Objective: We aimed to quantify SARS-CoV-2 specific antibodies’ seroprevalence among university students in Porto. Methods: A rapid point of care testing for SARS-CoV-2 specific immunoglobulin (Ig) M and IgG antibodies was performed, and a questionnaire was applied to 6512 voluntary students from September to December 2020. We computed the apparent IgM, IgG, and IgM or IgG prevalence, and the true prevalence and 95% credible intervals (95% CI) using Bayesian inference. Results: We found an apparent prevalence (IgM or IgG) of 9.7%, the true prevalence being 7.9% (95% CI 4.9–11.1). Prevalence was significantly higher among males (10.9% vs. 9.2%), international students (18.1% vs. 10.4% local vs. 8.8% nationally displaced), and increased with age. Those with a known risk contact, that experienced quarantine, had symptoms, or a previous negative molecular test had a higher seroprevalence. Of the 91 (1.4%) students who reported a molecular diagnosis, 86.8% were reactive for IgM or IgG. Conclusion: Based on immunological evidence infection was 5.6-fold the reported molecular diagnosis. The higher seroprevalence among male, older, and international students emphasizes the importance of identifying particular groups. Castanea sativa mill. flowers as potential chemopreventive agent against rat prostate cancer model

Introduction: Prostate cancer is one of the most common cancer among men, having a huge impact in their health [1]. This work aimed to evaluate the influence of a decoction extract obtained from C. sativa flowers (CF) on chemically and hormonally induced rat prostate cancer animal model. Material & Methods: All the animal experiments were approved by the Institutional Animals Ethics Committee and by Portuguese national authorities (DGAV no 021326). Forty male Wistar Unilever rats were randomly divided into four groups: control group (n=10), induced group (n=15), CF control group (n=5) and CF induced group (n=10). Animals from induced groups received a multistep induction protocol, which consisted of sequential administration of flutamide, testosterone propionate, the carcinogenic agent MNU and crystalline testosterone. The CF extract, rich in ellagitannins especially trigalloy-HHDP- glucose, was administered in the drinking water (3 mg/animal/day) for 49 weeks. Animals were sacrificed at 61 weeks of age and organs were collected, weighed and processed for light microscopy. Data were analysed using SPSS and GraphPad Prism software. Results: There were no significant differences in relative mean liver weight among groups exposed and not exposed to the CF extract and no animals developed severe hepatic changes. Animals from CF induced group developed less prostatic intraepithelial neoplasia than induced group. Also, animals exposed to the CF extract did not present areas of inflammation of the dorsolateral prostate lobe greater than 50% unlike the groups not exposed (p<0.05). The administration of CF in induced animals was able to decrease the activity of CAT and GST by 36% and 20%, respectively (p<0.05). Conclusions: These results suggest that CF extract was well tolerate by the animals and did not cause severe hepatic and renal toxicity. C. sativa flowers extract may be used as chemopreventive agent against prostate cancer and seems to have an antioxidant role

Direct enzymatic esterification of cotton and Avicel with wild-type and engineered cutinases

In this work, the surface of cellulose, either Avicel or cotton fabric, was modified using cutinases without any previous treatment to swell or to solubilise the polymer. Aiming further improvement of cutinase ester synthase activity on cellulose, an engineered cutinase was investigated. Wild-type cutinase from Fusarium solani and its fusion with the carbohydrate-binding module N1 from Cellulomonas fimi were able to esterify the hydroxyl groups of cellulose with distinct efficiencies depending on the acid substrate/solvent system used, as shown by titration and by ATR-FTIR. The carbonyl stretching peak area increased significantly after enzymatic treatment during 72 h at 30 °C. Cutinase treatment resulted in relative increases of 31 and 9 % when octanoic acid and vegetable oil were used as substrates, respectively. Cutinase-N1 treatment resulted in relative increases of 11 and 29 % in the peak area when octanoic acid and vegetable oil were used as substrates, respectively. The production and application of cutinase fused with the domain N1 as a cellulose ester synthase, here reported for the first time, is therefore an interesting strategy to pursuit. This work was co-funded by the European Social Fund through the management authority POPH and FCT, Postdoctoral fellowship reference: SFRH/BPD/47555/2008. Chagas Cardiomiopathy: The Potential of Diastolic Dysfunction and Brain Natriuretic Peptide in the Early Identification of Cardiac Damage

Chagas disease remains a major cause of morbidity and mortality in several countries of Latin America and has become a potential public health problem in countries where the disease is not endemic as a result of migration flows. Cardiac involvement represents the main cause of mortality, but its diagnosis is still based on nonspecific criteria with poor sensitivity. Early identification of patients with cardiac damage is desirable, since early treatment may improve prognosis. Diastolic dysfunction and elevated brain natriuretic peptide levels are present in different cardiomyopathies and in advanced phases of Chagas disease. However, there are scarce data about the role of these parameters in earlier forms of the disease. We conducted a study to assess the diastolic function, regional systolic abnormalities and brain natriuretic peptide levels in the different forms of Chagas disease. The main finding of our investigation is that diastolic dysfunction occurs before any cardiac dilatation or motion abnormality. In addition, BNP levels identify patients with diastolic dysfunction and Chagas disease with high specificity. The results reported in this study could help to early diagnose myocardial involvement and better stratify patients with Chagas disease
