    A structural connectivity convergence zone in the ventral and anterior temporal lobes: Data-driven evidence from structural imaging

    The hub-and-spoke model of semantic cognition seeks to reconcile embodied views of a fully distributed semantic network with patient evidence, primarily from semantic dementia, who demonstrate modality-independent conceptual deficits associated with atrophy centred on the ventrolateral anterior temporal lobe. The proponents of this model have recently suggested that the temporal cortex is a graded representational space where concepts become less linked to a specific modality as they are processed farther away from primary and secondary sensory cortices and towards the ventral anterior temporal lobe. To explore whether there is evidence that the connectivity patterns of the temporal lobe converge in its ventral anterior end the current study uses three dimensional Laplacian eigenmapping, a technique that allows visualisation of similarity in a low dimensional space. In this space similarity is encoded in terms of distances between data points. We found that the ventral and anterior temporal lobe is in a unique position of being at the centre of mass of the data points within the connective similarity space. This can be interpreted as the area where the connectivity profiles of all other temporal cortex voxels converge. This study is the first to explicitly investigate the pattern of connectivity and thus provides the missing link in the evidence that the ventral anterior temporal lobe can be considered a multi-modal graded hub

    Tractography dissection variability: What happens when 42 groups dissect 14 white matter bundles on the same dataset?

    White matter bundle segmentation using diffusion MRI fiber tractography has become the method of choice to identify white matter fiber pathways in vivo in human brains. However, like other analyses of complex data, there is considerable variability in segmentation protocols and techniques. This can result in different reconstructions of the same intended white matter pathways, which directly affects tractography results, quantification, and interpretation. In this study, we aim to evaluate and quantify the variability that arises from different protocols for bundle segmentation. Through an open call to users of fiber tractography, including anatomists, clinicians, and algorithm developers, 42 independent teams were given processed sets of human whole-brain streamlines and asked to segment 14 white matter fascicles on six subjects. In total, we received 57 different bundle segmentation protocols, which enabled detailed volume-based and streamline-based analyses of agreement and disagreement among protocols for each fiber pathway. Results show that even when given the exact same sets of underlying streamlines, the variability across protocols for bundle segmentation is greater than all other sources of variability in the virtual dissection process, including variability within protocols and variability across subjects. In order to foster the use of tractography bundle dissection in routine clinical settings, and as a fundamental analytical tool, future endeavors must aim to resolve and reduce this heterogeneity. Although external validation is needed to verify the anatomical accuracy of bundle dissections, reducing heterogeneity is a step towards reproducible research and may be achieved through the use of standard nomenclature and definitions of white matter bundles and well-chosen constraints and decisions in the dissection process

    Emotional experiences and psychological well-being in 51 countries during the COVID-19 pandemic.

    The COVID-19 pandemic presents challenges to psychological well-being, but how can we predict when people suffer or cope during sustained stress? Here, we test the prediction that specific types of momentary emotional experiences are differently linked to psychological well-being during the pandemic. Study 1 used survey data collected from 24,221 participants in 51 countries during the COVID-19 outbreak. We show that, across countries, well-being is linked to individuals' recent emotional experiences, including calm, hope, anxiety, loneliness, and sadness. Consistent results are found in two age, sex, and ethnicity-representative samples in the United Kingdom (n = 971) and the United States (n = 961) with preregistered analyses (Study 2). A prospective 30-day daily diary study conducted in the United Kingdom (n = 110) confirms the key role of these five emotions and demonstrates that emotional experiences precede changes in well-being (Study 3). Our findings highlight differential relationships between specific types of momentary emotional experiences and well-being and point to the cultivation of calm and hope as candidate routes for well-being interventions during periods of sustained stress.     Revisiting the human uncinate fasciculus, its subcomponents and asymmetries with stem-based tractography and microdissection validation

    International audienceDespite its significant functional and clinical interest, the anatomy of the uncinate fasciculus (UF) has received little attention. It is known as a ‘hook-shaped’ fascicle connecting the frontal and anterior temporal lobes and is believed to consist of multiple subcomponents. However, the knowledge of its precise connectional anatomy in humans is lacking, and its subcomponent divisions are unclear. In the present study, we evaluate the anatomy of the UF and provide its detailed normative description in 30 healthy subjects with advanced particle-filtering tractography with anatomical priors and robustness to crossing fibers with constrained spherical deconvolution. We extracted the UF by defining its stem encompassing all streamlines that converge into a compact bundle, which consisted not only of the classic hook-shaped fibers, but also of straight horizontally oriented. We applied an automatic-clustering method to subdivide the UF bundle and revealed five subcomponents in each hemisphere with distinct connectivity profiles, including different asymmetries. A layer-by-layer microdissection of the ventral part of the external and extreme capsules using Klingler’s preparation also demonstrated five types of uncinate fibers that, according to their pattern, depth, and cortical terminations, were consistent with the diffusion-based UF subcomponents. The present results shed new light on the UF cortical terminations and its multicomponent internal organization with extended cortical connections within the frontal and temporal cortices. The different lateralization patterns we report within the UF subcomponents reconcile the conflicting asymmetry findings of the literature. Such results clarifying the UF structural anatomy lay the groundwork for more targeted investigations of its functional role, especially in semantic language processing

    “Border Theory: A New Point of Access into Literature” seeks to explore the use of a border-theoretical approach to literature. Border theories by Johan Schimanski, Stephen Wolfe, David Newman, Homi Bhabha, and a set of different border planes – the symbolic, epistemological, topographic, temporal and textual border plane – provide the scholarly framework for this thesis. Border theory studies the notion of hybridity, diversity and doubling, discusses the border and the change between the visible and invisibility, reviews binary oppositions and goes on to explore how borders can move beyond binaries and create a new space – a third space. The thesis uses three novels by China Miéville to examine border theory: Un Lun Dun (2007), The City and the City (2009) and Embassytown (2011). Miéville is a science fiction writer that introduces the genre of weird fiction. The main idea is that border theory, as its own theoretical point of access into the field of literary studies, can contribute with a new aspect of literary analysis, and that China Miéville’s contemporary weird fiction invites and benefits from a border-theoretical analysis. Border theory creates a new access-point into the theoretical analysis of novels, and further explores and shows how borders are represented in literature. Un Lun Dun presents the reader with a city and its abcity where the border between the two is not easy to see and understand. In addition, the abcity UnLondon is filled with strange spaces and extraordinary characters that give no shortage of borders to explore. The City and the City is a detective story that crosses the border between two sister cities – Besźel and Ul Qoma – with an invisible law enforcement agency operating within the invisible border between the two cities. Embassytown uses language as a barrier and a border between species. Set in the far future, the novel describes and explores complex relations and divisions of space, time and people – and the complications surrounding the inability to communicate. These novels have been chosen because of their borders-inquisitive qualities, and together they can each be read as a piece of the puzzle to a more complex understanding of borders and border crossings – both in literature and in real life. “Border Theory: A New Point of Access into Literature – A border-theoretical reading of China Miéville’s Un Lun Dun, The City and the City and Embassytown” is a thesis with a theoretical approach to contemporary literature – exploring the literary field of border theory

    Tissue engineering and the intervertebral disc: the challenges

    Disc degeneration is a common disorder. Although the back pain that can develop in association with this is rarely life-threatening, the annual cost in terms of morbidity, lost productivity, medical expenses and workers’ compensation benefits is significant. Surgical intervention as practised currently is directed towards removing the damaged or altered tissue. Development of new treatment modalities is critical as there is a growing consensus that the strategies used currently for symptomatic degenerative disc disease may not be effective. Accordingly, there is a need to develop an entirely new way to treat this disorder; regenerative medicine and tissue engineering approaches appear particularly promising in this regard. This paper reviews some of the challenges that currently are limiting the clinical application of this approach to the treatment of disc degeneration
