Synthesis and expression of the Erythrina trypsin/tissue plasminogen activator (tPA) inhibitor encoding-gene : genetic dissection to correlate the interaction of Erythrina and Soybean trypsin inhibitors with tPA

Bibliography: pages 229-241.A trypsin inhibitor had previously been isolated from Erythrina caffra, a member of the Leguminosae family. The inhibitor, Erythrina trypsin inhibitor (ETI), is unique among plantderived inhibitors, in that in addition to trypsin, it inhibits chymotrypsin and tissue plasminogen activator (tPA). ETI was previously sequenced and its crystal structure determined from which it could be seen that ETI has good homology to soybean trypsin inhibitor (STI). However, STI does not inhibit tPA. From the three-dimensional structure of ETI it was known that the amino-terminus of the molecule forms a finger-like structure stabilized by hydrogen bonds and hydrophobic interactions. In addition, the N-terminal finger region is located in close proximity to the reactive site loop and the Nterminal residue (Val) is bound up in the finger region. In STI the N-terminal region is located in close proximity to the reactive site loop and is folded into a structure similar to that in ETI. While the crystal structure of STI was not detailed enough to determine secondary interactions such as hydrogen bonds it was hypothesized that the N-terminal region is stabilized as in ETI. It was further hypothesized that the N-terminal residue of STI (Asp), because of its hydrophilic nature, is not involved in the structured N-terminal finger region of this protein. This leaves this Asp residue of STI free to form an ion pair with Lys at position 60 in trypsin when STI and trypsin interact

B-spline Parameterization Based Flight Trajectory Optimization

Achieving the first heavier-than-air flight during the 20th century was certainly a great landmark in human history. However, flight itself generally is not the final objective. To perform the desired mission effectively, it is necessary to determine the path to follow according to the objective, for instance minimizing the elapsed time, the path length, etc. How do we deal with such problems? Which are the techniques to solve them? Trajectory Optimization is the subject that deals with such problems, and the object of study of the present dissertation. I propose to study the use of B-spline Parameterization for Flight Trajectory Optimization. A Python code was developed to implement the proposed method, revealing promising results for all three presented examples, proving the robustness and versatility of the proposed method. Future studies should explore different types of missions, including spatial trajectories, and various sets of waypoints. The study and implementation of the control to stimulate the desired flying device to follow the determined optimal trajectory would also constitute an interesting exercise.Materializar o primeiro voo de um dispositivo mais pesado do que o ar durante o século XX constituiu certamente um grande marco na história da humanidade. Contudo, o voo em si raramente constitui o objetivo final. Para se executar uma missão de forma efetiva, é necessário determinar a trajetória a ser seguida de acordo com o objetivo da missão, por exemplo minimizando o tempo decorrido, o comprimento da trajetória, etc. Como se lida com tais problemas? Quais são as técnicas adequadas para resolvê-las? A Otimização de Trajetórias é a disciplina que lida com estes tipos de problemas, e também o objeto de estudo desta dissertação. Proponho o uso da Parametrização B-spline para a Otimização de Trajetórias de Voo. Foi desenvolvido um código em Python que implementa o método proposto, tendo sido obtido resultados promissores para os três exemplos estudados, provando assim a robustez e a versatilidade do método proposto. Trabalhos futuros devem explorar diferentes tipos de missões, incluindo trajetórias espaciais, assim como diversas combinações de waypoints. O estudo e a implementação dos sinais de controlo que façam com que o veículo desejado siga a trajetória ótima determinada deverá constituir também um exercício interessante

Trabalho projetoThis project was elaborated with the intention to establish a communication plan for Century 21 Portugal, with the intention of promoting the foreign investment in the Portuguese real estate market. After analyzing the Portuguese market and the current strategy of Century 21 Portugal, it was found some opportunities that a brand, like Century 21 , can take advantage. The main idea of this project is to create a new communication plan and a new type of business for the brand Century 21 Portugal, using certain communication tools and techniques that will help the company to focus on critical factors and to reach their potential customers overseas. The objective of this "new direction" for the brand is to provide a better service to the property owners offering new possibilities of business, outside of Portugal. In long term, this "new direction" can be extended to all markets, with more people getting their second, or even the first home in other countries. With this project Century 21 can create just "one big and global real estate market", approaching all the markets, the owners of the properties and potential buyers from around the world.Este projeto foi elaborado com a intenção de criar um plano de comunicação para Century 21 Portugal, com o principal objectivo de promover o investimento estrangeiro no mercado imobiliário Português. Depois de analisar o mercado Português e a estratégia atual da Century 21 Portugal, verificou-se algumas oportunidades que uma marca, como Century 21, pode aproveitar neste mercado. A idéia principal deste projeto é a criação de um novo plano de comunicação e um novo tipo de negócio para a marca Century 21 Portugal, usando algumas ferramentas e técnicas de comunicação que ajudarão a empresa a concentrar-se em fatores críticos e alcançar potenciais clientes no estrangeiro. O objetivo desta "nova visão" para a marca é proporcionar um melhor serviços aos proprietários de imóveis oferecendo novas possibilidades de negócio, fora de Portugal. Como a música pode auxiliar no ensino de física

This study, whose theme is “How music can auxiliary in Physics teaching”, seeks to analyze how Physics is explored in the classroom by students and understand that Physics goes far beyond just the application of formulas. The work aims to understand how would be the inclusion of music in the classroom to auxiliary in the teaching of Physics, along with the use of the CAVAQUINHO as a display instrument. The proposed methodology is a case study, wich, after a review of the literature on the study of the use of music in teaching Physics, will be discussed the development of application of a questionnaire and the demonstration of a simple class with the theme “Physics and Music”, using the CAVAQUINHO as a teaching mediator instrument, in which 2 nd year high school students participated, in which students were able to analyze the quality of the class applied and the way in which the content was exposed. It was concluded that music can facilitate the teaching of Physics in light of the research carried out and how the inclusion of an instrument, whether musical or not, brings positive points to learning, drawing the student’s attention and bringing them to reality.Este estudo, cujo tema é “Como a música pode auxiliar no ensino de Física”, busca analisar como a Física é vista em sala de aula pelos alunos e compreender que a Física vai muito além do que apenas a aplicação de fórmulas. O trabalho tem como objetivo entender como seria a inclusão da música em sala de aula para auxiliar no ensino de física, junto com a utilização do cavaquinho como instrumento expositor de ensino. A metodologia proposta é um estudo de caso, após uma revisão da literatura sobre o estudo do uso da música no ensino de Física, será debatido o desenvolvimento da aplicação de um questionário e a demonstração de uma aula simples com o tema Física e Música, utilizando o cavaquinho como instrumento mediador de ensino, onde foram utilizadas turmas de 2o ano do Ensino Médio, a qual os alunos puderam analisar a qualidade da aula aplicada e a forma como foi exposto o conteúdo. Concluímos que a música pode facilitar o ensino de física diante das pesquisas realizadas e como a inclusão de um instrumento seja musical ou não, traz pontos positivos para a aprendizagem, chamando a atenção dos alunos e trazendo-os para realidade
