Spatially Extended Low Ionization Emission Regions (LIERs) at
We present spatially resolved emission diagnostics for eight
galaxies that demonstrate extended low ionization emission-line regions (LIERs)
over kpc scales. Eight candidates are selected based on their spatial extent
and emission line fluxes from slitless spectroscopic observations with the
HST/WFC3 G141 and G800L grisms in the well-studied GOODS survey fields. Five of
the candidates (62.5%) are matched to X-ray counterparts in the \textit{Chandra
X-Ray Observatory} Deep Fields. We modify the traditional
Baldwin-Philips-Terlevich (BPT) emission line diagnostic diagram to use
[SII]/(H+[NII]) instead of [NII]/H to overcome the blending of
[NII] and H+[NII] in the low resolution slitless grism spectra. We
construct emission line ratio maps and place the individual pixels in the
modified BPT. The extended LINER-like emission present in all of our
candidates, coupled with X-Ray properties consistent with star-forming galaxies
and weak [OIII]5007\AA\ detections, is inconsistent with purely
nuclear sources (LINERs) driven by active galactic nuclei. While recent
ground-based integral field unit spectroscopic surveys have revealed
significant evidence for diffuse LINER-like emission in galaxies within the
local universe , this work provides the first evidence for the

Post-mortem degradation of myosin heavy chain in intact fish muscle: Effects of pH and enzyme inhibitors
Fish muscle is rapidly degraded during post-mortem storage, due to proteolytic enzymes acting probably both on muscle cells and connective tissue. In this work we have developed a model system which may be used to study the enzymatic degradation occurring in intact post-mortem fish muscle. Degradation of myosin heavy chain (MHC) was monitored in muscle with pH adjusted to 6.05, 6.3 and 6.9 and in the presence of the enzyme inhibitors PMSF, EDTA, phenanthroline, pepstatin A, antipain, E-64 and the cysteine proteinase activator dithiothreithol (DTT). After storage, myofibrillar proteins were isolated and MHC-specific antibodies used to study the degradation in the different samples. MHC from muscle with pH 6.05 and 6.3 was degraded, while no severe degradation was observed at pH 6.9. Introduction of enzyme inhibitors into the muscle tissue clearly showed that mainly cysteine and aspartic proteinases are responsible for the in situ MHC degradation. This is supported by the severe breakdown of MHC in the muscle samples containing DTT.Peer reviewe
Genome-wide nucleosome map and cytosine methylation levels of an ancient human genome.
yesEpigenetic information is available from contemporary organisms, but is difficult to track back in evolutionary time.
Here, we show that genome-wide epigenetic information can be gathered directly from next-generation sequence reads of
DNA isolated from ancient remains. Using the genome sequence data generated from hair shafts of a 4000-yr-old Paleo-
Eskimo belonging to the Saqqaq culture, we generate the first ancient nucleosome map coupled with a genome-wide
survey of cytosine methylation levels. The validity of both nucleosome map and methylation levels were confirmed by the
recovery of the expected signals at promoter regions, exon/intron boundaries, and CTCF sites. The top-scoring nucleosome
calls revealed distinct DNA positioning biases, attesting to nucleotide-level accuracy. The ancient methylation
levels exhibited high conservation over time, clustering closely with modern hair tissues. Using ancient methylation
information, we estimated the age at death of the Saqqaq individual and illustrate how epigenetic information can be used
to infer ancient gene expression. Similar epigenetic signatures were found in other fossil material, such as 110,000- to
130,000-yr-old bones, supporting the contention that ancient epigenomic information can be reconstructed from a deep
past. Our findings lay the foundation for extracting epigenomic information from ancient samples, allowing shifts in
epialleles to be tracked through evolutionary time, as well as providing an original window into modern epigenomics
The Marriage Metaphor of Jesus and the Samaritan Woman: A New Interpretative Reading of John 4:1-45
This paper explores the marriage metaphor in John 4:1–45, arguing that Jesus’s encounter with the Samaritan woman is a symbolic representation of the relationship between Christ and the Church. Through an exegetical analysis of the text, the study examines key elements such as the double entendre of the well motif, living water, and the concept of worship. The results of this study reveal the depth of John\u27s narrative strategy in intentionally presenting Jesus as the Messianic Bridegroom, illustrated through his explicit use of Old Testament marriage motifs. The study concludes that the episode of Jesus and the Samaritan woman functions as a theological microcosm in God’s salvific plan
Do we all grieve the same? A multigroup test of the dimensional structure of prolonged grief disorder among Danish bereaved partners and children
Prolonged Grief Disorder (PGD) is a newly recognized mental disorder in ICD-11 and DSM-5-TR. Several studies using exploratory factor analysis have found a unidimensional structure of the Prolonged Grief-13 (PG-13) measure of PGD. The recently published ICD-11 proposal proposes a distinction between two clusters of symptoms: Separation distress symptoms and associated cognitive, emotional and behavioral symptoms. The aim of the current study is to test competing factor structures of PGD in Danish samples of bereaved. Confirmatory factor analysis was used to test competing models of PGD among two samples of in total 1093 adults that completed the questionnaires 6 months post loss of either a parent or a partner. Convergent and divergent validity was tested via the relationship to depression, anxiety, post-traumatic stress disorder (PTSD) and general wellbeing using regression analysis. The Danish version of the PG 13 appeared to be both valid and reliable. A two-factor model reflecting the division of core- and associated symptoms of prolonged grief disorder provided the best description of the PG-13 among Danish bereaved adults and there was evidence of partial structural invariance of the latent structure of PGD across bereavement types. Convergent and divergent validity analysis supported the validity of the two-factor model of PGD. Significant outcomes:: A latent variable model differing between core- and associated symptomatology of grief is supported. The Danish translation of PG-13 is a valid measure of prolonged grief symptomatology.</p
Do early symptoms of prolonged grief disorder lead to symptoms of posttraumatic stress disorder and depression?:A longitudinal register-based study of the two first years of bereavement
Introduction: Symptoms of prolonged grief disorder (PGD), depression, and posttraumatic stress disorder (PTSD) often emerge concurrently in bereavement. The understanding of temporal relationships between these syndromes in a general bereaved population is limited. This study aims to investigate temporal relationships between these syndromes from 2 months postloss throughout the two first years of bereavement. Method: Data were derived from a registry-based cohort study with 1,224 adult participants, who lost a spouse or parent. Participants completed self-report measures of PGD, depression, and PTSD at 2, 6, 11, 18, and 26 months postloss. Random intercept cross-lagged panel analyses examined the temporal relationships between PGD, PTSD, and depression. Results: In spousal and parental bereavement, high levels of grief symptoms at 2 months postloss predicted subsequent high symptoms of PTSD and depression at 6 months postloss, not vice versa. PGD, PTSD, and depression showed strong intertwined relationships over the two first years of bereavement. Between-person differences explained an increasingly large amount of variance in symptoms of PGD, PTSD, and depression over time. Losing a spouse and younger age was associated with higher symptoms of PGD, PTSD, and depression compared to losing a parent and older age. Conclusion: In the early years of bereavement, large differences exist between bereaved individuals in general levels of PGD, PTSD, and depression. Within bereaved individuals, the temporal relationships between these syndromes become increasingly complex and intertwined over time. Findings should be interpreted with respect to the nonclinical sample and self-report data used.</p
From food to pest: Conversion factors determine switches between ecosystem services and disservices
Ecosystem research focuses on goods and services, thereby ascribing beneficial values to the ecosystems. Depending on the context, however, outputs from ecosystems can be both positive and negative. We examined how provisioning services of wild animals and plants can switch between being services and disservices. We studied agricultural communities in Laos to illustrate when and why these switches take place. Government restrictions on land use combined with economic and cultural changes have created perceptions of rodents and plants as problem species in some communities. In other communities that are maintaining shifting cultivation practices, the very same taxa were perceived as beneficial. We propose conversion factors that in a given context can determine where an individual taxon is located along a spectrum from ecosystem service to disservice, when, and for whom. We argue that the omission of disservices in ecosystem service accounts may lead governments to direct investments at inappropriate targets
Mitochondrial proteomics on human fibroblasts for identification of metabolic imbalance and cellular stress
<p>Abstract</p> <p>Background</p> <p>Mitochondrial proteins are central to various metabolic activities and are key regulators of apoptosis. Disturbance of mitochondrial proteins is therefore often associated with disease. Large scale protein data are required to capture the mitochondrial protein levels and mass spectrometry based proteomics is suitable for generating such data. To study the relative quantities of mitochondrial proteins in cells from cultivated human skin fibroblasts we applied a proteomic method based on nanoLC-MS/MS analysis of iTRAQ-labeled peptides.</p> <p>Results</p> <p>When fibroblast cultures were exposed to mild metabolic stress – by cultivation in galactose medium- the amount of mitochondria appeared to be maintained whereas the levels of individual proteins were altered. Proteins of respiratory chain complex I and IV were increased together with NAD<sup>+</sup>-dependent isocitrate dehydrogenase of the citric acid cycle illustrating cellular strategies to cope with altered energy metabolism. Furthermore, quantitative protein data, with a median standard error below 6%, were obtained for the following mitochondrial pathways: fatty acid oxidation, citric acid cycle, respiratory chain, antioxidant systems, amino acid metabolism, mitochondrial translation, protein quality control, mitochondrial morphology and apoptosis.</p> <p>Conclusion</p> <p>The robust analytical platform in combination with a well-defined compendium of mitochondrial proteins allowed quantification of single proteins as well as mapping of entire pathways. This enabled characterization of the interplay between metabolism and stress response in human cells exposed to mild stress.</p
