    A pharmacometrics model to define docetaxel target in early breast cancer

    Aims: We aimed to study the relation between pharmacokinetics (PK) and pharmacodynamics (PD) of docetaxel in early breast cancer and recommend a target exposure. Methods: A PK/PD study was performed in 27 early breast cancer patients treated with doxorubicin and cyclophosphamide for 4 cycles followed by 4 cycles of docetaxel 75–100 mg/m2 infused every 21 days. Individual Bayesian estimates of docetaxel PK parameters were obtained using a nonparametric population PK model developed with data from patients with metastatic breast cancer who received dose-intensified docetaxel (300–350 mg/m2). Docetaxel area under the curve (AUC) and maximum concentration (Cmax) in each cycle and total cumulative AUC (AUCcum) were calculated and related to the incidence of adverse effects and tumour recurrence. Results: Docetaxel clearance showed no change over the 4 treatment cycles, but a gradual increase in the volume of distribution was observed. One third of the patients had at least 1 dose reduction of docetaxel due to toxicity. The mean AUC, AUCcum and Cmax in patients showing docetaxel-associated adverse events were significantly higher than in patients free of toxicity (P 4.5 mg*h/L and 3.5 mg/L, respectively, were risk factors for docetaxel toxicity, while an AUC <4.5 mg*h/L was associated with tumour recurrence. Conclusion: We report for the first time a relation between docetaxel exposure and toxicity and recommend specific targets of drug exposure with implications for the clinical management of early breast cancer patients.Fil: Aldaz, Azucena.     Al encabezar este artículo con el título Orden y desorden en La Ciutat del Sol de Joan Brossa queremos ver aquellos aspectos dinámicos de la obra. En esta ocasión el peso de la reflexión recae sobre el carnaval y los símbolos dinámicos de cambio que aparecen reflejados en la citada obra. Es especialmente relevante la simbología usada por Brossa en esta obra, ya que la ponen en relación directa con los anhelos de ilustración propios de la Revolución francesa, en el contexto gris y represivo de la España franquista de los años inmediatamente posteriores a la Guerra Civil Española. El artículo incorpora una puesta en escena de la obra para hacerla representable con numerosas licencias interpretativas: Incorporación de un prólogo o presentación que ayude al espectador a situarse en la serie de acciones que va a presenciar y un epílogo que subraye la naturaleza inconformista del planteamiento brossiano. Descomposición de la acción en un doble foco, de manera que el aspecto temporal queda enfatizado por la presencia de la memoria y el proyecto futuro encarnados por un colectivo de abuelos con sus nietos.     Monitoring Extreme Impacts of Rugulopteryx okamurae (Dictyotales, Ochrophyta) in El Estrecho Natural Park (Biosphere Reserve). Showing Radical Changes in the Underwater Seascape

    The invasive macroalga Rugulopteryx okamurae represents an unprecedented case of bioinvasion by marine macroalgae facing the European coasts. Since the first apparition of the species in the Strait of Gibraltar in 2015, its fast dispersion along the introduced habitats constitutes a real challenge to develop monitoring strategies that ahead of its impacts. The present study uses three different approaches to address impacts on the benthic ecosystems, at the same time offers relevant data for future management actions in El Estrecho Natural Park (PNE). Information obtained by monitoring permanent sentinel stations revealed a significant loss in resident species coverage after the moment of maximum growth in 2017. Thus, despite coverage of R. okamurae did not strongly varied in the latter years, impacts generated remain high in the habitats studied. Estimations of the invasive species coverage by combining cartographic image analysis and in situ data predicted a major occupation (over 85% coverage) between 10 and 30 m, coinciding with the maximum rocky surface areas (m2) mapped on the PNE. Furthermore, a Citizen Science research collaboration evidenced impacts on the benthic seascape through an ad hoc exploration of images that allowed a “before” and “after” comparison of the invasion process in the same geographic locations. This has made it possible to graphically demonstrate severe changes in the underwater seascape and, therefore, the general impact of this new biological invasion. The spatial colonization estimations combined with the impacts reported by both scientific [Sessile Bioindicators in Permanent Quadrats (SBPQ) sentinel stations] and civilian (Citizen Science) monitoring methodologies claim the urgent development of further studies that allow the design of monitoring strategies against R. okamurae expansion across the Mediterranean and Atlantic waters

    Assessment of elemental composition in commercial fish of the Bay of Cádiz, Southern Iberian Peninsula

    The assessment of trace metal content in our fish diet is important due to the adverse effect on human health. Despite the increasing interest about the fish quality, little information is available for Southern Spain, a region characterized by high seafood intake. Nine species from the Bay of Cádiz with high commercial value were selected. Similar values were measured in the nine studied species for most of the elements, except for the macroelements Ca and S, and the microelements Fe, Mn and As, which showed significant differences among species. Metal Pollution Index (MPI) did not differ among species, and it was similar to those obtained for other Atlantic and Mediterranean locations. The values measured for the nine species were below the health limits provided by World, European and Spanish legislations, indicating that, in general terms, consumption of these species is safe in the study area.We are very grateful to Dr. P. A Revista GAMA percorre tradições, registos, ações e intervenções artísticas, que assinalaram diversos momentos, com diferentes graus de difusão. Visa-se resgatar, recuperar a arte: é voltar a olhar, pelo escopo particular de um artista, a intervenção de um outro artista, com algum tempo de intervalo entre ambos. Este intervalo é um tempo de sedimentação, de filtragem, ou de recuperação de valores quase esquecidos. O tempo pode fornecer um ponto de vista privilegiado que permite novas interpretações, valorizações, interligações. Permite-se a reativação, o restauro: restauro não no sentido material, mas no sentido da reapresentação do conteúdo possibilitando reforçar um “museu imaginário,” livre de fronteiras e de hierarquias estabelecidas (Malraux). Os objetos de arte anteriores podem ter originado, ou vir a originar, novas obras, na sequência imprevisível dos discursos humanos mais significativos, reagrupando-se em termos de “ação” ou de “orientação,” de “imagem” ou de “palavra,” (Warburg) permitindo “restituir ao discurso o caráter de acontecimento” (Foucault).
