Reproducible Quantification of the Microstructure of Complex Quenched and Quenched and Tempered Steels Using Modern Methods of Machine Learning
Current conventional methods of evaluating microstructures are characterized by a high
degree of subjectivity and a lack of reproducibility. Modern machine learning (ML) approaches have
already shown great potential in overcoming these challenges. Once trained with representative
data in combination with objective ground truth, the ML model is able to perform a task properly
in a reproducible and automated manner. However, in highly complex use cases, it is often not
possible to create a definite ground truth. This study addresses this problem using the underlying
showcase of microstructures of highly complex quenched and quenched and tempered (Q/QT) steels.
A patch-wise classification approach combined with a sliding window technique provides a solution
for segmenting entire microphotographs where pixel-wise segmentation is not applicable since it
is hardly feasible to create reproducible training masks. Using correlative microscopy, consisting
of light optical microscope (LOM) and scanning electron microscope (SEM) micrographs, as well
as corresponding data from electron backscatter diffraction (EBSD), a training dataset of reference
states that covers a wide range of microstructures was acquired in order to train accurate and robust
ML models in order to classify LOM or SEM images. Despite the enormous complexity associated
with the steels treated here, classification accuracies of 88.8% in the case of LOM images and 93.7%
for high-resolution SEM images were achieved. These high accuracies are close to super-human
performance, especially in consideration of the reproducibility of the automated ML approaches
compared to conventional methods based on subjective evaluations through experts
Reproducibility of straylight measurement by C-Quant for assessment of retinal straylight using the compensation comparison method
Background: Straylight gives the appearance of a veil of light thrown over a person's retinal image when there is a strong light source present. We examined the reproducibility of the measurements by C-Quant, and assessed its correlation to characteristics of the eye and subjects' age. Participants and Methods: Five repeated straylight measurements were taken using the dominant eye of 45 healthy subjects (age 21-59) with a BCVA of 20/20: 14 emmetropic, 16 myopic, eight hyperopic and seven with astigmatism. We assessed the extent of reproducibility of straylight measures using the intraclass correlation coefficient. Results: The mean straylight value of all measurements was 1.01 (SD 0.23, median 0.97, interquartile range 0.85-1.1). Per 10years of age, straylight increased in average by 0.10 (95%CI 0.04 to 0.16, pâ<â0.01]. We found no independent association of refraction (range â5.25 dpt to +2 dpt) on straylight values (0.001; 95%CI â0.022 to 0.024, pâ=â0.92). Compared to emmetropic subjects, myopia reduced straylight (â.011; â0.024 to 0.02, pâ=â0.11), whereas higher straylight values (0.09; â0.01 to 0.20, pâ=â0.09) were observed in subjects with blue irises as compared to dark-colored irises when correcting for age. The intraclass correlation coefficient (ICC) of repeated measurements was 0.83 (95%CI 0.76 to 0.90). Conclusions: Our study showed that straylight measurements with the C-Quant had a high reproducibility, i.e. a lack of large intra-observer variability, making it appropriate to be applied in long-term follow-up studies assessing the long-term effect of surgical procedures on the quality of visio
Efficient reconstruction of prior austenite grains in steel from etched light optical micrographs using deep learning and annotations from correlative microscopy
The high-temperature austenite phase is the initial state of practically all
technologically relevant hot forming and heat treatment operations in steel
processing. The phenomena occurring in austenite, such as recrystallization or
grain growth, can have a decisive influence on the subsequent properties of the
material. After the hot forming or heat treatment process, however, the
austenite transforms into other microstructural constituents and information
on the prior austenite morphology are no longer directly accessible. There are
established methods available for reconstructing former austenite grain
boundaries via metallographic etching or electron backscatter diffraction
(EBSD) which both exhibit shortcomings. While etching is often difficult to
reproduce and strongly depend on the investigated steelâs alloying concept,
EBSD acquisition and reconstruction is rather time-consuming. But in fact,
though, light optical micrographs of steels contrasted with conventional Nital
etchant also contain information about the former austenite grains. However,
relevant features are not directly apparent or accessible with conventional
segmentation approaches. This work presents a deep learning (DL)
segmentation of prior austenite grains (PAG) from Nital etched light optical
micrographs. The basis for successful segmentation is a correlative
characterization from EBSD, light and scanning electron microscopy to
specify the ground truth required for supervised learning. The DL model
shows good and robust segmentation results. While the intersection over
union of 70% does not fully reflect the model performance due to the
inherent uncertainty in PAG estimation, a mean error of 6.1% in mean grain
size derived from the segmentation clearly shows the high quality of the result
ISRCTN12125882 - Influence of topical anti-VEGF (Ranibizumab) on the outcome of filtration surgery for glaucoma - Study Protocol
<p>Abstract</p> <p>Background</p> <p>Excessive wound healing, with scarring of the episcleral tissue or encapsulation of the filtering bleb is the main reason for failure in trabeculectomy. Ranibizumab, an inhibitor of the Vascular Endothelial Growth Factor (VEGF), is seen as a promising candidate to prevent or treat extensive wound healing. We describe the design of a two phased study, i) assessing the local tolerability and safety of topical ranibizumab and ii) assessing the efficacy of topical ranibizumab against placebo in patients who underwent trabeculectomy with mitomycin C combined with phacoemulsification and intra ocular lens (IOL) implantation.</p> <p>Methods/Design</p> <p>In the first phase five patients that had trabeculectomy with mitomycin C combined with phacoemulsification and IOL implantation will be treated with topical ranibizumab (Lucentis<sup>Âź</sup>) eye drops (2 mg/ml) four times daily for one month. The treatment will be started at the first postoperative day. Patients will be assessed for local and systemic side effects using a standardised schedule. In the second phase, after successful completion of phase 1, consenting eligible patients who underwent trabeculectomy with mitomycin C combined with phacoemulsification and IOL implantation will be randomised to either receive topical ranibizumab eye drops (2 mg/ml) four times daily for 1 month or placebo (BSS 4x/d for 1 month). Patients will be reviewed weekly for 4 weeks until conjunctival sutures are removed. Further follow up examinations are planned after 3 and six months. Assessment of differences in the intraocular eye pressure will be considered primary, and bleb appearance/vascularisation using a standardized photography and the Moorfields bleb grading system, postoperative intraocular pressure and conjunctival wound healing problems will be considered secondary outcome parameters.</p> <p>Discussion</p> <p>Anti-VEGF-antibodies might be more effective in preventing scaring and might have fewer toxic side effects than the currently used anti-metabolites and may replace them in the long term.</p> <p>Trial Registration</p> <p>ISRCTN: <a href="http://www.controlled-trials.com/ISRCTN12125882">ISRCTN12125882</a></p
Crowdsourced mapping of unexplored target space of kinase inhibitors
Despite decades of intensive search for compounds that modulate the activity of particular protein targets, a large proportion of the human kinome remains as yet undrugged. Effective approaches are therefore required to map the massive space of unexplored compound-kinase interactions for novel and potent activities. Here, we carry out a crowdsourced benchmarking of predictive algorithms for kinase inhibitor potencies across multiple kinase families tested on unpublished bioactivity data. We find the top-performing predictions are based on various models, including kernel learning, gradient boosting and deep learning, and their ensemble leads to a predictive accuracy exceeding that of single-dose kinase activity assays. We design experiments based on the model predictions and identify unexpected activities even for under-studied kinases, thereby accelerating experimental mapping efforts. The open-source prediction algorithms together with the bioactivities between 95 compounds and 295 kinases provide a resource for benchmarking prediction algorithms and for extending the druggable kinome. The IDG-DREAM Challenge carried out crowdsourced benchmarking of predictive algorithms for kinase inhibitor activities on unpublished data. This study provides a resource to compare emerging algorithms and prioritize new kinase activities to accelerate drug discovery and repurposing efforts

Using correlative microscopy, consisting of light optical microscope (LOM) and scanning electron microscope (SEM) micrographs, as well as corresponding data from electron backscatter diffraction (EBSD), a training dataset of reference states that covers a wide range of microstructures was acquired in order to train accurate and robust ML models in order to classify LOM or SEM images. Despite the enormous complexity associated with the steels treated here, classification accuracies of 88.8% in the case of LOM images and 93.7% for high-resolution SEM images were achieved. These high accuracies are close to super-human performance, especially in consideration of the reproducibility of the automated ML approaches compared to conventional methods based on subjective evaluations through experts
Fusion of Azaâ and Oxadiborepins with Furans in a Reversible RingâOpening Process Furnishes Versatile Building Blocks for Extended ÏâConjugated Materials
A modular synthesis of both difurooxaâ and difuroazadiborepins from a common precursor is demonstrated. Starting from 2,2âČâbifuran, after protection of the positions 5 and 5â with bulky silyl groups, formation of the novel polycycles proceeds through opening of the furan rings to a dialkyne and subsequent reâcyclization in the borylation step. The resulting bifuranâfused diborepins show pronounced stability, highly planar tricyclic structures, and intense blue light emission. Deprotection and transformation into dibrominated building blocks that can be incorporated into Ïâextended materials can be performed in one step. Detailed DFT calculations provide information about the aromaticity of the constituent rings of this polycycle
DithiopheneâFused Oxadiborepins and Azadiborepins: A New Class of Highly Fluorescent Heteroaromatics
Access to dithiopheneâfused oxadiborepins and the first azadiborepins attained via a modular synthesis route are presented. The new compounds emit intense blue light, some of which demonstrate fluorescence quantum yields close to unity. Cyclic voltammetry (CV) revealed electrochemically reversible oneâelectron reduction processes. The weak aromatic character of the novel 1,2,7âazadiborepin ring is demonstrated with inâdepth theoretical investigations using nucleusâindependent chemical shift (NICS) scans and anisotropy of the induced current density (ACID) calculations
