CitotoksiÄnost, inhibicija agregacije trombocita i antioksidativna aktivnost ekstrakata biljke Curcuma amada Roxb.
Mango ginger (Curcuma amada Roxb.) is a unique spice having morphological resemblance to ginger but imparts raw mango flavour. The sequential extraction of mango ginger rhizome powder was carried out using hexane, chloroform, ethyl acetate, acetone, methanol and water. The phenolic content was the highest in methanol extract, followed by acetone, ethyl acetate and water extracts. Among these, chloroform extract exhibited high lipid peroxidation inhibitory activity and metal chelating activity, whereas ethyl acetate extract showed high DPPH radical scavenging activity and superoxide radical scavenging activity. Mango ginger extracts also showed potential platelet aggregation inhibitory activity and cytotoxicity properties.Curcuma amada Roxb. jedinstveni je zaÄin jer morfoloĆĄki nalikuje Äumbiru, a po okusu je sliÄan mangu. Genetic diversity and differentiation within and between cultivated (Vitis vinifera L. ssp. sativa) and wild (Vitis vinifera L. ssp. sylvestris) grapes
Genetic characterization of 502 diverse grape accessions including 342 cultivated (V. vinifera ssp. sativa) and 160 wild (V. vinifera ssp. sylvestris) grapes showed considerable genetic diversity among accessions. A total of 117 alleles were detected across eight SSR loci with the average of 14 alleles per locus. The genetic diversity of wild grapes was slightly lower than that observed in the cultivated grapes probably due to small populations and severe natural selection leading to drift and loss of alleles and heterozygosity in wild grapes. The distance cluster analysis (CA) supported the classical ecogeographic groups with moderate genetic differentiation among them. There was a greater affinity of Occidentalis grape to wild grape from the Caucasus than other groups. However, a number of low to moderate frequency alleles that are present in the cultivated grape are not represented in the wild grape.
Fabrication, Mechanical and Wear Properties of Aluminum (Al6061)-Silicon Carbide-Graphite Hybrid Metal Matrix Composites
In recent times, the use of aluminum alloy-based Hybrid Metal Matrix Composites (HMMCs) is being increased in aerospace and automotive applications. HMMCs compensate for the low desirable properties of each filler used. However, the mechanical properties of HMMCs are not well understood. In particular, microstructural investigations and wear optimization studies of HMMCs are not clear. Therefore, further studies are required. The present study is aimed at fabricating and mechanical and wear characterizing and microstructure investigating of Silicon Carbide (SiC) and Graphite (Gr) added in Aluminum (Al) alloy Al6061 HMMCs. The addition of SiC particles was in the range from 0 to 9 weight percentage (wt.%) in steps of 3, along with the addition of 1 wt.% Gr in powder form. The presence of alloying elements in the Al6061 alloy was identified using the Energy Dispersive X-Ray Analysis (EDX). The dispersion of SiC and Gr particles in the alloy was investigated using metallurgical microscope and Scanning Electron Microscopy (SEM). The gain in strength can be attributed to the growth in dislocation density. The nature of fracture was quasi-cleavage. The microstructure examination reveals the uniform dispersion of the reinforcement. Density, hardness, and Ultimate Tensile Strength values observed to be increased with increased contents of SiC reinforcement. Besides, wear studies were performed in dry sliding conditions. Optimization studies were performed to investigate the effect of parameters that affecting the wear. The sliding wear resistance was noticed to be improved concerning higher amounts of reinforcement leading to a decrease in delamination and adhesive wear. The predicted values for the wear rate have also been compared with the experimental results and good correlation is obtained

Patterns of genomic and phenomic diversity in wine and table grapes.
Grapes are one of the most economically and culturally important crops worldwide, and they have been bred for both winemaking and fresh consumption. Here we evaluate patterns of diversity across 33 phenotypes collected over a 17-year period from 580 table and wine grape accessions that belong to one of the world's largest grape gene banks, the grape germplasm collection of the United States Department of Agriculture. We find that phenological events throughout the growing season are correlated, and quantify the marked difference in size between table and wine grapes. By pairing publicly available historical phenotype data with genome-wide polymorphism data, we identify large effect loci controlling traits that have been targeted during domestication and breeding, including hermaphroditism, lighter skin pigmentation and muscat aroma. Breeding for larger berries in table grapes was traditionally concentrated in geographic regions where Islam predominates and alcohol was prohibited, whereas wine grapes retained the ancestral smaller size that is more desirable for winemaking in predominantly Christian regions. We uncover a novel locus with a suggestive association with berry size that harbors a signature of positive selection for larger berries. Our results suggest that religious rules concerning alcohol consumption have had a marked impact on patterns of phenomic and genomic diversity in grapes
Genetic structure and domestication history of the grape
The grape is one of the earliest domesticated fruit crops and, since antiquity, it has been widely cultivated and prized for its fruit and wine. Here, we characterize genome-wide patterns of genetic variation in over 1,000 samples of the domesticated grape, Vitis vinifera subsp. vinifera, and its wild relative, V. vinifera subsp. sylvestris from the US Department of Agriculture grape germ-plasm collection. We find support for a Near East origin of vinifera and present evidence of introgression from local sylvestris as the grape moved into Europe. High levels of genetic diversity and rapid linkage disequilibrium (LD) decay have been maintained in vinifera, which is consistent with a weak domestication bottleneck followed by thousands of years of widespread vegetative propagation. The considerable genetic diversity within vinifera, however, is contained within a complex network of close pedigree relationships that has been generated by crosses among elite cultivars. We show that first-degree relationships are rare between wine and table grapes and among grapes from geographically distant regions. Our results suggest that although substantial genetic diversity has been maintained in the grape subsequent to domestication, there has been a limited exploration of this diversity. We propose that the adoption of vegetative propagation was a double-edged sword: Although it provided a benefit by ensuring true breeding cultivars, it also discouraged the generation of unique cultivars through crosses. The grape currently faces severe pathogen pressures, and the long-term sustainability of the grape and wine industries will rely on the exploitation of the grape's tremendous natural genetic diversity
Genetic structure and differentiation in cultivated fig (Ficus carica L.)
One hundred ninety-four germplasm accessions of fig representing the four fig types, Common, Smyrna, San Pedro, and Caprifig were analyzed for genetic diversity, structure, and differentiation using genetic polymorphism at 15 microsatellite loci. The collection showed considerable polymorphism with observed number of alleles per locus ranging from four for five different loci, MFC4, LMFC14, LMFC22, LMFC31 and LMFC35 to nine for LMFC30 with an average of 4.9 alleles per locus. Seven of the 15 loci included in the genetic structure analyses exhibited significant deviation from panmixia, of which two showed excess and five showed deficiency of heterozygote. The cluster analysis (CA) revealed ten groups with 32 instances of synonymy among cultivars and groups differed significantly for frequency and composition of alleles for different loci. The principal components analysis (PCA) confirmed the results of CA with some groups more differentiated than the others. Further, the model based Bayesian approach clustering suggested a subtle population structure with mixed ancestry for most figs. The gene diversity analysis indicated that much of the total variation is found within groups (HG/HTÂ =Â 0.853; 85.3%) and the among groups within total component (GGTÂ =Â 0.147) accounted for the remaining 14.7%, of which ~64% accounted for among groups within clusters (GGCÂ =Â 0.094) and ~36% among clusters (GCTÂ =Â 0.053). The analysis of molecular variance (AMOVA) showed approximately similar results with nearly 87% of variation within groups and ~10% among groups within clusters, and ~3% among clusters. Overall, the gene pool of cultivated fig analyzed possesses substantial genetic polymorphism but exhibits narrow differentiation. It is evident that fig accessions from Turkmenistan are somewhat genetically different from the rest of the Mediterranean and the Caucasus figs. The long history of domestication and cultivation with widespread dispersal of cultivars with many synonyms has resulted in a great deal of confusion in the identification and classification of cultivars in fig
Transcriptome profiling of grapevine seedless segregants during berry development reveals candidate genes associated with berry weight
IndexaciĂłn: Web of Science; PubMedBackground
Berry size is considered as one of the main selection criteria in table grape breeding programs. However, this is a quantitative and polygenic trait, and its genetic determination is still poorly understood. Considering its economic importance, it is relevant to determine its genetic architecture and elucidate the mechanisms involved in its expression. To approach this issue, an RNA-Seq experiment based on Illumina platform was performed (14 libraries), including seedless segregants with contrasting phenotypes for berry weight at fruit setting (FST) and 6â8 mm berries (B68) phenological stages.
Results
A group of 526 differentially expressed (DE) genes were identified, by comparing seedless segregants with contrasting phenotypes for berry weight: 101 genes from the FST stage and 463 from the B68 stage. Also, we integrated differential expression, principal components analysis (PCA), correlations and network co-expression analyses to characterize the transcriptome profiling observed in segregants with contrasting phenotypes for berry weight. After this, 68 DE genes were selected as candidate genes, and seven candidate genes were validated by real time-PCR, confirming their expression profiles.
Conclusions
We have carried out the first transcriptome analysis focused on table grape seedless segregants with contrasting phenotypes for berry weight. Our findings contributed to the understanding of the mechanisms involved in berry weight determination. Also, this comparative transcriptome profiling revealed candidate genes for berry weight which could be evaluated as selection tools in table grape breeding programs.http://bmcplantbiol.biomedcentral.com/articles/10.1186/s12870-016-0789-
Incontinentia pigmenti presenting as hypodontia in a 3-year-old girl: a case report

Abstract

Introduction

Incontinentia pigmenti or Bloch-Sulzberger syndrome is a rare X-linked dominant disease that mainly affects the skin, eyes, hair, central nervous system and teeth. The disease is predominant among women. Although dermatologic manifestations are among the most important aspects for the diagnosis of the syndrome, they are less damaging to the patient and do not require treatment. However, oral involvement characterized by hypodontia of deciduous and permanent teeth is important for the diagnosis and treatment of the patient.

Case presentation

We report the case of a 3-year-old girl with ophthalmologic and neurologic disturbances, cutaneous manifestations and hypodontia. Since the patient did not present more damaging manifestations such as neurologic and/or ophthalmologic problems, her most severe complications were related to dental anomalies. The importance of integrated dental treatment, which combines pediatric dentistry, orthodontics and conventional prosthesis, is emphasized.

Conclusion

Hypodontia is a frequent finding in incontinentia pigmenti, and dentists should be aware of this condition in order to help with the diagnosis.</p
Genetic Structure of the Polymorphic Metrosideros (Myrtaceae) Complex in the Hawaiian Islands Using Nuclear Microsatellite Data
Five species of Metrosideros (Myrtaceae) are recognized in the Hawaiian Islands, including the widespread M. polymorpha, and are characterized by a multitude of distinctive, yet overlapping, habit, ecological, and morphological forms. It remains unclear, despite several previous studies, whether the morphological variation within Hawaiian Metrosideros is due to hybridization, genetic polymorphism, phenotypic plasticity, or some combination of these processes. The Hawaiian Metrosideros complex has become a model system to study ecology and evolution; however this is the first study to use microsatellite data for addressing inter-island patterns of variation from across the Hawaiian Islands.Ten nuclear microsatellite loci were genotyped from 143 individuals of Metrosideros. We took advantage of the bi-parental inheritance and rapid mutation rate of these data to examine the validity of the current taxonomy and to investigate whether Metrosideros plants from the same island are more genetically similar than plants that are morphologically similar. The Bayesian algorithm of the program structure was used to define genetic groups within Hawaiian Metrosideros and the closely related taxon M. collina from the Marquesas and Austral Islands. Several standard and nested AMOVAs were conducted to test whether the genetic diversity is structured geographically or taxonomically. The results suggest that Hawaiian Metrosideros have dynamic gene flow, with genetic and morphological diversity structured not simply by geography or taxonomy, but as a result of parallel evolution on islands following rampant island-island dispersal, in addition to ancient chloroplast capture. Results also suggest that the current taxonomy requires major revisions in order to reflect the genetic structure revealed in the microsatellite data
