Local models of Shimura varieties, I. Geometry and combinatorics
We survey the theory of local models of Shimura varieties. In particular, we
discuss their definition and illustrate it by examples. We give an overview of
the results on their geometry and combinatorics obtained in the last 15 years.
We also exhibit their connections to other classes of algebraic varieties such
as nilpotent orbit closures, affine Schubert varieties, quiver Grassmannians

The 27-day versus 13.5-day variations in the solar Lyman-alpha radiation and the radio wave absorption in the lower ionosphere over Europe
In order to clarify the question of solar periods in absorption, the pattern was studied of the solar Lyman-alpha radiation (the principal ionizing agent of the lower ionosphere) and of the radio wave absorption at five widely spaced places in Europe. When the solar Lyman-alpha flux variability is very well developed, then it dominates in the lower ionospheric variability. The most pronounced Lyman-alpha variation on time scale day-month is the solar rotation variation (about 27 days). When the Lyman-alpha variability is developed rather poorly, as it is typical for periods dominated by the 13.5 day variability, then the lower ionospheric variability appears to be dominated by variations of meteorological origin. DFN-02, Sumatriptan 10 mg Nasal Spray with Permeation Enhancer, for the Acute Treatment of Migraine: A Randomized, Double-Blind, Placebo-Controlled Study Assessing Functional Disability and Subject Satisfaction with Treatment.
BackgroundThe commercial formulation of sumatriptan nasal spray is an effective option for migraine patients requiring or preferring a non-oral route of drug administration, but its utility is limited by poor absorption and tolerability issues. DFN-02, a new formulation of sumatriptan 10 mg nasal spray, is co-formulated with a permeation enhancer that gives it pharmacokinetics comparable to subcutaneous sumatriptan. As reported previously, DFN-02 was significantly better than placebo on multiple efficacy endpoints at 2 h postdose, including pain freedom, absence of the most bothersome symptom, and pain relief, and its safety and tolerability profiles were excellent.ObjectiveThe objective of this study was to assess the efficacy of acute treatment of migraine with DFN-02, including its effect on migraine-related functional disability and patient satisfaction with treatment.MethodsThis was a multicenter, randomized, double-blind, placebo-controlled efficacy and safety study of DFN-02 in adults with episodic migraine. Functional disability and subject satisfaction with treatment were prespecified endpoints, assessed in real-time by subjects, using an electronic diary.ResultsIn total, 107 subjects were randomized. DFN-02 was significantly superior to placebo for the reduction in functional disability score from predose level at 2 h after treatment (- 1.2 vs. - 0.6, p < 0.001). Subjects treated with DFN-02 were also more likely to be satisfied or very satisfied than subjects treated with placebo at 2 h postdose (70.0% vs. 44.2%, p = 0.027). Using the Patient Perception of Migraine Questionnaire-Revised at 24 h postdose, DFN-02 mean scores were significantly superior to placebo for the subscales of efficacy (65.2 vs. 42.5, p = 0.016) and function (68.9 vs. 42.1, p = 0.001), and for total score (71.0 vs. 56.6, p = 0.016); global medication effectiveness (p = 0.027); and overall satisfaction (p = 0.019). Placebo was significantly better than DFN-02 on the tolerability subscale (94.8 vs. 88.5, p = 0.026). At 24 h postdose, subjects reported significantly higher satisfaction with DFN-02 compared with satisfaction reported pre-randomization regarding their usual migraine medication (p = 0.012).ConclusionDFN-02 was superior to placebo for the relief of migraine-related functional disability, and provided greater satisfaction than placebo or subjects' usual acute treatment.Trial registrationClinicalTrials.gov identifier: NCT02856802
Quantitative analysis of cell organization in the external region of the olive fruit
Definitions of the cells that constitute the exocarp or exterior tissue of fleshy fruits are often vague, sometimes providing contradictory descriptions of the epidermis plus none or varying numbers of underlying cell layers for the same species. This study uses a morphometric approach to investigate how cell dimensions, cell number, and their relation with genetically based fruit size differences can contribute to a characterization of tissue organization in the external fruit region, using the olive drupe as an example.We determined cell area, radial and tangential widths, and cell number in the epidermis and 20 subepidermal cell layers of mature fruits of four olive cultivars that range in fruit size. Variation of these measurements among cell layers and the implied cellular contributions to fruit expansion revealed two different subepidermal regions, but with constant widths and layer numbers for all cultivars: (1) the first four cell layers (1-4), which have similar behavior to the epidermis; and (2) the following five cell layers (5-9), which are more similar to the mesocarp. The results provide new insights about cell patterns in the external region of the olive fruit and suggest that layers 1-4 together with the epidermis may act as a multiseriate exocarp and layers 5-9 may act as an outer mesocarp. © 2012 by The University of Chicago. Networking Effects on Cooperation in Evolutionary Snowdrift Game
The effects of networking on the extent of cooperation emerging in a
competitive setting are studied. The evolutionary snowdrift game, which
represents a realistic alternative to the well-known Prisoner's Dilemma, is
studied in the Watts-Strogatz network that spans the regular, small-world, and
random networks through random re-wiring. Over a wide range of payoffs, a
re-wired network is found to suppress cooperation when compared with a
well-mixed or fully connected system. Two extinction payoffs, that characterize
the emergence of a homogeneous steady state, are identified. It is found that,
unlike in the Prisoner's Dilemma, the standard deviation of the degree
distribution is the dominant network property that governs the extinction

Multiple mechanisms determine ER network morphology during the cell cycle in Xenopus egg extracts
In metazoans the endoplasmic reticulum (ER) changes during the cell cycle, with the nuclear envelope (NE) disassembling and reassembling during mitosis and the peripheral ER undergoing extensive remodeling. Here we address how ER morphology is generated during the cell cycle using crude and fractionated Xenopus laevis egg extracts. We show that in interphase the ER is concentrated at the microtubule (MT)-organizing center by dynein and is spread by outward extension of ER tubules through their association with plus ends of growing MTs. Fusion of membranes into an ER network is dependent on the guanosine triphosphatase atlastin (ATL). NE assembly requires fusion by both ATL and ER-soluble N-ethyl-maleimide–sensitive factor adaptor protein receptors. In mitotic extracts, the ER converts into a network of sheets connected by ER tubules and loses most of its interactions with MTs. Together, these results indicate that fusion of ER membranes by ATL and interaction of ER with growing MT ends and dynein cooperate to generate distinct ER morphologies during the cell cycle
