    Improved extractability of carotenoids from tomato peels as side benefits of PEF treatment of tomato fruit for more energy-efficient steam-assisted peeling

    Abstract The combination of steam blanching (SB) with Pulsed Electric Fields (PEF) treatments of whole tomatoes, in addition to reducing the energy required for tomato peeling, can significantly contribute to the recovery of carotenoids from the peels. In this work, PEF (0.25-0-75 kV/cm, 1 kJ/kg) and SB (1 min at 50–70 °C), as pre-treatment prior to hand peeling, were investigated to assess their ability, separately and in combination, to induce the cell permeabilization of tomato peels, and hence to improve the carotenoids extraction in acetone (4 h at 25 °C). PEF and SB, by inducing significant damages at the cuticular level, caused the increase of the yield in total carotenoids (up to 188% for PEF and 189% for SB) and antioxidant power (up to 372% for PEF and 305% for SB) with respect to the peels from untreated tomatoes. The application of a combined treatment (PEF + SB) significantly increased the carotenoid content and the antioxidant power of the extracts, with a synergistic effect observed already at 60 °C (37.9 mg/100 g fresh weight tomato peels). HPLC analyses revealed that lycopene was the main carotenoid extracted and that neither PEF nor SB caused any selective release or degradation of lycopene. Results obtained from this study demonstrate that the integration of PEF in the processing line of tomato fruits prior to SB contributes to the valorization of tomato processing by-products

    Passively mode-locked fiber laser by utilizing TTG film on a D-shaped fiber as a saturable absorber

    In this paper, we propose a mode-locked fiber laser by utilizing single layer Trivial Transfer Graphene film (TTGF) as a saturable absorber (SA). The SA was deposited on the top of a side-polished D-shaped fiber. The SA was then integrated in the ring cavity configuration, with a clockwise light propagation. Three distinctive 2 m erbium doped fibers (EDFs) those are Metro-Gain15 EDF, Iso-Gain6 EDF and Iso-Gain12 EDF were used as gain media, interchangeable in the experiment. From the results, the Metro-Gain15 EDF gives the most proficient gain medium on generating a passively mode-locked fiber laser. The Metro-Gain15 Erbium doped mode-locked Laser was successfully producing ultrashort pulse with 8 nm spectral band-width, 13 MHz of repetition rate and 915 fs pulse duration. These outcomes demonstrated that TTGF deposited on the D-shaped fiber is a suitable component as an SA to produce a stable output passively mode-locked fiber laser for many optical fiber applications

    Validation of Multicultural Counselling Competencies Scale among Malaysian Counsellor Trainees: A Confirmatory Factor Analysis

    The competency of multicultural counselling is an aspect which needs to be mastered by a counsellor in order to provide good counselling services to clients especially when dealing with clients of different culture. The purpose of this research is to examine the validity and reliability of the Multicultural Counselling Competencies Scale which was developed based on the local culture by using confirmatory factor analysis (CFA). A total of 290 final year students participating in the first-degree counselling program from six public universities were selected randomly. The results of the analysis showed that the fit value for the comparative fit index (CFI), Tucker Lewis index (TLI), and normed fit index (NFI) respectively had fulfilled the fit value of 0.90 and above as suggested by the literature. The analysis also showed the average variance extracted (AVE) values for all three constructs were 0.669 (awareness), 0.764 (skills) and 0.642 (knowledge) respectively, indicating the validity convergent and discriminant reliability were achieved. CFA analysis has removed 7 out of 26 original items due to low loading factor. Overall, CFA analysis has successfully proven that competencies of multicultural counselling questionnaire as a multidimensional construct as it has been theorized and is appropriate for measuring the competencies of multicultural counselling in Malaysia

Cardiac dysfunction has only been described in adults thus far. We describe 3 cases of previously healthy children presenting with shock and COVID-19-related cardiac inflammation.     Cellobiohydrolase B of Aspergillus niger over-expressed in Pichia pastoris stimulates hydrolysis of oil palm empty fruit bunches

    Background. Aspergillus niger, along with many other lignocellulolytic fungi, has been widely used as a commercial workhorse for cellulase production. A fungal cellulase system generally includes three major classes of enzymes i.e.,β-glucosidases, endoglucanases and cellobiohydrolases. Cellobiohydrolases (CBH) are vital to the degradation of crystalline cellulose present in lignocellulosic biomass. However, A. niger naturally secretes low levels of CBH. Hence, recombinant production of A. niger CBH is desirable to increase CBH production yield and also to allow biochemical characterisation of the recombinant CBH from A. niger. Methods. In this study, the gene encoding a cellobiohydrolase B (cbhB) from A. niger ATCC 10574 was cloned and expressed in the methylotrophic yeast Pichia pastoris X-33. The recombinant CBHB was purified and characterised to study its biochemical and kinetic characteristics. To evaluate the potential of CBHB in assisting biomass conversion, CBHB was supplemented into a commercial cellulase preparation (Cellic ® CTec2) and was used to hydrolyse oil palm empty fruit bunch (OPEFB), one of the most abundant lignocellulosic waste from the palm oil industry. To attain maximum saccharification, enzyme loadings were optimised by response surface methodology and the optimum point was validated experimentally. Hydrolysed OPEFB samples were analysed using attenuated total reflectance FTIR spectroscopy (ATR-FTIR) to screen for any compositional changes upon enzymatic treatment. Results. Recombinant CBHB was over-expressed as a hyperglycosylated protein attached to N-glycans. CBHB was enzymatically active towards soluble substrates such as 4-methylumbelliferylβ-D-cellobioside (MUC), p-nitrophenyl-cellobioside (pNPC) and p-nitrophenyl-cellobiotrioside (pNPG3) but was not active towards crystalline substrates like Avicel ® and Sigmacell cellulose. Characterisation of purified CBHB using MUC as the model substrate revealed that optimum catalysis occurred at 50 °C and pH 4 but the enzyme was stable between pH 3 to 10 and 30 to 80 °C. Although CBHB on its own was unable to digest crystalline substrates, supplementation of CBHB (0.37%) with Cellic ® CTec2 (30%) increased saccharification of OPEFB by 27%. Compositional analyses of the treated OPEFB samples revealed that CBHB supplementation reduced peak intensities of both crystalline cellulose Iα and Iβ in the treated OPEFB samples. Discussion. Since CBHB alone was inactive against crystalline cellulose these data suggested that it might work synergistically with other components of Cellic R CTec2. CBHB supplements were desirable as they further increased hydrolysis of OPEFB when the performance of Cellic® CTec2 was theoretically capped at an enzyme loading of 34% in this study. Hence, A. niger CBHB was identified as a potential supplementary enzyme for the enzymatic hydrolysis of OPEFB

    Reaction optimization of Aspergillus niger α-L-arabinofuranosidase for improved arabinose production from kenaf stem

    There are abundant of lignocellulosic biomass readily available with varying compositions. Kenaf (Hibiscus cannabinus) is one of this lignocellulosic biomass that has a high content of hemicellulose. This particular hemicellulose is composed of high arabinoxylan, which is a xylan backbone with arabinofuranosyl branches. In order to hydrolyze arabinoxylan, a branching enzyme is needed. Therefore, α-L-arabinofuranosidase from Aspergillus niger ATCC120120 (AnabfA) was used to hydrolyzed pre-treated kenaf and the reaction conditions were optimized using central composite design (CCD) to produce a significant amount of arabinose. There were 20 experiments conducted with 1.68 star points and 6 replicates at the centre points. The reaction conditions that were optimized are enzyme loading, substrate concentration and reaction time in which resulted with 88 U AnabfA activity, 0.9% (w/v) and 48 h, respectively. These optimized conditions managed to increase the yield of arabinose with 47.17 mg/g arabinose produced

    Bright ligand-activatable fluorescent protein for high-quality multicolor live-cell super-resolution microscopy

    We introduce UnaG as a green-to-dark photoswitching fluorescent protein capable of high-quality super-resolution imaging with photon numbers equivalent to the brightest photoswitchable red protein. UnaG only fluoresces upon binding of a fluorogenic metabolite, bilirubin, enabling UV-free reversible photoswitching with easily controllable kinetics and low background under Epi illumination. The on- and off-switching rates are controlled by the concentration of the ligand and the excitation light intensity, respectively, where the dissolved oxygen also promotes the off-switching. The photo-oxidation reaction mechanism of bilirubin in UnaG suggests that the lack of ligand-protein covalent bond allows the oxidized ligand to detach from the protein, emptying the binding cavity for rebinding to a fresh ligand molecule. We demonstrate super-resolution single-molecule localization imaging of various subcellular structures genetically encoded with UnaG, which enables facile labeling and simultaneous multicolor imaging of live cells. UnaG has the promise of becoming a default protein for high-performance super-resolution imaging. Photoconvertible proteins occupy two color channels thereby limiting multicolour localisation microscopy applications.     The Glaciozyma antarctica genome reveals an array of systems that provide sustained responses towards temperature variations in a persistently cold habitat

    Extremely low temperatures present various challenges to life that include ice formation and effects on metabolic capacity. Psyhcrophilic microorganisms typically have an array of mechanisms to enable survival in cold temperatures. In this study, we sequenced and analysed the genome of a psychrophilic yeast isolated in the Antarctic region, Glaciozyma antarctica. The genome annotation identified 7857 protein coding sequences. From the genome sequence analysis we were able to identify genes that encoded for proteins known to be associated with cold survival, in addition to annotating genes that are unique to G. antarctica. For genes that are known to be involved in cold adaptation such as anti-freeze proteins (AFPs), our gene expression analysis revealed that they were differentially transcribed over time and in response to different temperatures. This indicated the presence of an array of adaptation systems that can respond to a changing but persistent cold environment. We were also able to validate the activity of all the AFPs annotated where the recombinant AFPs demonstrated anti-freeze capacity. This work is an important foundation for further collective exploration into psychrophilic microbiology where among other potential, the genes unique to this species may represent a pool of novel mechanisms for cold survival

    Service quality of private hospitals: The Iranian Patients' perspective

    <p>Abstract</p> <p>Background</p> <p>Highly competitive market in the private hospital industry has caused increasing pressure on them to provide services with higher quality. The aim of this study was to determine the different dimensions of the service quality in the private hospitals of Iran and evaluating the service quality from the patients' perspective.</p> <p>Methods</p> <p>A cross-sectional study was conducted between October and November 2010 in Tehran, Iran. The study sample was composed of 983 patients randomly selected from 8 private general hospitals. The study questionnaire was the SERVQUAL questionnaire, consisting of 21 items in service quality dimensions.</p> <p>Results</p> <p>The result of factor analysis revealed 3 factors, explaining 69% of the total variance. The total mean score of patients' expectation and perception was 4.91(SD = 0.2) and 4.02(SD = 0.6), respectively. The highest expectation and perception related to the tangibles dimension and the lowest expectation and perception related to the empathy dimension. The differences between perception and expectation were significant (p < 0.001). There was a significant difference between the expectations scores based on gender, education level, and previous hospitalization in that same hospital. Also, there was a significant difference between the perception scores based on insurance coverage, average length of stay, and patients' health conditions on discharge.</p> <p>Conclusion</p> <p>The results showed that SERVQUAL is a valid, reliable, and flexible instrument to monitor and measure the quality of the services in private hospitals of Iran. Our findings clarified the importance of creating a strong relationship between patients and the hospital practitioners/personnel and the need for hospital staff to be responsive, credible, and empathetic when dealing with patients.</p
