The Role of BDNF/TrkB Signaling in Acute Amphetamine-Induced Locomotor Activity and Opioid Peptide Gene Expression in the Rat Dorsal Striatum
Exposure to psychostimulants increases brain-derived neurotrophic factor (BDNF) mRNA and protein levels in the cerebral cortex and subcortical structures. Because BDNF is co-localized with dopamine and glutamate in afferents to the striatum of rats, it may be co-released with those neurotransmitters upon stimulation. Further, there may be an interaction between the intracellular signaling cascades activated by dopamine, glutamate, and TrkB receptors in medium spiny striatal neurons. In the present study, the effect of acute amphetamine administration on TrkB phosphorylation, as an indirect indicator of activation, and striatal gene expression, was evaluated. In Experiment 1, 15 min or 2 h after a single saline or amphetamine (2.5 mg/kg, i.p.) injection, the caudate–putamen (CPu), nucleus accumbens (NAc), and dorsomedial prefrontal cortex (dmPFC) were extracted and processed for phospho (p)-TrkB immunoreactivity. Immunoprecipitation analyses indicated that neither the tyrosine phosphorylation (p-Tyr) or autophosphorylation sites of TrkB (706) were changed in NAc, CPu, or dmPFC 15 min after amphetamine administration. In contrast, p-Tyr and the PLCγ phosphorylation site of TrkB (816) were increased in the NAc and CPu 2 h after amphetamine. In Experiment 2, intra-striatal infusion of the tyrosine kinase inhibitor, K252a, increased amphetamine-induced vertical activity but not total distance traveled. In addition, K252a inhibited amphetamine-induced preprodynorphin, but not preproenkephalin, mRNA expression in the striatum. These data indicate that acute amphetamine administration induces p-TrkB activation and signaling in a time- and brain region-dependent manner and that TrkB/BDNF signaling plays an important role in amphetamine-induced behavior and striatal gene expression
Modulation Instability of Ultrashort Pulses in Quadratic Nonlinear Media beyond the Slowly Varying Envelope Approximation
We report a modulational instability (MI) analysis of a mathematical model
appropriate for ultrashort pulses in cascaded quadratic-cubic nonlinear media
beyond the so-called slowly varying envelope approximation. Theoretically
predicted MI properties are found to be in good agreement with numerical
simulation. The study shows the possibility of controlling the generation of MI
and formation of solitons in a cascaded quadratic-cubic media in the few cycle
regimes. We also find that stable propagation of soliton-like few-cycle pulses
in the medium is subject to the fulfilment of the modulation instability
criteria
Nonclassical statistics of intracavity coupled waveguides: the quantum optical dimer
A model is proposed where two nonlinear waveguides are contained
in a cavity suited for second-harmonic generation. The evanescent wave coupling
between the waveguides is considered as weak, and the interplay between this
coupling and the nonlinear interaction within the waveguides gives rise to
quantum violations of the classical limit. These violations are particularly
strong when two instabilities are competing, where twin-beam behavior is found
as almost complete noise suppression in the difference of the fundamental
intensities. Moreover, close to bistable transitions perfect twin-beam
correlations are seen in the sum of the fundamental intensities, and also the
self-pulsing instability as well as the transition from symmetric to asymmetric
states display nonclassical twin-beam correlations of both fundamental and
second-harmonic intensities. The results are based on the full quantum Langevin
equations derived from the Hamiltonian and including cavity damping effects.
The intensity correlations of the output fields are calculated
semi-analytically using a linearized version of the Langevin equations derived
through the positive-P representation. Confirmation of the analytical results
are obtained by numerical simulations of the nonlinear Langevin equations

Textile sensors for stab and cut detection
Manufacturers are aiming for more flexible and lightweight protective clothing to increase wearing comfort. A cardigan with a knitted stab-resistant inlay and an alarm system is presented. The stab-resistant inlay is based on a multilayer ultra-high molecular weight poly ethylene (UHMW-PE) fabric. Stab resistance was evaluated according to the standard of the Association of Test Laboratories for Bullet, Stab or Pike Resistant Materials and Construction Standard (VPAM 2011). Furthermore sensors for the detection of cuts and pressure were integrated. Both sensors can trigger alarms if the wearer is attacked. Normal pressure occurring through leaning on a wall or sitting is filtered out and does not trigger an alarm
Effects of osteopontin inhibition on radiosensitivity of MDA-MB-231 breast cancer cells
<p>Abstract</p> <p>Background</p> <p>Osteopontin (OPN) is a secreted glycophosphoprotein that is overexpressed in various tumors, and high levels of OPN have been associated with poor prognosis of cancer patients. In patients with head and neck cancer, high OPN plasma levels have been associated with poor prognosis following radiotherapy. Since little is known about the relationship between OPN expression and radiosensitivity, we investigated the cellular and radiation induced effects of OPN siRNA in human MDA-MB-231 breast cancer cells.</p> <p>Methods</p> <p>MDA-MB-231 cells were transfected with OPN-specific siRNAs and irradiated after 24 h. To verify the OPN knockdown, we measured the OPN mRNA and protein levels using qRT-PCR and Western blot analysis. Furthermore, the functional effects of OPN siRNAs were studied by assays to assess clonogenic survival, migration and induction of apoptosis.</p> <p>Results</p> <p>Treatment of MDA-MB-231 cells with OPN siRNAs resulted in an 80% decrease in the OPN mRNA level and in a decrease in extracellular OPN protein level. Transfection reduced clonogenic survival to 42% (p = 0.008), decreased the migration rate to 60% (p = 0.15) and increased apoptosis from 0.3% to 1.7% (p = 0.04). Combination of OPN siRNA and irradiation at 2 Gy resulted in a further reduction of clonogenic survival to 27% (p < 0.001), decreased the migration rate to 40% (p = 0.03) and increased apoptosis to 4% (p < 0.005). Furthermore, OPN knockdown caused a weak radiosensitization with an enhancement factor of 1.5 at 6 Gy (p = 0.09) and a dose modifying factor (DMF<sub>10</sub>) of 1.1.</p> <p>Conclusion</p> <p>Our results suggest that an OPN knockdown improves radiobiological effects in MDA-MB-231 cells. Therefore, OPN seems to be an attractive target to improve the effectiveness of radiotherapy.</p
Quantum properties of transverse pattern formation in second-harmonic generation
We investigate the spatial quantum noise properties of the one dimensional
transverse pattern formation instability in intra-cavity second-harmonic
generation. The Q representation of a quasi-probability distribution is
implemented in terms of nonlinear stochastic Langevin equations. We study these
equations through extensive numerical simulations and analytically in the
linearized limit. Our study, made below and above the threshold of pattern
formation, is guided by a microscopic scheme of photon interaction underlying
pattern formation in second-harmonic generation. Close to the threshold for
pattern formation, beams with opposite direction of the off-axis critical wave
numbers are shown to be highly correlated. This is observed for the fundamental
field, for the second harmonic field and also for the cross-correlation between
the two fields. Nonlinear correlations involving the homogeneous transverse
wave number, which are not identified in a linearized analysis, are also
described. The intensity differences between opposite points of the far fields
are shown to exhibit sub-Poissonian statistics, revealing the quantum nature of
the correlations. We observe twin beam correlations in both the fundamental and
second-harmonic fields, and also nonclassical correlations between them.Comment: 18 pages, 17 figures, submitted to Phys. Rev.
Systems of education governance and cultures of justice in Ireland, Scotland and Pakistan
This chapter compares the issue of cultures of justice in the systems of education governance in three education systems: Ireland, Scotland and Pakistan. The focus for the comparison are the current policies which shape the regulation of education. These policies were reviewed to identify key issues relating to social justice and equality, decision-making and accountability. From the analysis of each system, three central issues were identified: firstly, the improvement of a state education system; secondly, the degree of decentralisation and centralisation in governance structures and thirdly, the expectations placed on school leaders. The chapter concludes by discussing the tensions between the drive for system improvement and opportunities for school leaders to build strategies to address issues of inequality in schools
Investigations on antibody binding to a micro-cantilever coated with a BAM pesticide residue
The attachment of an antibody to an antigen-coated cantilever has been investigated by repeated experiments, using a cantilever-based detection system by Cantion A/S. The stress induced by the binding of a pesticide residue BAM (2,6 dichlorobenzamide) immobilized on a cantilever surface to anti-BAM antibody is measured using the CantiLab4© system from Cantion A/S with four gold-coated cantilevers and piezo resistive readout. The detection mechanism is in principle label-free, but fluorescent-marked antibodies have been used to subsequently verify the binding on the cantilever surface. The bending and increase in mass of each cantilever has also been investigated using a light interferometer and a Doppler Vibrometer. The system has been analyzed during repeated measurements to investigate whether the CantiLab4© system is a suited platform for a pesticide assay system
