    Effect of hyperbaric oxygen on mesenchymal stem cells for lumbar fusion in vivo

    <p>Abstract</p> <p>Background</p> <p>Hyperbaric oxygen (HBO) therapy has been proved in improving bone healing, but its effects on mesenchymal stem cells (MSCs) <it>in vivo </it>is not clear. The aims of this study are to clarify whether the HBO therapy has the same enhancing effect on MSCs with regard to bone formation and maturation and to ascertain whether the transplanted MSCs survive in the grafted area and contribute to new bone formation.</p> <p>Methods</p> <p>Twenty-three adult rabbits underwent posterolateral fusion at L4-L5 level. The animals were divided into three groups according to the material implanted and subsequent treatment: (1) Alginate carrier (n = 6); (2) Alginate-MSCs composite (n = 11); and (3) Alginate-MSCs composite with HBO therapy (n = 6). After 12 weeks, spine fusion was examined using radiographic examination, manual testing, and histological examination. Using a PKH fluorescence labeling system, whether the transplanted MSCs survived and contributed to new bone formation in the grafted area after HBO therapy was also examined.</p> <p>Results</p> <p>The bilateral fusion areas in each animal were evaluated independently. By radiographic examination and manual palpation, union for the Alginate, Alginate-MSCs, and Alginate-MSCs-HBO groups was 0 of 12, 10 of 22, and 6 of 12 respectively. The difference between the Alginate-MSCs and Alginate-MSCs-HBO groups was not significant (P = 0.7997). The fluorescence microscopy histological analysis indicated that the transplanted PKH67-labeled MSCs survived and partly contributed to new bone formation in the grafted area.</p> <p>Conclusions</p> <p>This study demonstrated that the preconditioned MSCs could survive and yield bone formation in the grafted area. HBO therapy did not enhance the osteogenic ability of MSCs and improve the success of spine fusion in the rabbit model. Although there was no significant effect of HBO therapy on MSCs for spine fusion, the study encourages us to research a more basic approach for determining the optimal oxygen tension and pressure that are required to maintain and enhance the osteogenic ability of preconditioned MSCs. Further controlled <it>in vivo </it>and <it>in vitro </it>studies are required for achieving a better understanding of the effect of HBO treatment on MSCs.</p

    Elevated creatine kinase activity in primary hepatocellular carcinoma

    BACKGROUND: Inconsistent findings have been reported on the occurrence and relevance of creatine kinase (CK) isoenzymes in mammalian liver cells. Part of this confusion might be due to induction of CK expression during metabolic and energetic stress. METHODS: The specific activities and isoenzyme patterns of CK and adenylate kinase (AdK) were analysed in pathological liver tissue of patients undergoing orthotopic liver transplantation. RESULTS: The brain-type, cytosolic BB-CK isoenzyme was detected in all liver specimens analysed. Conversely, CK activity was strongly increased and a mitochondrial CK (Mi-CK) isoenzyme was detected only in tissue samples of two primary hepatocellular carcinomas (HCCs). CONCLUSION: The findings do not support significant expression of CK in normal liver and most liver pathologies. Instead, many of the previous misconceptions in this field can be explained by interference from AdK isoenzymes. Moreover, the data suggest a possible interplay between p53 mutations, HCC, CK expression, and the growth-inhibitory effects of cyclocreatine in HCC. These results, if confirmed, could provide important hints at improved therapies and cures for HCC

    Impaired Thymic Export and Apoptosis Contribute to Regulatory T-Cell Defects in Patients with Chronic Heart Failure

    Animal studies suggest that regulatory T (T(reg)) cells play a beneficial role in ventricular remodeling and our previous data have demonstrated defects of T(reg) cells in patients with chronic heart failure (CHF). However, the mechanisms behind T(reg-)cell defects remained unknown. We here sought to elucidate the mechanism of T(reg-)cell defects in CHF patients.We performed flow cytometry analysis and demonstrated reduced numbers of peripheral blood CD4(+)CD25(+)FOXP3(+)CD45RO(-)CD45RA(+) naĂŻve T(reg) (nT(reg)) cells and CD4(+)CD25(+)FOXP3(+)CD45RO(+)CD45RA(-) memory T(reg) (mT(reg)) cells in CHF patients as compared with non-CHF controls. Moreover, the nT(reg)/mT(reg) ratio (p<0.01), CD4(+)CD25(+)FOXP3(+)CD45RO(-) CD45RA(+)CD31(+) recent thymic emigrant T(reg) cell (RTE-T(reg)) frequency (p<0.01), and T-cell receptor excision circle levels in T(reg) cells (p<0.01) were lower in CHF patients than in non-CHF controls. Combined annexin-V and 7-AAD staining showed that peripheral T(reg) cells from CHF patients exhibited increased spontaneous apoptosis and were more prone to interleukin (IL)-2 deprivation- and CD95 ligand-mediated apoptosis than those from non-CHF individuals. Furthermore, analyses by both flow cytometry and real-time polymerase chain reaction showed that T(reg)-cell frequency in the mediastinal lymph nodes or Foxp3 expression in hearts of CHF patients was no higher than that of the non-CHF controls.Our data suggested that the T(reg)-cell defects of CHF patients were likely caused by decreased thymic output of nascent T(reg) cells and increased susceptibility to apoptosis in the periphery

    Effect of high root temperature and excessive insolation upon growth

    Summary. 1 1. Under ordinary environmental conditions of temperature and sunlight the growth of peas, as of barley, is seriously hindered by overcrowding, even when each plant receives a similar supply of food and water. Not only is less dry weight produced, but the pods become thin and distorted and fail to develop their seeds properly. 2 Growth tends to be depressed in hot sunny weather when no protection is afforded. The chief detrimental factors concerned appear to be high temperatures at the roots associated with strong and prolonged sunshine, though the two factors acting individually are much less potent for harm. Under these conditions crowding shelters the roots from overheating and the leaves from too much sunlight, and up to a certain point crowded plants make better growth than those spaced well apart. Overcrowding, however, still depresses growth, probably because the light and root temperature reductions are too great. 3 Provided insolation is not excessive the amount of daily fluctuation of root temperature over a total range of about 22oC. (6?7-28-9oC.) has comparatively little influence upon growth; high maxima and low minima give similar results to low maxima and relatively high minima, provided the average mean temperatures are not too dissimilar. 4 With high root temperatures a difference in the degree of insolation or in the angle of incidence of the sun's rays may have a considerable influence on growth, a slight easing off of the solar conditions enabling much better growth to be made. 5 With very strong sunshine reduction of high maximum root temperatures (from 29oC. upwards) allows of satisfactory growth, when unprotected plants are rapidly killed. The inhibitory action of too high temperatures at the roots is thus clearly shown. Nevertheless, the growth so made is less good than under more normal conditions of insolation, thus demonstrating the harmful action of too powerful sunlight, when all the root temperatures rule high. 6 Boot temperatures appear to be of greater importance than atmospheric temperatures, as good growth can be made in hot atmospheres provided the roots are kept relatively cool. 7 There is some reason to believe that the minima are of as much importance as the maxima, i.e. that plants can withstand very high maximum temperatures provided there is a considerable drop to the minima, but cannot put up with the constant conditions of heat induced by fairly high maxima, and high minim
