Biology of growth of Hoplias aff. malabaricus (Bloch, 1794) in a shallow pampean lake Argentina
The trahira Hoplias aff. malabaricus is a top predator in pampean shallow lakes and is highly appreciated by recreational anglers and artisanal fishermen. Trahira growth from Yalca shallow lake was determined by lepidological analysis and age validated by marginal increment. When growth was fitted to the von Bertalanffy model, annual classes exhibited a bimodal pattern as a result of the presence of spring and summer annual cohorts associated with a three month spawning season, each period in turn showing different growth patterns. The trahira population-age structure at Yalca shallow lake showed a truncated profile with very low numbers of large adults and few individuals older than three to four years, thus producing an unbalanced length-structure population. Growth parameters and growth performance were similar to the corresponding parameters estimated for other shallow pampean lakes of the region, but strongly diverged from the data for those populations inhabiting subtropical and tropical environments. Such differences could be accounted for by dissimilarity in metabolic rates associated with thermal differences accompanying seasonal variability among latitudes as well as by the development of adaptive physiologic and demographic responses to cope with the high thermal amplitude and hydrologic instability observed in pampean lakes. Fil: Balboni, Leandro. Novel 2-amino-isoflavones exhibit aryl hydrocarbon receptor agonist or antagonist activity in a species/cell-specific context
The aryl hydrocarbon receptor (AhR) mediates the induction of a variety of xenobiotic metabolism genes. Activation of the AhR occurs through binding to a group of structurally diverse compounds, most notably dioxins, which are exogenous ligands. Isoflavones are part of a family which include some well characterised endogenous AhR ligands. This paper analysed a novel family of these compounds, based on the structure of 2-amino-isoflavone. Initially two luciferase-based cell models, mouse H1L6.1c2 and human HG2L6.1c3, were used to identify whether the compounds had AhR agonistic and/or antagonistic properties. This analysis showed that some of the compounds were weak agonists in mouse and antagonists in human. Further analysis of two of the compounds, Chr-13 and Chr-19, was conducted using quantitative real-time PCR in rat H4IIE and human MCF-7 cells. The results indicated that Chr-13 was an agonist in rat but an antagonist in human cells. Chr-19 was shown to be an agonist in rat but more interestingly, a partial agonist in human. Luciferase induction results not only revealed that subtle differences in the structure of the compound could produce species-specific differences in response but also dictated the ability of the compound to be an AhR agonist or antagonist. Substituted 2-amino-isoflavones represent a novel group of AhR ligands that must differentially interact with the AhR ligand binding domain to produce their species-specific agonist or antagonist activity and future ligand binding analysis and docking studies with these compounds may provide insights into the differential mechanisms of action of structurally similar compounds
Prediction of Metabolic Profiles from Transcriptomics Data in Human Cancer Cell Lines
The Metabolome and Transcriptome are mutually communicating within cancer cells, and this interplay is translated into the existence of quantifiable correlation structures between gene expression and metabolite abundance levels. Studying these correlations could provide a novel venue of understanding cancer and the discovery of novel biomarkers and pharmacological strategies, as well as laying the foundation for the prediction of metabolite quantities by leveraging information from the more widespread transcriptomics data. In the current paper, we investigate the correlation between gene expression and metabolite levels in the Cancer Cell Line Encyclopedia dataset, building a direct correlation network between the two molecular ensembles. We show that a metabolite/transcript correlation network can be used to predict metabolite levels in different samples and datasets, such as the NCI-60 cancer cell line dataset, both on a sample-by-sample basis and in differential contrasts. We also show that metabolite levels can be predicted in principle on any sample and dataset for which transcriptomics data are available, such as the Cancer Genome Atlas (TCGA)
Clinicopathological findings in cats tested for feline immunodeficiency virus (FIV) and feline leukaemia virus (FeLV)
This retrospective study aimed to evaluate the clinicopathological changes in a population of cats tested for feline immunodeficiency virus (FIV) and feline leukaemia virus (FeLV), in an Italian Veterinary University Hospital, in the period between January 2002 and May 2016. During the period of 14 years, 1834 cats were tested, and of these 241/1834 (13.1%) were positive for FIV antibodies and 92/1834 (5%) cats were positive for FeLV antigen. These data confirm the presence of a high prevalence of these viruses on Italian territory. To the authors’ knowledge, this study describes findings that have never been evaluated before, such as iron status in retrovirus-infected cats and urinalysis in FeLV-positive cats. In this study, FIV-positive cats were more likely to have higher serum protein concentration and lower albumin-globulin ratio than other groups of cats. Lower urine specific gravity and higher urine protein to creatinine ratio were also detected for FIV-positive cats when compared with negative and healthy cats. FeLV-positive cats were more likely to have cytopenia, decreased haemoglobin, haematocrit and RBC compared with other groups of cats. The data obtained underline the importance of considering retroviral infections in the presence of a broad spectrum of risk factors and laboratory anomalies
Novel sequence variants of viral hexon and fibre genes in two dogs with canine adenovirus type 1-associated disease
There is little information on sequence variation of canine adenovirus type 1 (CAdV-1), the aetiological agent of infectious canine hepatitis (ICH). This study reports hexon and fibre gene sequence variants of CAdV-1 in a dog with systemic ICH and a dog with the ocular form of the disease (\ue2\u80\u98blue eye\ue2\u80\u99) in Northern Italy in 2013. One of the sequence variants matched a CAdV-1 fox sequence previously detected in Italy
Making GDPR Usable: A Model to Support Usability Evaluations of Privacy
We introduce a new model for evaluating privacy that builds on the criteria
proposed by the EuroPriSe certification scheme by adding usability criteria.
Our model is visually represented through a cube, called Usable Privacy Cube
(or UP Cube), where each of its three axes of variability captures,
respectively: rights of the data subjects, privacy principles, and usable
privacy criteria. We slightly reorganize the criteria of EuroPriSe to fit with
the UP Cube model, i.e., we show how EuroPriSe can be viewed as a combination
of only rights and principles, forming the two axes at the basis of our UP
Cube. In this way we also want to bring out two perspectives on privacy: that
of the data subjects and, respectively, that of the controllers/processors. We
define usable privacy criteria based on usability goals that we have extracted
from the whole text of the General Data Protection Regulation. The criteria are
designed to produce measurements of the level of usability with which the goals
are reached. Precisely, we measure effectiveness, efficiency, and satisfaction,
considering both the objective and the perceived usability outcomes, producing
measures of accuracy and completeness, of resource utilization (e.g., time,
effort, financial), and measures resulting from satisfaction scales. In the
long run, the UP Cube is meant to be the model behind a new certification
methodology capable of evaluating the usability of privacy, to the benefit of
common users. For industries, considering also the usability of privacy would
allow for greater business differentiation, beyond GDPR compliance.Comment: 41 pages, 2 figures, 1 table, and appendixe
Critical role for prokineticin 2 in CNS autoimmunity
Objective: To investigate the potential role of prokineticin 2 (PK2), a bioactive peptide involved in multiple biological functions including immune modulation, in CNS autoimmune demyelinating disease.
Methods: We investigated the expression of PK2 in mice with experimental autoimmune encephalomyelitis (EAE), the animal model of multiple sclerosis (MS), and in patients with relapsing-remitting MS. We evaluated the biological effects of PK2 on expression of EAE and on development of T-cell response against myelin by blocking PK2 in vivo with PK2 receptor antagonists. We treated with PK2 immune cells activated against myelin antigen to explore the immune-modulating effects of this peptide in vitro.
Results: Pk2 messenger RNA was upregulated in spinal cord and lymph node cells (LNCs) of mice with EAE. PK2 protein was expressed in EAE inflammatory infiltrates and was increased in sera during EAE. In patients with relapsing-remitting MS, transcripts for PK2 were significantly increased in peripheral blood mononuclear cells compared with healthy controls, and PK2 serum concentrations were significantly higher. A PK2 receptor antagonist prevented or attenuated established EAE in chronic and relapsing-remitting models, reduced CNS inflammation and demyelination, and decreased the production of interferon (IFN)-γ and interleukin (IL)-17A cytokines in LNCs while increasing IL-10. PK2 in vitro increased IFN-γ and IL-17A and reduced IL-10 in splenocytes activated against myelin antigen.
Conclusion: These data suggest that PK2 is a critical immune regulator in CNS autoimmune demyelination and may represent a new target for therapy
Canine circovirus and Canine adenovirus type 1 and 2 in dogs with parvoviral enteritis
Canine parvovirus type 2 (CPV-2) is one of the most relevant pathogens associated with enteritis in dogs and is frequently
reported in association with the detection of other pathogens in faeces. In this study the concomitant presence of Canine
circovirus (CanineCV) and Canine adenovirus (CAdV) DNA in faecal or intestine samples of 95 dogs with parvovirus
enteritis sampled in Italy (1995–2017) was investigated and the viruses identified were genetically characterised. Potential
correlations with the antigenic variant of CPV-2 and with signalment data and outcome were evaluated. Twenty-eight of 95
(29.5%) CPV-2 infected dogs tested positive to other viruses: 7/28 were also positive to CanineCV, 1/28 to CAdV-1, 18/28
to CAdV-2, 1/28 to CanineCV and CAdV-2, and 1/28 to CAdV-1 and CAdV-2. The frequency of CAdV DNA detection and
coinfections was significantly higher in purebred dogs compared to mixed breed ones (P = 0.002 and 0.009, respectively). The
presence of coinfection was not associated with any other relevant data available, including CPV-2 variant and final outcome.
The detection of CanineCV in a dog sampled in 2009 allowed to backdating its circulation in dogs. The eight CanineCV
completely sequenced were phylogenetically related to the CanineCV identified in dogs, wolves and a badger from Europe,
USA, Argentina and China. Nine CAdV were partially sequenced and phylogenetic analysis showed a separate branch for
the oldest CAdV-2 identified (1995). From the results obtained in this study population, CanineCV and CAdV coinfections
in dogs with parvoviral enteritis did not result in more severe disease
