Enforced Expression of the Transcriptional Coactivator OBF1 Impairs B Cell Differentiation at the Earliest Stage of Development
OBF1, also known as Bob.1 or OCA-B, is a B lymphocyte-specific transcription factor which coactivates Oct1 and Oct2 on B cell specific promoters. So far, the function of OBF1 has been mainly identified in late stage B cell populations. The central defect of OBF1 deficient mice is a severely reduced immune response to T cell-dependent antigens and a lack of germinal center formation in the spleen. Relatively little is known about a potential function of OBF1 in developing B cells. Here we have generated transgenic mice overexpressing OBF1 in B cells under the control of the immunoglobulin heavy chain promoter and enhancer. Surprisingly, these mice have greatly reduced numbers of follicular B cells in the periphery and have a compromised immune response. Furthermore, B cell differentiation is impaired at an early stage in the bone marrow: a first block is observed during B cell commitment and a second differentiation block is seen at the large preB2 cell stage. The cells that succeed to escape the block and to differentiate into mature B cells have post-translationally downregulated the expression of transgene, indicating that expression of OBF1 beyond the normal level early in B cell development is deleterious. Transcriptome analysis identified genes deregulated in these mice and Id2 and Id3, two known negative regulators of B cell differentiation, were found to be upregulated in the EPLM and preB cells of the transgenic mice. Furthermore, the Id2 and Id3 promoters contain octamer-like sites, to which OBF1 can bind. These results provide evidence that tight regulation of OBF1 expression in early B cells is essential to allow efficient B lymphocyte differentiation
Collective and broken pair states of 65,67Ga
Excited states of 65Ga and 67Ga nuclei were populated through the 12C(58Ni,αp) and 12C(58Ni,3p) reactions, respectively, and investigated by in-beam γ-ray spectroscopic methods. The NORDBALL array equipped with a charged particle ball and 11 neutron detectors was used to detect the evaporated particles and γ rays. The level schemes of 65,67Ga were constructed on the basis of γγ-coincidence relations up to 8.6 and 10 MeV excitation energy, and Iπ=27/2 and 33/2+ spin and parity, respectively. The structure of 65,67Ga nuclei was described in the interacting boson-fermion plus broken pair model, including quasiproton, quasiproton-two-quasineutron, and three-quasiproton fermion configurations in the boson-fermion basis states. Most of the states were assigned to quasiparticle + phonon and three quasiparticle configurations on the basis of their electromagnetic decay properties
Exploring the Support Needs of Family Caregivers of Patients with Brain Cancer Using the CSNAT: A Comparative Study with Other Cancer Groups
A substantial burden is placed on family caregivers of patients diagnosed with brain cancers. Despite this, the support needs of the caregivers are often under-recognised and not addressed adequately in current routine and patient centred clinical care. The Care Support Needs Assessment Tool (CSNAT) is a validated instrument designed to systematically identify and address caregiver needs. It has been trialled in an Australian palliative care community setting using a stepped wedge cluster design involving 322 family carers of terminally ill patients. The current article reports on a subset from this trial, 29 caregivers of patients with primary brain cancer, and compares their profile and outcomes to those of other cancer groups. Caregiver strain was assessed using the Family Appraisal of Caregiving Questionnaire, caregiver physical and mental wellbeing using SF12 and caregiver workload using a questionnaire on support with activities of daily living (ADL). In comparison to caregivers of patients with all other cancers, the primary brain cancer group had significantly higher levels of caregiver strain, lower levels of mental wellbeing and a higher level of ADL workload. Their physical wellness also deteriorated significantly over time.An action plan approach led to practical solutions for addressing highlighted concerns. Four themes evolved from the family caregivers’ feedback interviews: The extremely challenging caregiver experience with brain cancer; the systematic and practical approach of the CSNAT during rapid changes; connection with health professionals, feeling acknowledged and empowered; and timely advice and assurance of support during the caregiving journey. This preliminary study has demonstrated that the CSNAT provides a practical and useful tool for assessing the support needs of family caregivers of patients with brain cancer and has provided the basis for a larger scale, longitudinal study that allows a more detailed characterisation of the evolving caregiver needs at different stages of the disease
Silencing and Nuclear Repositioning of the λ5 Gene Locus at the Pre-B Cell Stage Requires Aiolos and OBF-1
The chromatin regulator Aiolos and the transcriptional coactivator OBF-1 have been implicated in regulating aspects of B cell maturation and activation. Mice lacking either of these factors have a largely normal early B cell development. However, when both factors are eliminated simultaneously a block is uncovered at the transition between pre-B and immature B cells, indicating that these proteins exert a critical function in developing B lymphocytes. In mice deficient for Aiolos and OBF-1, the numbers of immature B cells are reduced, small pre-BII cells are increased and a significant impairment in immunoglobulin light chain DNA rearrangement is observed. We identified genes whose expression is deregulated in the pre-B cell compartment of these mice. In particular, we found that components of the pre-BCR, such as the surrogate light chain genes λ5 and VpreB, fail to be efficiently silenced in double-mutant mice. Strikingly, developmentally regulated nuclear repositioning of the λ5 gene is impaired in pre-B cells lacking OBF-1 and Aiolos. These studies uncover a novel role for OBF-1 and Aiolos in controlling the transcription and nuclear organization of genes involved in pre-BCR function

Temporal variation in vegetative development of Caulerpa scalpelliformis (Chlorophyta) from Baleia beach, Ilha Grande bay (Rio de janeiro, brazil)
Caulerpa scalpelliformis grows on rocky and sandy bottoms at different depths in Jacuacanga Cove, Ilha Grande Bay, where it can form dense patches. The invasive behavior of this and other species of Caulerpa is well documented in the literature. This study analyzed the variation of the vegetative development of C. scalpelliformis from Baleia Beach (23º01'63''S and 44º14'18''W) in Jacuacanga Cove, from June 2003 to September 2004, including plants from rocky and sandy substrates. Morphometric and dry weight data from the erect and prostrate portions were used. Plants were collected from 20 x 20 cm² plots (n=3), randomly positioned on the rocky boulders (≅1.5 m depth) and on contiguous sandy bottom (≅3.0 m depth). During the study period, C. scalpelliformis occurred on both substrates as a pseudo-perennial species, showing partial loss of the erect portions from winter to spring; the height and dry weight of the erect portions increased from summer to autumn on both substrates. The temporal variation of C. scalpelliformis vegetative development in Baleia Beach was similar to that described for the invasive C. racemosa and C. taxifolia from different geographical regions of the world.Caulerpa scalpelliformis cresce sobre substrato rochoso e arenoso a diferentes profundidades na Enseada de Jacuacanga, Baía da Ilha Grande, onde pode formar bancos densos. O comportamento invasor desta e de outras espécies de Caulerpa é bem documentado na literatura. Este estudo analisou a variação do desenvolvimento vegetativo de C. scalpelliformis da Praia da Baleia (23º01'63''S e 44º14'18''W), Enseada de Jacuacanga, de junho de 2003 a setembro de 2004, considerando plantas do substrato rochoso e do fundo de areia. Dados morfométricos e de peso seco das porções prostrada e ereta foram usados. Plantas foram coletadas em quadrados de 20 cm de lado (n=3), posicionados aleatoriamente sobre o substrato rochoso (profundidade ≅ 1,5 m) e sobre o substrato arenoso (profundidade ≅ 3,0 m). Durante o período de estudo, C. scalpelliformis ocorreu nos dois substratos como espécie pseudo-perene, mostrando perda parcial das porções eretas do talo do inverno para a primavera; altura e massa seca das porções eretas mostraram tendência de aumento do verão para o outono nos dois substratos. A variação temporal do desenvolvimento vegetativo de C. scalpelliformis na Praia da Baleia foi similar à descrita para C. racemosa e C. taxifolia, espécies invasoras em diferentes regiões do mundo
