    Alcohol consumption among adult males in urban area of Thanlyin Township, Yangon Region, Myanmar

    Background: Alcohol consumption is a major cause of morbidity and mortality worldwide. It is frequently related to health and behavioural problems as well as socio-economic hardship. Therefore, this study was conducted to determine the prevalence and risk factors of alcohol consumption among adult males residing in urban area of Thanlyin Township, Yangon Region.Methods: A cross-sectional study was conducted among 380 adult males. Multi-stage random sampling was applied. Data entry and analysis was done using Stata 11.0 statistical package.Results: The prevalence of current alcohol drinking, ex-drinking and never drinking were 20.5%, 9.0% and 70.5%, respectively. There was a significant decreasing trend of alcohol consumption across the levels of age-group. Age, education status and practicing other health-risk behaviours such as smoking and betel chewing were detected as significant risk factors of alcohol consumption. Ever smokers and ever betel chewers were about 4 times more likely to be ever alcohol user compared to their counterparts even if age and education level were adjusted. By controlling smoking and betel chewing habits, 79.2% and 76.6% of existing prevalence of alcohol consumption among respondents would be reduced, respectively.Conclusions: There is an urgent need to curb the habit of alcohol consumption among adult males living in urban area, especially young adults. Alcohol and tobacco control policies in Myanmar should be strengthened or reinforced. Tobacco control program also needs to be intensified. Health education and health promotion activities should be enhanced in order to reduce alcohol consumption in the country.

    Klotho-beta overexpression as a novel target for suppressing proliferation and fibroblast growth factor receptor-4 signaling in hepatocellular carcinoma

    <p>Abstract</p> <p>Background</p> <p>We had previously demonstrated overexpression of fibroblast growth factor receptor-4 (FGFR4) in hepatocellular carcinoma (HCC). However, additional molecular mechanisms resulting in amplified FGFR4 signaling in HCC remain under-studied. Here, we studied the mechanistic role of its co-receptor klotho-beta (KLB) in driving elevated FGFR4 activity in HCC progression.</p> <p>Results</p> <p>Quantitative real-time PCR analysis identified frequent elevation of KLB gene expression in HCC tumors relative to matched non-tumor tissue, with a more than two-fold increase correlating with development of multiple tumors in patients. KLB-silencing in Huh7 cells decreased cell proliferation and suppressed FGFR4 downstream signaling. While transient repression of KLB-FGFR4 signaling decreased protein expression of alpha-fetoprotein (AFP), a HCC diagnostic marker, prolonged inhibition enriched for resistant HCC cells exhibiting increased liver stemness.</p> <p>Conclusions</p> <p>Elevated KLB expression in HCC tissues provides further credence to the oncogenic role of increased FGFR4 signaling in HCC progression and represents a novel biomarker to identify additional patients amenable to anti-FGFR4 therapy. The restricted tissue expression profile of KLB, together with the anti-proliferative effect observed with KLB-silencing, also qualifies it as a specific and potent therapeutic target for HCC patients. The enrichment of a liver stem cell-like population in response to extended KLB-FGFR4 repression necessitates further investigation to target the development of drug resistance.</p

    Clinical importance of the Mandalay spitting cobra (Naja mandalayensis) in Upper Myanmar – Bites, envenoming and ophthalmia

We confirm and extend the known distributions and habitats of both N. mandalayensis and N. kaouthia in Upper Myanmar. Clinical symptoms of local and systemic envenoming by N. mandalayensis are described for the first time. These included local swelling, blistering and necrosis and life-threatening systemic neurotoxicity. More information is needed about the clinical phenotype and management of bites by N. mandalayensis, the commoner of the two cobras in Upper Myanmar. Since the current cobra antivenom manufactured in Myanmar has lower pre-clinical efficacy against N. mandalayensis than N. kaouthia, there is a need for more specific antivenom therapy.Published versio

    Bioactive compounds screening, antimicrobial activities of leave extract from two palatable plants: Piper betle and Murraya koenigii (Curry leaves)

    Introduction: Piper betle Linn is one of the most commonly used compounding plants for ethno-medical purposes, with its extract generally used in modern products to enrich their functional efficacy. The extraction methods always lead to differences in the antimicrobial efficacy of methanol extracts of bioactive compounds. Purpose: The study was conducted to screen for bioactive compounds and determine their antimicrobial efficacy in a methanol extract of Piper betle and Murraya koenigi leaves from five different regions. Methodology: The phytochemical screening done according to the procedure that is implied in from Patil, et.al, with minor modifications by the researchers of the current study. Antimicrobial activity was determined; efficacy was measured by disc diffusion analysis. Results: Phytochemical screening revealed the presence of saponin, tannin, terpenoids, alkaloids and flavonoids in the extracts. The methanolic extract of betel leaves from all the selected regions except from Bau exhibited antimicrobial activities. Among them, extract from Kuching and Simunjan have no effect on E. coli. The methanolic extract of curry leaves from Kuching, Balai ringai and Bau have antimicrobial activities against Staph aureus and those from Balai ringai is also active against E.coli. Conclusion: Although there were previous reports of phytochemical screening and antimicrobial activities from the extract of these plants, there were still lack of research conducted on the specimens especially from our local community (Sarawak). The outcome of this study will help us to identify the bioactive compounds of the local samples and give us some pictures of their activities on how to ensure these plants can be brought forward based on the origin of the sample.     Co-occurrence of Point Mutations in the Voltage-Gated Sodium Channel of Pyrethroid-Resistant Aedes aegypti Populations in Myanmar

    Background:Single amino acid substitutions in the voltage-gated sodium channel associated with pyrethroid resistance constitute one of the main causative factors of knockdown resistance in insects. The kdr gene has been observed in several mosquito species; however, point mutations in the para gene of Aedes aegypti populations in Myanmar have not been fully characterized. The aim of the present study was to determine the types and frequencies of mutations in the para gene of Aedes aegypti collected from used tires in Yangon City, Myanmar.Methodology/Principal Findings:We determined high pyrethroid resistance in Aedes aegypti larvae at all collection sites in Yangon City, by using a simplified knockdown bioassay. We showed that V1016G and S989P mutations were widely distributed, with high frequencies (84.4% and 78.8%, respectively). By contrast, we were unable to detect I1011M (or I1011V) or L1014F mutations. F1534C mutations were also widely distributed, but with a lower frequency than the V1016G mutation (21.2%). High percentage of co-occurrence of the homozygous V1016G/S989P mutations was detected (65.7%). Additionally, co-occurrence of homozygous V1016G/F1534C mutations (2.9%) and homozygous V1016G/F1534C/S989P mutations (0.98%) were detected in the present study.Conclusions/Significance:Pyrethroid insecticides were first used for malaria control in 1992, and have since been constantly used in Myanmar. This intensive use may explain the strong selection pressure toward Aedes aegypti, because this mosquito is generally a domestic and endophagic species with a preference for indoor breeding. Extensive use of DDT for malaria control before the use of this chemical was banned may also explain the development of pyrethroid resistance in Aedes aegypti
