    Antioxidant oils and Salmonella enterica Typhimurium reduce tumor in an experimental model of hepatic metastasis

    Fruit seeds high in antioxidants have been shown to have anticancer properties and enhance host protection against microbial infection. Recently we showed that a single oral dose of Salmonella enterica serovar Typhimurium expressing a truncated human interleukin-2 gene (SalpIL2) is avirulent, immunogenic, and reduces hepatic metastases through increased natural killer cell populations in mice. To determine whether antioxidant compounds enhance the antitumor effect seen in SalpIL2-treated animals, we assayed black cumin (BC), black raspberry (BR), and milk thistle (MT) seed oils for the ability to reduce experimental hepatic metastases in mice. In animals without tumor, BC and BR oil diets altered the kinetics of the splenic lymphocyte response to SalpIL2. Consistent with previous reports, BR and BC seed oils demonstrated independent antitumor properties and moderate adjuvant potential with SalpIL2. MT oil, however, inhibited the efficacy of SalpIL2 in our model. Based on these data, we conclude that a diet high in antioxidant oils promoted a more robust immune response to SalpIL2, thus enhancing its antitumor efficacy

    Single Cell Transcriptome Data Analysis Defines the Heterogeneity of Peripheral Nerve Cells in Homeostasis and Regeneration

    The advances in single-cell RNA sequencing technologies and the development of bioinformatics pipelines enable us to more accurately define the heterogeneity of cell types in a selected tissue. In this report, we re-analyzed recently published single-cell RNA sequencing data sets and provide a rationale to redefine the heterogeneity of cells in both intact and injured mouse peripheral nerves. Our analysis showed that, in both intact and injured peripheral nerves, cells could be functionally classified into four categories: Schwann cells, nerve fibroblasts, immune cells, and cells associated with blood vessels. Nerve fibroblasts could be sub-clustered into epineurial, perineurial, and endoneurial fibroblasts. Identified immune cell clusters include macrophages, mast cells, natural killer cells, T and B lymphocytes as well as an unreported cluster of neutrophils. Cells associated with blood vessels include endothelial cells, vascular smooth muscle cells, and pericytes. We show that endothelial cells in the intact mouse sciatic nerve have three sub-types: epineurial, endoneurial, and lymphatic endothelial cells. Analysis of cell type-specific gene changes revealed that Schwann cells and endoneurial fibroblasts are the two most important cell types promoting peripheral nerve regeneration. Analysis of communication between these cells identified potential signals for early blood vessel regeneration, neutrophil recruitment of macrophages, and macrophages activating Schwann cells. Through this analysis, we also report appropriate marker genes for future single cell transcriptome data analysis to identify cell types in intact and injured peripheral nerves. The findings from our analysis could facilitate a better understanding of cell biology of peripheral nerves in homeostasis, regeneration, and disease.</jats:p

    Police ethics and integrity: Can a new code overturn the blue code?

    This paper analyses police officer perspectives on the seriousness of potential misconduct or unethical behaviour, and the factors that might shape whether they would report their colleagues' misdemeanours. It compares responses from police officers in UK three forces, looking at potentially corrupt behaviours described in a series of scenarios. The discussion includes why some types of misdemeanour seem more likely to be reported and the potential effects of a newly introduced formal Code of Ethics. In terms of differences between ranks and roles, and different responses from different services, the study suggests that the way police culture operates is significant and needs to be more widely addressed. The study used scenario based questionnaires to elicit views about the seriousness of certain police behaviours and to ask whether officers would report colleagues' misdemeanours. It develops a previous survey by one of the authors which conducted a similar survey published in 2005. Using the same questionnaire the new study examined a larger and more diverse sample of serving officers (n=520). This new study compares responses from police officers in UK three forces, geographically distributed across the country and have differing characteristics in terms of size, rurality, population density and policing priorities

    Rethinking the law and politics of democratic police accountability

    This paper evaluates the work and impact of a number of Police and Crime Commissioners (PCCs) in England and Wales and attempts to refocus public discourse and scrutiny on their Police and Crime Plans as a key prism through which their performance should be measured. Drawing upon the literature published by various PCCs, the Stevens Commission, the Home Affairs Committee and numerous academics, the paper will argue that a major reform of democratic police accountability in England and Wales is needed.     Stress Hyperglycemia in Critically Ill Patients: Insight Into Possible Molecular Pathways

    Severe sepsis, systemic inflammatory response syndrome (SIRS), and traumatic brain injury are frequently associated with hyperglycemia in non-diabetic patients. In patients suffering from any of these conditions, hyperglycemia at admission to an intensive care unit (ICU) is directly correlated with increased mortality or morbidity. Although there was initial enthusiasm for insulin treatment to blood glucose levels below 110 mg/dL in these patients, recent understanding suggests that the potential for hypoglycemic complications make this approach potentially dangerous. More moderate glucose control seems to be more beneficial than the aggressive glucose lowering initially suggested. An important publication has shown that hyperlactatemia accompanying hyperglycemia could be the real culprit in bad outcomes. This suggests that coupling moderate glucose lowering with therapeutic agents which might treat the underlying metabolic disturbances in these conditions may be a better strategy. The key metabolic disturbance in these three conditions seems to be persistent glycolysis as an energy source even in the presence of adequate tissue oxygenation (the Warburg Effect). We look at recent advances in understanding aerobic glycolysis and possibly the action of DPP4 on incretins resulting in insulin dysregulation and suggest key metabolic pathways involved in hyperglycemia regulation

    Doubly estranged: racism, the body and reflection

Drawing on a large qualitative dataset it will argue that young migrant group boys in Dublin’s north inner city suffer from a break with their embodied selves as they are pushed between habitual and reflective action. The dual elements of “double estrangement” will be outlined, firstly, through the contention that visible difference and dispositions of the body mark minority boys out as not belonging within peer exchanges in three primary schools. Secondly, by arguing this has the effect of heightening a boy’s self-consciousness of their body as an object of value estranging them from their habitual embodied being

    This article is a review of issues associated with measuring ethnicity and using ethnicity measures in social science research. The review is oriented towards researchers who undertake secondary analyses of large-scale multipurpose social science datasets. The article begins with an outline of two main approaches used in social surveys to measure ethnicity, the ‘mutually exclusive category’ approach and the ‘multiple characteristics’ approach. We also describe approaches to the use of ethnicity measures in cross-national comparative research. We emphasise the value of sensitivity analyses. We also encourage researchers to carefully consider the possible relationships between ethnicity and other important variables in order to avoid spurious interpretations of the effects of ethnicity

    On the Functional Significance of the P1 and N1 Effects to Illusory Figures in the Notch Mode of Presentation

    The processing of Kanizsa figures have classically been studied by flashing the full “pacmen” inducers at stimulus onset. A recent study, however, has shown that it is advantageous to present illusory figures in the “notch” mode of presentation, that is by leaving the round inducers on screen at all times and by removing the inward-oriented notches delineating the illusory figure at stimulus onset. Indeed, using the notch mode of presentation, novel P1and N1 effects have been found when comparing visual potentials (VEPs) evoked by an illusory figure and the VEPs to a control figure whose onset corresponds to the removal of outward-oriented notches, which prevents their integration into one delineated form. In Experiment 1, we replicated these findings, the illusory figure was found to evoke a larger P1 and a smaller N1 than its control. In Experiment 2, real grey squares were placed over the notches so that one condition, that with inward-oriented notches, shows a large central grey square and the other condition, that with outward-oriented notches, shows four unconnected smaller grey squares. In response to these “real” figures, no P1 effect was found but a N1 effect comparable to the one obtained with illusory figures was observed. Taken together, these results suggest that the P1 effect observed with illusory figures is likely specific to the processing of the illusory features of the figures. Conversely, the fact that the N1 effect was also obtained with real figures indicates that this effect may be due to more global processes related to depth segmentation or surface/object perception
