    Detection, quantification and genotyping of Herpes Simplex Virus in cervicovaginal secretions by real-time PCR: a cross sectional survey

    BACKGROUND: Herpes Simplex Virus (HSV) Genital Ulcer Disease (GUD) is an important public health problem, whose interaction with HIV results in mutually enhancing epidemics. Conventional methods for detecting HSV tend to be slow and insensitive. We designed a rapid PCR-based assay to quantify and type HSV in cervicovaginal lavage (CVL) fluid of subjects attending a Genito-Urinary Medicine (GUM) clinic. Vaginal swabs, CVL fluid and venous blood were collected. Quantitative detection of HSV was conducted using real time PCR with HSV specific primers and SYBR Green I. Fluorogenic TaqMan Minor Groove Binder (MGB) probes designed around a single base mismatch in the HSV DNA polymerase I gene were used to type HSV in a separate reaction. The Kalon test was used to detect anti-HSV-2 IgG antibodies in serum. Testing for HIV, other Sexually Transmitted Infections (STI) and related infections was based on standard clinical and laboratory methods. RESULTS: Seventy consecutive GUM clinic attendees were studied. Twenty-seven subjects (39%) had detectable HSV DNA in CVL fluid; HSV-2 alone was detected in 19 (70%) subjects, HSV-1 alone was detected in 4 (15%) subjects and both HSV types were detected in 4 (15%) subjects. Eleven out of 27 subjects (41%) with anti-HSV-2 IgG had detectable HSV-2 DNA in CVL fluid. Seven subjects (10%) were HIV-positive. Three of seven (43%) HIV-infected subjects and two of five subjects with GUD (40%) were secreting HSV-2. None of the subjects in whom HSV-1 was detected had GUD. CONCLUSION: Quantitative real-time PCR and Taqman MGB probes specific for HSV-1 or -2 were used to develop an assay for quantification and typing of HSV. The majority of subjects in which HSV was detected had low levels of CVL fluid HSV, with no detectable HSV-2 antibodies and were asymptomatic

    Multiple and Precessing Collimated Outflows in the Planetary Nebula IC 4634

    With its remarkable double-S shape, IC 4634 is an archetype of point-symmetric planetary nebulae (PN). In this paper, we present a detailed study of this PN using archival HST WFPC2 and ground-based narrow-band images to investigate its morphology, and long-slit spectroscopic observations to determine its kinematics and to derive its physical conditions and excitation. The data reveal new structural components, including a distant string of knots distributed along an arc-like feature 40"-60" from the center of the nebula, a skin of enhanced [O III]/H-alpha ratio enveloping the inner shell and the double-S feature, and a triple-shell structure. The spatio-kinematical study also finds an equatorial component of the main nebula that is kinematically independent from the bright inner S-shaped arc. We have investigated in detail the bow shock-like features in IC 4634 and found that their morphological, kinematical and emission properties are consistent with the interaction of a collimated outflow with surrounding material. Indeed, the morphology and kinematics of some of these features can be interpreted using a 3D numerical simulation of a collimated outflow precessing at a moderate, time-dependent velocity. Apparently, IC 4634 has experienced several episodes of point-symmetric ejections oriented at different directions with the outer S-shaped feature being related to an earlier point-symmetric ejection and the outermost arc-like string of knots being the relic of an even much earlier point-symmetric ejection. There is tantalizing evidence that the action of these collimated outflows has also taken part in the shaping of the innermost shell and inner S-shaped arc of IC 4634.Comment: 16 pages, 11 figures, accepted for publication in The Astrophysical Journa

    Proper Motions of the Jets in the Region of HH 30 and HL/XZ Tau. Evidence for a Binary Exciting Source of the HH 30 Jet

    We present [SII] images of the HH 30 and HL/XZ Tau region obtained at two epochs, as well as long-slit optical spectroscopy of the HH 30 jet. We measured proper motions of about 100-300 km/s for the HH 30 jet and counterjet, and of about 120 km/s for the HL Tau jet. Inclination angles with respect to the plane of the sky are 0-40 deg for the HH 30 jet and 60 deg for the HL Tau jet. Comparison with previous observations suggests that most of the jet knots consist of persisting structures. Also, we corroborate that the HH 30-N knots correspond to the head of the HH 30 jet. The overall HH 30 jet structure can be well described by a wiggling ballistic jet, arising either by the orbital motion of the jet source around a primary or by precession of the jet axis because of the tidal effects of a companion. In the first scenario, the orbital period would be 53 yr and the total mass 0.25-2 solar masses. In the precession scenario, the mass of the jet source would be 0.1-1 solar masses, the orbital period <1 yr, and the mass of the companion less than a few times 0.01 solar masses, thus being a substellar object or a giant exoplanet. In both scenarios a binary system with a separation <18 AU (<0.13 arcsec) is required. Since the radius of the flared disk observed with the HST is about 250 AU, we conclude that this disk appears to be circumbinary rather than circumstellar, suggesting that the search for the collimating agent of the HH 30 jet should be carried out at much smaller scales.Comment: 42 pages, 7 figures, 5 tables.     Systemic Effector and Regulatory Immune Responses to Chlamydial Antigens in Trachomatous Trichiasis

    Trachomatous trichiasis (TT) caused by repeated or chronic ocular infection with Chlamydia trachomatis is the result of a pro-fibrotic ocular immune response. At the conjunctiva, the increased expression of both inflammatory (IL1B, TNF) and regulatory cytokines (IL10) have been associated with adverse clinical outcomes. We measured in vitro immune responses of peripheral blood to a number of chlamydial antigens. Peripheral blood effector cells (CD4, CD69, IFNγ, IL-10) and regulatory cells (CD4, CD25, FOXP3, CTLA4/GITR) were readily stimulated by C. trachomatis antigens but neither the magnitude (frequency or stimulation index) or the breadth and amount of cytokines produced in vitro [IL-5, IL-10, IL-12 (p70), IL-13, IFNγ, and TNFα] were significantly different between TT cases and their non-diseased controls. Interestingly we observed that CD4+ T cells account for <50% of the IFNγ positive cells induced following stimulation. Further investigation in individuals selected from communities where exposure to ocular infection with C. trachomatis is endemic indicated that CD3−CD56+ (classical natural killer cells) were a major early source of IFNγ production in response to C. trachomatis elementary body stimulation and that the magnitude of this response increased with age. Future efforts to unravel the contribution of the adaptive immune response to conjunctival fibrosis should focus on the early events following infection and the interaction with innate immune mediated mechanisms of inflammation in the conjunctiva

    The antikaon nuclear potential in hot and dense matter

    The antikaon optical potential in hot and dense nuclear matter is studied within the framework of a coupled-channel self-consistent calculation taking, as bare meson-baryon interaction, the meson-exchange potential of the J\"ulich group. Typical conditions found in heavy-ion collisions at GSI are explored. As in the case of zero temperature, the angular momentum components larger than L=0 contribute significantly to the finite temperature antikaon optical potential at finite momentum. It is found that the particular treatment of the medium effects has a strong influence on the behavior of the antikaon potential with temperature. Our self-consistent model, in which antikaons and pions are dressed in the medium, gives a moderately temperature dependent antikaon potential which remains attractive at GSI temperatures, contrary to what one finds if only nuclear Pauli blocking effects are included.Comment: 30 pages, 8 figures, references added.     Insulin secretion in patients with latent autoimmune diabetes (LADA): half way between type 1 and type 2 diabetes: action LADA 9

    Background: The study of endogenous insulin secretion may provide relevant insight into the comparison of the natural history of adult onset latent autoimmune diabetes (LADA) with types 1 and 2 diabetes mellitus. The aim of this study was to compare the results of the C-peptide response to mixed-meal stimulation in LADA patients with different disease durations and subjects with type 2 and adult-onset type 1 diabetes. Methods: Stimulated C-peptide secretion was assessed using the mixed-meal tolerance test in patients with LADA (n = 32), type 1 diabetes mellitus (n = 33) and type 2 diabetes mellitus (n = 30). All patients were 30 to 70 years old at disease onset. The duration of diabetes in all groups ranged from 6 months to 10 years. The recruitment strategy was predefined to include at least 10 subjects in the following 3 disease onset categories for each group: 6 to 18 months, 19 months to 5 years and 5 to 10 years. Results: At all time-points of the mixed-meal tolerance test, patients with LADA had a lower stimulated C-peptide response than the type 2 diabetes group and a higher response than the type 1 diabetes group. The same results were found when the peak or area under the C-peptide curve was measured. When the results were stratified by time since disease onset, a similar pattern of residual insulin secretory capacity was observed. Conclusions: The present study shows that the magnitude of stimulated insulin secretion in LADA is intermediate between that of type 1 and type 2 diabetes mellitus

    Identificación y caracterización de genes implicados en la maduración y la calidad de la fresa

    Ripening is a critical step for the development of flavor quality in fruits. This character has significantly declined in many fleshy fruits over recent decades. This is particularly significant in strawberry (Fragaria × ananassa), where current cultivars are derived from a narrow germplasm collection. Improving fruit quality requires two important breakthroughs: 1) a precise understanding of the fruit ripening process that will allow the targeting of relevant genes, and 2) the identification of novel alleles responsible for fruit quality traits. In our project (TRANSFR-Q, Starting Grant-ERC), we aim at the identification and characterization of key transcription factors involved in fruit ripening regulation and their target genes, in order to infer the Gene Regulatory Network controlling this process. On the other hand, we are carrying out a Genome-Wide Association Study using a germplasm collection of the woodland strawberry (Fragaria vesca) in order to identify loci involved in important traits such as aroma, fruit size, and resistance to pathogens. Finally, we have implemented the use of the genome-editing tool CRISPR/Cas9 in the cultivated strawberry, which we expect to open opportunities for engineering this species to improve traits of economic importance.ERC Starting Grant ERC-2014-StG 63813

    Influence of Large-scale Interplanetary Structures on the Propagation of Solar Energetic Particles: The Multispacecraft Event on 2021 October 9

    An intense solar energetic particle (SEP) event was observed on 2021 October 9 by multiple spacecraft distributed near the ecliptic plane at heliocentric radial distances R ≲ 1 au and within a narrow range of heliolongitudes. A stream interaction region (SIR), sequentially observed by Parker Solar Probe (PSP) at R = 0.76 au and 48° east from Earth (ϕ = E48°), STEREO-A (at R = 0.96 au, ϕ = E39°), Solar Orbiter (SolO; at R = 0.68 au, ϕ = E15°), BepiColombo (at R = 0.33 au, ϕ = W02°), and near-Earth spacecraft, regulated the observed intensity-time profiles and the anisotropic character of the SEP event. PSP, STEREO-A, and SolO detected strong anisotropies at the onset of the SEP event, which resulted from the fact that PSP and STEREO-A were in the declining-speed region of the solar wind stream responsible for the SIR and from the passage of a steady magnetic field structure by SolO during the onset of the event. By contrast, the intensity-time profiles observed near Earth displayed a delayed onset at proton energies ≳13 MeV and an accumulation of ≲5 MeV protons between the SIR and the shock driven by the parent coronal mass ejection (CME). Even though BepiColombo, STEREO-A, and SolO were nominally connected to the same region of the Sun, the intensity-time profiles at BepiColombo resemble those observed near Earth, with the bulk of low-energy ions also confined between the SIR and the CME-driven shock. This event exemplifies the impact that intervening large-scale interplanetary structures, such as corotating SIRs, have in shaping the properties of SEP events

    A coding polymorphism in matrix metalloproteinase 9 reduces risk of scarring sequelae of ocular Chlamydia trachomatis infection

    BACKGROUND: Trachoma, an infectious disease of the conjunctiva caused by Chlamydia trachomatis, is an important global cause of blindness. A dysregulated extracellular matrix (ECM) proteolysis during the processes of tissue repair following infection and inflammation are thought to play a key role in the development of fibrotic sequelae of infection, which ultimately leads to blindness. Expression and activity of matrix metalloproteinase 9 (MMP-9), a major effector of ECM turnover, is up-regulated in the inflamed conjunctiva of trachoma subjects. Genetic variation within the MMP9 gene affects in vitro MMP9 expression levels, enzymatic activity and susceptibility to various inflammatory and fibrotic conditions. METHODS: We genotyped 651 case-control pairs from trachoma endemic villages in The Gambia for coding single nucleotide polymorphisms (SNPs) in the MMP9 gene using the high-throughput Sequenom(® )system. Single marker and haplotype conditional logistic regression (CLR) analysis for disease association was performed. RESULTS: The Q279R mutation located in exon 6 of MMP9 was found to be associated with lower risk for severe disease sequelae of ocular Chlamydia trachomatis infection. This mutation, which leads to a nonsynonymous amino-acid change within the active site of the enzyme may reduce MMP-9-induced degradation of the structural components of the ECM during inflammatory episodes in trachoma and its associated fibrosis. CONCLUSION: This work supports the hypothesis that MMP-9 has a role in the pathogenesis of blinding trachoma
