para estudios futuros y de esa forma contribuir a los estudios comparativos dentro del campo de las investigaciones budistas.Luego de la revisión preliminar de los materiales disponibles, se hizo hincapié en el estudio en paralelo de dos corrientes básicas: la definida por el Yüngdrung Bön y la Budista. Este esquema, desarrollado por los dos investigadores asistentes, ha permitido establecer un mapa conceptual que define los diferentes componentes de la conducta ética en tres niveles: el nivel de Pratimoksa o liberación individual, el nivel de la práctica del Bodhisattva, concentrado en las perfecciones, y el nivel del adepto tántrico, para aquellos que estuvieran dispuestos a seguir ese camino. Una vez establecidos esos niveles, se pudo encontrar que existía una recurrencia de conceptos dentro de las categorías de sila referentes a la perspectiva, a la práctica y a la conducta en las diferentes escuelas estudiadas. Se ha avanzado en la sistematización del marco teórico para construir un corpus documental que permita realizar el análisis de conjunto y elaborar el informe final.The present project has the objective of contributing to the comparative knowledge of the different schools of Buddhism, focusing in three schools considered important in this religious philosophy: Mahayana Buddhism, the main school in China, Korea and Japan; Vajrayana Tantric Buddhism, main style of Tíbet, Nepal and Mongolia (and in some Japanese schools) and finally Yundrung Bön, the Tibetan original tradition of spirituality, based on the teaching of its founder, Sherab Miwoche, considered a Buddha before the time of Buddha Sakyamuni by his followers. In this study we will focus on the area of moral discipline (skt.Sila), which is developed by the three schoolsof Buddhism mentioned. We will realize an exploration of the conceptual-qualitative type whichshould allow a clear definition of basic concepts. It is our intention to generate a conceptual mapwhich can be used as a taxonomical framework in future studies and in that way, contribute to theresearch of Buddhism.After a preliminary revision of the available resources, we decided to focus on two branches:the Vajrayana Buddhist one and the Yungdrung Bön. This scheme, developed by the assistant researchers,has allowed to study the conduct in three levels: the Pratimoksha level of individualliberation, the level of the Bodhisattva practice of perfections and the level of the tantric adept, forthose who are willing to follow that road. Once those levels were established, we found a recurrenceof concepts in the categories of Sila as pertain the perspective, the practice and the conductin the different schools that we have studied. We have made progress in the systematization of thetheoretical framework to build a documental corpus which allows us to do a comparative analysisand final conclusion

    The CBRB regulon: Promoter dissection reveals novel insights into the CbrAB expression network in Pseudomonas putida

    CbrAB is a high ranked global regulatory system exclusive of the Pseudomonads that responds to carbon limiting conditions. It has become necessary to define the particular regulon of CbrB and discriminate it from the downstream cascades through other regulatory components. We have performed in vivo binding analysis of CbrB in P. putida and determined that it directly controls the expression of at least 61 genes; 20% involved in regulatory functions, including the previously identified CrcZ and CrcY small regulatory RNAs. The remaining are porines or transporters (20%), metabolic enzymes (16%), activities related to protein translation (5%) and orfs of uncharacterised function (38%). Amongst the later, we have selected the operon PP2810-13 to make an exhaustive analysis of the CbrB binding sequences, together with those of crcZ and crcY. We describe the implication of three independent non-palindromic subsites with a variable spacing in three different targets; CrcZ, CrcY and operon PP2810-13 in the CbrAB activation. CbrB is a quite peculiar σN—depen-dent activator since it is barely dependent on phosphorylation for transcriptional activation. With the depiction of the precise contacts of CbrB with the DNA, the analysis of the multi-merisation status and its dependence on other factors such as RpoN o IHF, we propose a model of transcriptional activation.Ministerio de Economía y Competitividad BIO2014-57545-
