Effects of GLP-1 receptor agonists on neurological complications of diabetes
Emerging evidence suggests that treatment with glucagon-like peptide-1 receptor agonists (GLP-1 RAs) could be an interesting treatment strategy to reduce neurological complications such as stroke, cognitive impairment, and peripheral neuropathy. We performed a systematic review to examine the evidence concerning the effects of GLP-1 RAs on neurological complications of diabetes. The databases used were Pubmed, Scopus and Cochrane. We selected clinical trials which analysed the effect of GLP-1 RAs on stroke, cognitive impairment, and peripheral neuropathy. We found a total of 19 studies: 8 studies include stroke or major cardiovascular events, 7 involve cognitive impairment and 4 include peripheral neuropathy. Semaglutide subcutaneous and dulaglutide reduced stroke cases. Liraglutide, albiglutide, oral semaglutide and efpeglenatide, were not shown to reduce the number of strokes but did reduce major cardiovascular events. Exenatide, dulaglutide and liraglutide improved general cognition but no significant effect on diabetic peripheral neuropathy has been reported with GLP-1 RAs. GLP-1 RAs are promising drugs that seem to be useful in the reduction of some neurological complications of diabetes. However, more studies are neededThis work was supported by the Instituto de Salud Carlos III (PI19/01375) co-financed with FEDER programme fund.

Unveiling the Synergistic Interplay of Neuropeptides for Novel Therapeutic Approaches in Neurodegenerative and Depressive Disorders.
The intricate relationship between hippocampal neurogenesis dysregulation and neurodegenerative diseases such as Alzheimer's, as well as depression, has sparked an urgent call for innovative therapeutic strategies. Our groundbreaking study delves into the interaction of Neuropeptide Y (NPY) and galanin (GAL) agonists, two neuromodulatory systems with a substantial presence in the limbic system, and their potential neurogenic impact on both the dorsal and ventral hippocampus.
Through meticulous examination of the subchronic e"ects of NPY Y1 (Y1R) and GAL2 (GALR2) agonists on hippocampal cell proliferation, survival, and neuroprotective factor expression, we reveal a fascinating cascade of cellular responses. These include increased cell proliferation (PCNA), enhanced hippocampal cell survival (BrdU), and induction of neuroprotective factors (BDNF).
Our functional assessment showcases the resulting improvements in spatial memory performance in the object-in-place task and antidepressant-like e"ects in the forced swimming test. These outcomes are attributed to the synergistic interaction between Y1R and GALR2 receptors, which promote neuronal survival and neurite outgrowth in hippocampal cells.
This pioneering research paves the way for the development of heterobivalent agonist pharmacophores that target Y1R-GALR2 heterocomplexes. By acting on neuronal precursor cells in the dentate gyrus of the dorsal hippocampus, these novel compounds hold immense promise as transformative therapies for cognitive and a"ective impairments in neurodegenerative and depressive diseases.Supported by the Proy_Excel_2021_0613 (Junta de Andalucía) and Proyecto Puente (B4-2021) (UMA), Spain to MN.
Dry spells in the central basin of Duero river
rachas a 40’1 días.
The aim of this work is to characterize the climate of the most arid area in the Duero
basin. It has been paid attention to periods without precipitation (dry spells) and the estimation of probability and frequency of these periods using statistical methods of easy application.
Average annual precipitation is below 400 mm and the annual number of dry days is
high. Using a rainfall threshold of 0’1 mm in the definition of a dry day, the mean of dry days
is 291 and the mean length of dry spells is 9’6 days. If the threshold is increased to 10 mm, the
annual number of dry days is 356 and the mean length of dry spells is 40’1. Although the
study area is located in the northern part of Iberian Peninsula, the analysis of the previous data
and the length, intensity and probability of the maximum annual dry spells, confirm that its
climatic characteristics are more similar to data from Mediterranean observatories than those
from Atlantic area. Finally, as much the first-order Markov chain as the Gumbel distribution
of extremes are suitable to estimate the probability and frequency of dry spells respectively.

GALR2 and Y1R agonists intranasal infusion enhanced adultventral hippocampal neurogenesis and antidepressant‐likeeffects involving BDNF actions
Dysregulation of adult hippocampal neurogenesis is linked to major depressive disorder(MDD), with more than 300 million people diagnosed and worsened by the COVID‐19pandemic. Accumulating evidence for neuropeptideY (NPY) and galanin (GAL) interactionwas shown in various limbic system regions at molecular‐, cellular‐, and behavioral‐specific levels. The purpose of the current work was to evaluate the proliferating role ofGAL2 receptor (GALR2) and Y1R agonists interaction upon intranasal infusion in theventral hippocampus. We studied their hippocampal proliferating actions using theproliferating cell nuclear antigen (PCNA) on neuroblasts or stem cells and the expressionof the brain‐derived neurothrophic factor (BDNF). Moreover, we studied the formation ofY1R–GALR2 heteroreceptor complexes and analyzed morphological changes inhippocampal neuronal cells. Finally, the functional outcome of the NPY and GALinteraction on the ventral hippocampus was evaluated in the forced swimming test. Wedemonstrated that the intranasal infusion of GALR2 and the Y1R agonists promotesneuroblasts proliferation in the dentate gyrus of the ventral hippocampus and the induction of the neurotrophic factor BDNF. These effects were mediated by theincreased formation of Y1R–GALR2 heteroreceptor complexes, which may mediate theneurites outgrowth observed on neuronal hippocampal cells. Importantly, BDNF actionwas found necessary for the antidepressant‐like effects after GALR2 and the Y1Ragonists intranasal administration. Our data may suggest the translational development ofnew heterobivalent agonist pharmacophores acting on Y1R–GALR2 heterocomplexes inthe ventral hippocampus for the novel therapy of MDD or depressive‐affecting diseases.
