    Determination of uranium and zirconium by flow injection analysis

    As an integral part of chemical quality control of nuclear materials a method for determination of uranium and zirconium, in a mixture is presented. A simple, cheap, selective and quantitative Flow Injection Analysis (FIA) system was developed. Zirconium and uranium were determinate in presence of each other and no prior separation was needed. Arsenazo III was used as a colorimetric reagent and parameters such as acidity and reagents concentration were studied and optimized. An analytical throughput of 30 sample determination per hour was obtained

    La discrimination des textuels est essentielle pour la lecture de nouveaux mots (lecture recombinant). Cette étude a employé des pseudomots dictés et imprimés avec un pseudo-alphabet pour étudier les effets de la similitude entre les mots et de lextension de lunité textuelle enseignée sur le développement de la lecture recombinant. Dans lExp. 1, universitaires ont appris des relations conditionnelles entre pseudomots dictés et imprimés qui ont différé des mots incorrects dans 25% et 75% des éléments. Dans l´Exp. 2, dautres universitaires ont appris a relation des lettres ou des syllabes dictées à correspondantes imprimées et après ils ont appris les relations avec les pseudo-mots. Lacquisition des discriminations était plus rapide à Condition Différence 75%; la lecture recombinante a été liée à la quantité de mots enseignés (Exp. 1). La lecture a montré plus dexactitude juste après lapprentissage des relations avec des syllabes, mais la lecture recombinante a été favorisée par lapprentissage des lettres. Combiner lenseignement des mots plus différents et des unités denseignement petites peut accélérer la lecture recombinante.Discriminative control by textual elements is a requisite for reading new words (recombinative reading). This study used pseudowords printed with pseudo-alphabet to evaluate the effects of the similarity between words and the size of teaching units on the development of recombinative reading. In Exp. 1, undergraduate students learned conditional relations between spoken and printed pseudowords which differed from incorrect words in 25% and 75% of the elements. In Exp. 2, undergraduate students learned to relate spoken letters or syllables to the corresponding printed units and afterwards they learned the conditional relations with pseudowords. The acquisition of discriminations was faster during the Condition 75%; recombinative reading was related to the amount of trained words (Exp. 1). Reading accuracy was higher after learning syllables, but more recombinative reading occurred after learning letters (Exp. 2). The teaching that combines dissimilar words and units smaller than words may accelerate recombinative reading.El control discriminatorio por elementos textuales es necesario para que se lea nuevas palabras (lectura con recombinación). Ese estudio utilizó pseudopalabras impresas en pseudo-alfabeto para investigar los efectos de la semejanza entre palabras y del tamaño de la unidad textual enseñada sobre el desarrollo de la lectura con recombinación. En el Exp. 1 universitarios aprendieron relaciones condicionales entre pseudopalabras dictadas e impresas que se diferenciaban de las palabras incorrectas en 25% y 75% de los elementos. En el Exp. 2, otros estudiantes aprendieron a relacionar letras o sílabas dictadas con sus correspondientes unidades impresas y después aprendieron las relaciones involucrando las pseudopalabras. Adquisición de las discriminaciones fue más rápida en la Condición Diferencia 75%; la lectura con recombinación estuvo relacionada con la cantidad de palabras entrenadas (Exp.1). Exp. 2 demostró mayor precisión de lectura después de la aprendizaje de silabas, pero la lectura con recombinación fue favorecida por la aprendizaje de letras. No Experimento 2 outros universitários aprenderam a relacionar letras ou sílabas ditadas às correspondentes modalidades impressas e subsequentemente aprenderam relações envolvendo pseudopalavras. A aquisição das discriminações foi mais rápida na Condição Diferença 75%; a leitura recombinativa dependeu da quantidade de palavras ensinadas (Exp.1). A leitura foi mais precisa após a aprendizagem de sílabas, mas ocorreu mais recombinação após a aprendizagem de letras (Exp. 2).     Phytochemical screening, cytotoxicity and acute toxicity of Annona vepretorum Mart (Annonaceae) leaf extracts

    Purpose: To investigate the phytochemistry, cytotoxicity and acute toxicity of leaf extracts from Annona vepretorum.Methods: The crude extracts were obtained by maceration with hexane and  methanol. The crude methanol extract was suspended in a 3:7 (v/v) mixture of methanol (MeOH) and water (H2O) and partitioned with hexane, chloroform (CHCl3) and ethyl acetate (AcOEt) in ascending order of polarity to obtain the respective  extracts. In the investigation of phytochemical profile, the extracts were evaluated on thin layer chromatography (TLC) plates of silica gel. Cytotoxicity was tested using 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) and (3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium) (MTS) assays against tumor cell lines, viz,HCT-116 (colon), SF-295 (brain), HL-60 (leukemic) and Sarcoma-180. Acute toxicity study was performed by administration of a single oral dose of 2 g/kg body weight of the extracts to mice and the animals were observed for 14 days.Results: Phytochemical screening results showed that A. vepretorum extracts contain alkaloids, flavonoids and terpenes. Methanol and chloroform extracts exhibited high cytotoxic activity against HCT-116, HL-60 and Sarcoma-180. Moreover, the extracts displayed low toxicity in mice, as no deaths and pronounced toxic effects were observed.Conclusion: A. vepretorum contains a variety of secondary metabolites which may confer on this species high cytotoxic activity.     Surveillance of hemorrhagic fever and/or neuroinvasive disease: challenges of diagnosis

      OBJECTIVE To evaluate the performance of post mortem laboratory analysis in identifying the causes of hemorrhagic fever and/or neuroinvasive disease in deaths by arbovirus infection. METHODS Retrospective cross-sectional study based on the differential analysis and final outcome obtained in patients whose samples underwent laboratory testing for arboviruses at the Pathology Center of the Adolfo Lutz Institute, in São Paulo, Brazil. RESULTS Of the 1355 adults clinically diagnosed with hemorrhagic fever and/or neuroinvasive disease, the most commonly attributed cause of death and the most common final outcome was dengue fever. Almost half of the samples tested negative on all laboratory tests conducted. CONCLUSION The failure to identify the causative agent in a great number of cases highlights a gap in the diagnosis of deaths of unknown etiology. Additional immunohistochemical and molecular assessments need to be added to the post-mortem protocol if all laboratory evaluations performed fail to identify a causative agent. While part of our findings may be due to technical issues related to sample fixation, better information availability when making the initial diagnosis is crucial. Including molecular approaches might lead to a significant advancement in diagnostic accuracy

    Use practices and profile of microbiological contamination of lab coats in medical school

    The coats are a potential microbiological transmission means and its misuse brings risks to health professionals and population. The goal of this study was to know the usage practices and the profile of contamination of the smock worn by professors and students of the University Center Serra dos Órgãos (UNIFESO). It is a quantitative study, cross-sectional and exploratory. Respecting ethical principles, a questionnaire on the use of the white coat was applied and the microbiological analysis was held. The data were analyzed manually, expressed in percentages and demonstrated through tables. As a result, 73 (seventy-three) coats studied showed varied flora and predominance of Gram-positive cocci. The region of the wrist, the most contaminated, presented a UFC 23 average per field. As for the usage profile, the majority, 42, (57%) are between the ages of 18 and 25 years, 46 (63%) are female, 32 (44%) are medical students in the 3rd term and only 13 (18%) are professors. On the frequency of changing of lab coat, 35 (48%) change it once a week, however with inappropriate handling. After use, 36 (49%) do not wash the coat separately, 43 (58%) do not use disinfectant products, 54 (73%) use ironing as an additional measure against microbiological contamination. It is concluded that there is imperious need for awareness of scholars and professors of appropriate use/handling of the lab coat and the importance of clothing in the epidemiology of infections intra/extra health care environment.Os jalecos são um veículo potencial de transmissão microbiológica e seu uso inadequado traz riscos à saúde de profissionais e da população. O objetivo deste estudo foi o de conhecer as práticas de utiliza- ção e o perfil de contaminação dos jalecos brancos usados como uniforme por docentes e discentes do Centro Universitário Serra dos Orgãos (UNIFESO). Trata-se de um estudo quantitativo, transversal e exploratório. Respeitados os princípios éticos, aplicou-se questionário sobre o modo de uso e se realizou a análise microbiológica. Os dados foram analisados de forma manual, expressos em percentuais e demonstrados por meio de tabelas. Como resultado, 73 (setenta e três) jalecos estudados apresentaram variada flora, com predomínio de cocos Gram positivos. A região do punho, a mais contaminada, apresentou em média 23 UFC por campo. Quanto ao perfil de uso, a maioria, 42, (57%) tem idade entre 18 e 25 anos, 46 (63%) são do sexo feminino, 27 (37%) são acadêmicos de medicina do 3o período e apenas 13 (18%) são docentes. Sobre a frequência de troca do jaleco, 35 (48%) trocam uma vez por semana, entretanto com inadequado manuseio. Após uso, 36 (49%) não lavam o jaleco separadamente, 43 (58%) não usam produto desinfetante e 54 (73%) utilizam a passagem a ferro como medida complementar de contenção microbiológica. Conclui-se que há imperiosa necessidade de conscientização dos acadêmicos e docentes sobre adequados uso e manuseio do jaleco e importância da vestimenta na epidemiologia das infecções intra e extra-ambientes da saúd

    Novel ocellatin peptides mitigate LPS-induced ROS formation and NF-kB activation in microglia and hippocampal neurons

Therefore, this study aimed to determine the primary structure and investigate peptides obtained from the cutaneous secretions of the amphibian, Leptodactylus vastus, as a source of bioactive molecules. The peptides obtained possessed the amino acid sequences, GVVDILKGAAKDLAGH and GVVDILKGAAKDLAGHLASKV, with monoisotopic masses of [M + H]± = 1563.8 Da and [M + H]± = 2062.4 Da, respectively. The molecules were characterized as peptides of the class of ocellatins and were named as Ocellatin-K1(1-16) and Ocellatin-K1(1-21). Functional analysis revealed that Ocellatin-K1(1-16) and Ocellatin-K1(1-21) showed weak antibacterial activity. However, treatment of mice with these ocellatins reduced the nitrite and malondialdehyde content. Moreover, superoxide dismutase enzymatic activity and glutathione concentration were increased in the hippocampus of mice. In addition, Ocellatin-K1(1-16) and Ocellatin-K1(1-21) were effective in impairing lipopolysaccharide (LPS)-induced reactive oxygen species (ROS) formation and NF-kB activation in living microglia. We incubated hippocampal neurons with microglial conditioned media treated with LPS and LPS in the presence of Ocellatin-K1(1-16) and Ocellatin-K1(1-21) and observed that both peptides reduced the oxidative stress in hippocampal neurons. Furthermore, these ocellatins demonstrated low cytotoxicity towards erythrocytes.     Characterization of Dengue Virus Type 2: New Insights on the 2010 Brazilian Epidemic

    Dengue viruses (DENV) serotypes 1, 2, and 3 have been causing yearly outbreaks in Brazil. In this study, we report the re-introduction of DENV2 in the coast of São Paulo State. Partial envelope viral genes were sequenced from eighteen patients with dengue fever during the 2010 epidemic. Phylogenetic analysis showed this strain belongs to the American/Asian genotype and was closely related to the virus that circulated in Rio de Janeiro in 2007 and 2008. The phylogeny also showed no clustering by clinical presentation, suggesting that the disease severity could not be explained by distinct variants or genotypes. The time of the most recent common ancestor of American/Asian genotype and the São Paulo and Rio de Janeiro (SP/RJ) monophyletic cluster was estimated to be around 40 and 10 years, respectively. Since this virus was first identified in Brazil in 2007, we suggest that it was already circulating in the country before causing the first documented outbreak. This is the first description of the 2010 outbreak in the State of São Paulo, Brazil, and should contribute to efforts to control and monitor the spread of DENVs in endemic areas

    Alguns dados sobre a Fauna entomológica da ilha das Flores - Açores

    IV Expedição Científica do Departamento de Biologia - Flores 1989Com este trabalho, realizado em Julho de 1989 nas Flores - a ilha mais ocidental do Arquipélago dos Açores -, acrescentaram-se onze espécies de Lepidópteros à lista referenciada para aquela ilha, pertencendo uma à família Lycaenidae (Lampides boeticus L.), oito a familia Noctuidae (Agrotis ipsilon HFN., Brotolomia meticulosa L., Chrysodeixis chalcites ESPER., Heliothis armigera HBN., Noctua atlantica WARREN, Noctua pronuba L., Peridroma saucia HBN., Sesamia nonagrioides LEF.), uma à família Nymphalidae (Vanessa atalanta L.) e uma a família Pyralidae (Glyphodes unionalis HBN.). Entre os demais insectos, foram identificadas cerca de duas dezenas e meia de espécies, distribuídas pelas Ordens Dermaptera, Orthoptera, Dictyoptera, Heteroptera, Homoptera, Coleoptera, Neuroptera, Diptera, Hymenoptera e Collembola. Salienta-se ainda a importância, do ponto de vista agronómico, das pragas Mythimna unipuncta (HAWORTH) e Xestia c-nigrun L. naquela ilha.RÉSUMÉ: Avec ce travail, réalisé en Juillet 1989 a Flores - l'île plus occidental de l'archipel des Açores, onze espèces de Lépidoptères ont été ajoutées à la liste des espèces connus pour cette île, dont une appartient a la famille Lycaenidae (Lampides boelicus L.), huit à la famille Noctuidae (Agrotis ipsilon HFN., Brotolomia meticulosa L. On remarque I'importance, du point de vue agronomique, des ravageurs Mythimna unipuncra (HAWORTH) et Xestia c-nigrum L. dans cette île
