    Potent Adjuvanticity of a Pure TLR7-Agonistic Imidazoquinoline Dendrimer

    Engagement of toll-like receptors (TLRs) serve to link innate immune responses with adaptive immunity and can be exploited as powerful vaccine adjuvants for eliciting both primary and anamnestic immune responses. TLR7 agonists are highly immunostimulatory without inducing dominant proinflammatory cytokine responses. We synthesized a dendrimeric molecule bearing six units of a potent TLR7/TLR8 dual-agonistic imidazoquinoline to explore if multimerization of TLR7/8 would result in altered activity profiles. A complete loss of TLR8-stimulatory activity with selective retention of the TLR7-agonistic activity was observed in the dendrimer. This was reflected by a complete absence of TLR8-driven proinflammatory cytokine and interferon (IFN)-γ induction in human PBMCs, with preservation of TLR7-driven IFN-α induction. The dendrimer was found to be superior to the imidazoquinoline monomer in inducing high titers of high-affinity antibodies to bovine α-lactalbumin. Additionally, epitope mapping experiments showed that the dendrimer induced immunoreactivity to more contiguous peptide epitopes along the amino acid sequence of the model antigen.This work was supported by National Institutes of Health/National Institute of Allergy and Infectious Diseases contract HHSN272200900033C

    Dynamic transformation of small Ni particles during methanation of CO under fluctuating reaction conditions monitored by X-ray absorption spectroscopy

    A 10 wt.-% Ni/Al2_{2}O3_{3} catalyst with Ni particles of about 4 nm was prepared and applied in the methanation of CO2_{2} under dynamic reaction conditions. Fast phase transformations between metallic Ni, NiO and NiCO3_{3} were observed under changing reaction atmospheres using operando X-ray absorption spectroscopy (XAS). Removing H2_{2} from the feed gas and, thus, simulating a H2_{2} dropout during the methanation reaction led to oxidation of the active sites. The initial reduced state of the Ni particles could not be recovered under methanation atmosphere (H2_{2}/CO2_{2} = 4); this was only possible with an effective reactivation step applying H2_{2} at increased temperatures. Furthermore, the cycling of the gas atmospheres resulted in a steady deactivation of the catalyst. Operando XAS is a powerful tool to monitor these changes and the behavior of the catalyst under working conditions to improve the understanding of the catalytic processes and deactivation phenomena

    One-dimensional massive quantum particles (or 1+1-dimensional random walks) with short-ranged multi-particle interactions are studied by exact renormalization group methods. With repulsive pair forces, such particles are known to scale as free fermions. With finite mm-body forces (m = 3,4,...), a critical instability is found, indicating the transition to a fermionic bound state. These unbinding transitions represent new universality classes of interacting fermions relevant to polymer and membrane systems. We explore dynamics inside and between the far right and the far left, as well as the relation between the structure of the network and sentiment. The 2015 Greek political context offers a unique opportunity to investigate political communication in times of political intensity and crisis.     Doing what others do: norms, science, and collective action on global warming

    Does rhetoric highlighting social norms or mentioning science in a communication affect individuals’ beliefs about global warming and/or willingness to take action? We draw from framing theory and collective-interest models of action to motivate hypotheses that are tested in two large web-based survey-experiments using convenience samples. Our results show that attitudes about global warming, support for policies that would reduce carbon emissions, and behavioral intentions to take voluntary action are strongly affected by norm- and science-based interventions. This has implications for information campaigns targeting voluntary efforts to promote lifestyle changes that would reduce greenhouse gas emissions

    Numerical observation of non-axisymmetric vesicles in fluid membranes

    By means of Surface Evolver (Exp. Math,1,141 1992), a software package of brute-force energy minimization over a triangulated surface developed by the geometry center of University of Minnesota, we have numerically searched the non-axisymmetric shapes under the Helfrich spontaneous curvature (SC) energy model. We show for the first time there are abundant mechanically stable non-axisymmetric vesicles in SC model, including regular ones with intrinsic geometric symmetry and complex irregular ones. We report in this paper several interesting shapes including a corniculate shape with six corns, a quadri-concave shape, a shape resembling sickle cells, and a shape resembling acanthocytes. As far as we know, these shapes have not been theoretically obtained by any curvature model before. In addition, the role of the spontaneous curvature in the formation of irregular crenated vesicles has been studied. The results shows a positive spontaneous curvature may be a necessary condition to keep an irregular crenated shape being mechanically stable.Comment: RevTex, 14 pages.     Processed data on the night-time use of screen-based media devices and adolescents' sleep quality and health-related quality of life

    The data presented in this article relate to the research article entitled “Night-time screen-based media device use and adolescents' sleep and health-related quality of life” [1]. The present data reports findings from the investigation of the relationship between night-time screen-based media devices (SBMD) use and both sleep quality and health-related quality of life (HRQoL) among 11 to 12-year-olds. Baseline data from a large cohort of 6,616 adolescents from 39 schools in and around London, UK, participating in the Study of Cognition Adolescents and Mobile Phone (SCAMP) were analysed. Self-report data on adolescents’ use of any SBMD (mobile phone, tablet, laptop, television etc.) were the main exposures of interest. Mobile phone and television were the most commonly used portable and non-portable device, respectively. Sleep variables were derived from self-reported weekday and/or weekend bedtime, sleep onset latency (SOL) and wake time. Sleep quality was assessed using four standardised dimensions from the Swiss Health Survey. HRQoL was estimated using the KIDSCREEN-10 questionnaire

    Genome-Wide Association Studies of Coffee Intake in UK/US Participants of European Ancestry Uncover Gene-Cohort Influences

    Coffee is one of the most widely consumed beverages. We performed a genome-wide association study (GWAS) of coffee intake in US-based 23andMe participants (N =130,153) and identified 7 significant loci, with many replicating in three multi-ancestral cohorts. We examined genetic correlations and performed a phenome-wide association study across thousands of biomarkers and health and lifestyle traits, then compared our results to the largest available GWAS of coffee intake from UK Biobank (UKB; N =334,659). The results of these two GWAS were highly discrepant. We observed positive genetic correlations between coffee intake and psychiatric illnesses, pain, and gastrointestinal traits in 23andMe that were absent or negative in UKB. Genetic correlations with cognition were negative in 23andMe but positive in UKB. The only consistent observations were positive genetic correlations with substance use and obesity. Our study shows that GWAS in different cohorts could capture cultural differences in the relationship between behavior and genetics
