Ethyl Glucuronide in Scalp and Non-head Hair: An Intra-individual Comparison
Aims: Analysis of ethyl glucuronide (EtG), a minor metabolite of ethanol, is a valid tool for the assessment of social and chronic excessive alcohol consumption. Standardized analysis of EtG is usually done in head hair. As head hair cannot always be provided, alternative hair matrices become more and more interesting. Therefore, a study was performed that compared the intra-individual EtG concentrations in scalp hair and non-head hair (chest, arm, leg and axillary hair). Methods: Hair samples were collected from 68 subjects undergoing an expert assessment for fitness to drive. Aqueous extracts of the hair matrix were cleaned by solid-phase extraction, using an Oasis MAX column. EtG was first derivatized with perfluoropentanoic anhydride and then quantified by GC-MS/MS in negative chemical ionization mode, using EtG-d5 as internal standard. Results: For categorizing drinking behaviour, the two EtG cut-off values recommended by the Society of Hair Testing were applied for all different hair types. For chest, arm and leg hair, correct classification ratios were >83%. This corresponds to sensitivity values >78% and specificities >75%. Such values indicate together with φ coefficients (rφ) > 0.7 a high correlation of the categorization of the drinking behaviour based on these body hair EtG concentrations compared with the indexing based on scalp hair EtG-values. However, it must be taken into consideration that the time frame represented by non-head hair may extend way back. Conclusions: These results indicate that chest, arm and leg hair can be a valid alternative to assess the drinking behaviour of a subject if head hair is not available; whereas axillary hair is not suitable as alternative matri
Automating Vehicles by Deep Reinforcement Learning using Task Separation with Hill Climbing
Within the context of autonomous driving a model-based reinforcement learning
algorithm is proposed for the design of neural network-parameterized
controllers. Classical model-based control methods, which include sampling- and
lattice-based algorithms and model predictive control, suffer from the
trade-off between model complexity and computational burden required for the
online solution of expensive optimization or search problems at every short
sampling time. To circumvent this trade-off, a 2-step procedure is motivated:
first learning of a controller during offline training based on an arbitrarily
complicated mathematical system model, before online fast feedforward
evaluation of the trained controller. The contribution of this paper is the
proposition of a simple gradient-free and model-based algorithm for deep
reinforcement learning using task separation with hill climbing (TSHC). In
particular, (i) simultaneous training on separate deterministic tasks with the
purpose of encoding many motion primitives in a neural network, and (ii) the
employment of maximally sparse rewards in combination with virtual velocity

Filling a blank on the map: 60 years of fisheries in Equatorial Guinea
Despite a scarcity of pertinent information, it has been possible to reconstruct time series of marine fisheries catches for Equatorial Guinea from 1950 to 2010 using per capita fish consumption and population numbers for small-scale fisheries, catch rates and number of vessels for industrial fisheries and discard rates to estimate the discarded bycatch. Small-scale fisheries, industrial large-scale fisheries, domestic and legal and illegal foreign fisheries and their discards are all included. Total catches were estimated at 2.7 million tonnes over the time period considered, of which 653 000 t were caught domestically compared to 187 000 t reported by FAO. This shows that fisheries have more importance for Equatorial Guinea's food security than the official data suggest. In contrast to what is suggested by official figures, fisheries were shown to be strongly impacted by civil and political unrest; notably, they declined overall because of civil and political conflicts, socio-demographic dynamics, and a growing role of the newly discovered oil resources, which directly and indirectly threaten the food security of the people of Equatorial Guinea

MontanAqua : Wasserbewirtschaftung in Zeiten von Knappheit und globalem Wandel. Wasserbewirtschaftungsoptionen für die Region Crans-Montana-Sierre im Wallis
Das nationale Forschungsprogramm NFP 61 «Nachhaltige Wassernutzung » des Schweizerischen Nationalfonds hat sich zum Ziel gesetzt, wissenschaftliche Grundlagen zur nachhaltigen Wasserbewirtschaftung in der Schweiz zu liefern. Als Teil dieses Forschungsvorhabens wurde im Rahmen des Projektes MontanAqua die Wasserbewirtschaftung der Region Crans-Montana-Sierre (Wallis) untersucht. Es ging dabei darum, in enger Zusammenarbeit mit den in der Region betroffenen Akteuren nachhaltige Wassernutzungsstrategien für die Zukunft zu entwickeln. MontanAqua hat sich vertieft mit den bestehenden Systemen der Wasserbewirtschaftung auf der regionalen Skala (11 Gemeinden) auseinandergesetzt. Dazu wurden die zukünftigen Auswirkungen der klimatischen und sozioökonomischen Veränderungen einbezogen. Das Forschungsteam analysierte die aktuelle Situation anhand von quantitativen, qualitativen sowie kartografischen Methoden und kombinierte diese mit Modellberechnungen. Für die Modellierung der Zukunft wurden regionale Klimaszenarien und vier mit lokalen Akteuren entwickelte sozioökonomische Szenarien verwendet. Dieser Überblick fasst die Resultate des Projektes MontanAqua zusammen. Fünf wesentliche Fragen werden beantwortet und fünf Kernbotschaften erläutert. Zudem sind Empfehlungen für die Verantwortlichen der regionalen und kantonalen Wasserbewirtschaftung formuliert

Constitutively elevated levels of SOCS1 suppress innate responses in DF-1 immortalised chicken fibroblast cells.
The spontaneously immortalised DF-1 cell line is rapidly replacing its progenitor primary chicken embryo fibroblasts (CEFs) for studies on avian viruses such as avian influenza but no comprehensive study has as yet been reported comparing their innate immunity phenotypes. We conducted microarray analyses of DF-1 and CEFs, under both normal and stimulated conditions using chicken interferon-α (chIFN-α) and the attenuated infectious bursal disease virus vaccine strain PBG98. We found that DF-1 have an attenuated innate response compared to CEFs. Basal expression levels of Suppressor of Cytokine Signalling 1 (chSOCS1), a negative regulator of cytokine signalling in mammals, are 16-fold higher in DF-1 than in CEFs. The chSOCS1 “SOCS box” domain (which in mammals, interacts with an E3 ubiquitin ligase complex) is not essential for the inhibition of cytokine-induced JAK/STAT signalling activation in DF-1. Overexpression of SOCS1 in chIFN-α-stimulated DF-1 led to a relative decrease in expression of interferon-stimulated genes (ISGs; MX1 and IFIT5) and increased viral yield in response to PBG98 infection. Conversely, knockdown of SOCS1 enhanced induction of ISGs and reduced viral yield in chIFN-α-stimulated DF-1. Consequently, SOCS1 reduces induction of the IFN signalling pathway in chicken cells and can potentiate virus replication
Highly pathogenic avian influenza virus infection in chickens but not ducks is associated with elevated host immune and pro-inflammatory responses
Highly pathogenic avian influenza (HPAI) H5N1 viruses cause severe infection in chickens at near complete mortality, but corresponding infection in ducks is typically mild or asymptomatic. To understand the underlying molecular differences in host response, primary chicken and duck lung cells, infected with two HPAI H5N1 viruses and a low pathogenicity avian influenza (LPAI) H2N3 virus, were subjected to RNA expression profiling. Chicken cells but not duck cells showed highly elevated immune and pro-inflammatory responses following HPAI virus infection. HPAI H5N1 virus challenge studies in chickens and ducks corroborated the in vitro findings. To try to determine the underlying mechanisms, we investigated the role of signal transducer and activator of transcription-3 (STAT-3) in mediating pro-inflammatory response to HPAIV infection in chicken and duck cells. We found that STAT-3 expression was down-regulated in chickens but was up-regulated or unaffected in ducks in vitro and in vivo following H5N1 virus infection. Low basal STAT-3 expression in chicken cells was completely inhibited by H5N1 virus infection. By contrast, constitutively active STAT-3 detected in duck cells was unaffected by H5N1 virus infection. Transient constitutively-active STAT-3 transfection in chicken cells significantly reduced pro-inflammatory response to H5N1 virus infection; on the other hand, chemical inhibition of STAT-3 activation in duck cells increased pro-inflammatory gene expression following H5N1 virus infection. Collectively, we propose that elevated pro-inflammatory response in chickens is a major pathogenicity factor of HPAI H5N1 virus infection, mediated in part by the inhibition of STAT-3

Spliced Leader Trapping Reveals Widespread Alternative Splicing Patterns in the Highly Dynamic Transcriptome of Trypanosoma brucei
Trans-splicing of leader sequences onto the 5′ends of mRNAs is a widespread phenomenon in protozoa, nematodes and some chordates. Using parallel sequencing we have developed a method to simultaneously map 5′splice sites and analyze the corresponding gene expression profile, that we term spliced leader trapping (SLT). The method can be applied to any organism with a sequenced genome and trans-splicing of a conserved leader sequence. We analyzed the expression profiles and splicing patterns of bloodstream and insect forms of the parasite Trypanosoma brucei. We detected the 5′ splice sites of 85% of the annotated protein-coding genes and, contrary to previous reports, found up to 40% of transcripts to be differentially expressed. Furthermore, we discovered more than 2500 alternative splicing events, many of which appear to be stage-regulated. Based on our findings we hypothesize that alternatively spliced transcripts present a new means of regulating gene expression and could potentially contribute to protein diversity in the parasite.
