    Generally failures of different aircraft components and parts are revealed and examined by the use of non-destructive examination methods. In further detailed explanation and interpretation of failures optical and scanning electron microscopy are used. This paper deals with a problem of a crack on aircraft wheel rim made from aluminium alloy 2014-T6.The crack was observed during regular control by the maintenance unit for non-destructive examination of the Slovenian air carrier Adria Airways. The crack on the rim of an aircraft wheel investigated was a typical fatigue crack. At same time a numerous pits were found which served as stress concentrations on the rim surface

At same time a numerous pits were found which served as stress concentrations on the rim surface.Oštećenja različitih dijelova i komponenti aviona otkrivena su i ispitivana primjenom nedestruktivnih metoda ispitivanja. Za detaljna objašnjavanja i interpretaciju oštećenja korištene su metode optičke i pretražne elektronske mikroskopije. Ovaj se članak bavi problemom pukotine na naplatku avionskog kotača izrađenog od aluminijeve slitine 2014-T6. Pukotina je zapažena za vrijeme redovite kontrole od strane slovenskog zračnog prijevoznika Adria Airways. Ispitivanja su pokazala, da je zapažena pukotina na naplatku avionskog kotača bila tipična umorna pukotina. Također je utvrđeno, da brojna oštećenja u obliku rupica pronađena na površini naplatka djeluju kao koncentratori naprezanja

    Strengthening the host immune system to fully exploit its potential as antimicrobial defense is vital in countering antibiotic resistance. Chemical compounds released during bidirectional host–pathogen cross-talk, which follows a sensing-response paradigm, can serve as protective mediators. A potent, diffusible messenger is hydrogen peroxide (H(2)O(2)), but its consequences on extracellular pathogens are unknown. Here we show that H(2)O(2), released by the host on pathogen contact, subverts the tyrosine signaling network of a number of bacteria accustomed to low-oxygen environments. This defense mechanism uses heme-containing bacterial enzymes with peroxidase-like activity to facilitate phosphotyrosine (p-Tyr) oxidation. An intrabacterial reaction converts p-Tyr to protein-bound dopa (PB-DOPA) via a tyrosinyl radical intermediate, thereby altering antioxidant defense and inactivating enzymes involved in polysaccharide biosynthesis and metabolism. Disruption of bacterial signaling by DOPA modification reveals an infection containment strategy that weakens bacterial fitness and could be a blueprint for antivirulence approaches

    Cys-Ph-TAHA: a lanthanide binding tag for RDC and PCS enhanced protein NMR

    Here we present Cys-Ph-TAHA, a new nonadentate lanthanide tag for the paramagnetic labelling of proteins. The tag can be easily synthesized and is stereochemically homogenous over a wide range of temperatures, yielding NMR spectra with a single set of peaks. Bound to ubiquitin, it induced large residual dipolar couplings and pseudocontact shifts that could be measured easily and agreed very well with the protein structure. We show that Cys-Ph-TAHA can be used to label large proteins that are biochemically challenging such as the Lac repressor in a 90 kDa ternary complex with DNA and inducer

    Spherical crystallization of drugs is the process of obtaining larger particles by agglomeration during crystallization. The most common techniques used to obtain such particles are spherical agglomeration and quasi-emulsion solvent diffusion. Ammonia diffusion systems and crystallo-co-agglomeration are extensions of these techniques. By controlling process parameters during crystallization, such as temperature, stirring rate, type and amount of solvents, or excipient selection, it is possible to control the formation of agglomerates and obtain spherical particles of the desired size, porosity, or hardness. Researchers have reported that the particles produced have improved micromeritic, physical, and mechanical properties, which make them suitable for direct compression. In some cases, when additional excipients are incorporated during spherical crystallization, biopharmaceutical parameters including the bioavailability of drugs can also be tailored.Sferična kristalizacija je postopek izdelave večjih delcev z aglomeracijo manjših med samo kristalizacijo. Najpogosteje uporabljeni tehniki za izdelavo takšnih delcev sta sferična aglomeracija in kvaziemulzija z difuzijo topila. Sistem z difuzijo amoniaka in kristalo-ko-aglomeracija sta razširitvi teh dveh metod. Z nadzorovanjem procesnih parametrov med kristalizacijo, kot sta temperatura in hitrost mešanja, z izbiro lastnosti in množine topil ter z izbiro pomožnih snovi, lahko vplivamo na nastanek aglomeratov in izdelamo sferične delce želenih velikosti, primerne poroznosti ali trdote. Raziskovalci poročajo, da imajo izdelani delci izboljšane pretočne lastnosti, izboljšane druge fizikalne in mehanske lastnosti zaradi česar so primerni za direktno tabletiranje. V nekaterih primerih lahko ob vgradnji ustreznih pomožnih snovi, ki jih dodamo med procesom sferične kristalizacije, izboljšamo tudi biofarmacevtske lastnosti zdravilnih učinkovin vključno s povečanjem biološke uporabnosti

    Nanocomposite foams from iron oxide stabilized dicyclopentadiene high internal phase emulsions

    Nanocomposite polyHIPE foams with open-cellular morphology were obtained using nanoparticles (γFe2O3/Fe3O4γFe_2O_3/Fe_3O_4), surfactant (Pluronic L121) or nanoparticle/surfactant stabilized dicyclopentadiene high internal phase emulsions (DCPD HIPEs). Upon curing, cavity sizes were found to vary drastically between 950 ± 360 µm down to 7 ±3 µm de- pending on the HIPE formulations. As-obtained nanocomposite polyHIPE foams were functionalized using elemental bromine in THF. Upon bromination the nanoparticles are moved from the cavities surfaces into the bulk phase of the polymer scaffold, which affects the inductive-heating capability of the magnetic nanocomposite foams decreasing it by the factor of 2.V tem strokovnem članku opisujem pripravo makroporoznih nanokompozitnih pen iz diciklopentadienskih HIP emulzij stabiliziranimi z γFe2O3/Fe3O4γFe_2O_3/Fe_3O_4 nanodelci, t.i. Pickering emulzij in HIP emulzij stabiliziranimi s kombinacijo surfaktanta in nanodelcev. Tako pripravljene nanokompozitne pene smo primerjali s penami pripravljenimi iz HIP emulzij stabiliziranimi le s surfaktanti in ugotovili da se premer por spreminja in sicer od 7 ± 3 m do 950 ± 360 µm. V nadaljevanju smo nanokompozitne makroporzne pene funkcionalizirali z elementarnim bromom, ter raziskali vpliv bromiranja na samo lokacijo nanodelcev in sposobnost γFe2O3/Fe3O4γFe_2O_3/Fe_3O_4 nanodelcev da ob prisotnosti zunanjega alternirajočega magnetnega polja proizvajajo toploto. V ta namen smo bromirano nanokompozitno peno izpostavili zunanjemu izmeničnemu magnetnem polju, ter ugotovili da se sposobnost segrevanje zmanjša za faktor 2

    Morphokinetic Characteristics and Developmental Potential of In Vitro Cultured Embryos from Natural Cycles in Patients with Poor Ovarian Response

    Background. Patients with poor ovarian response to ovarian hyperstimulation represent an interesting group for studying the impact of embryo cleavage irregularities on clinical outcome since all embryos, regardless of their quality, are usually transferred to the uterus. The aim of our study was to follow the morphokinetics of fertilized oocytes from natural cycles in poor responders. Methods. Zygotes from 53 cycles were cultured in vitro for 3 days. The morphokinetics of their development and transfer outcomes were retrospectively analyzed for the normally and irregularly cleaved embryos. Results. Of all embryos, 30.2% had single and 20.8% multiple cleavage irregularities with the following prevalence: developmental arrest 30.2%, direct cleavage to more than two cells 24.5%, chaotic cleavage 13.2%, and reverse cleavage 11.3%. These embryos had longer pronuclear phases, first cytokinesis, second embryo cell cycles, and less synchronized divisions. The transfer of normally developing embryos resulted in an implantation rate of 30.8% and a delivery rate of 23.1%, but irregularly cleaved embryos did not implant. Conclusions. The use of time-lapse microscopy in poor responder patients identified embryos with cleavage abnormalities that are related with no or extremely low implantation potential. Gained information about embryo quality is important for counselling patients about their expectations
