    The Yellow Coral Dendrophyllia cornigera in a Warming Ocean

    Ocean warming is expected to impinge detrimentally on marine ecosystems worldwide up to impose extreme environmental conditions capable to potentially jeopardize the good ecological status of scleractinian coral reefs at shallow and bathyal depths. The integration of literature records with newly acquired remotely operated vehicle (ROV) data provides an overview of the geographic distribution of the temperate coral Dendrophyllia cornigera spanning the eastern Atlantic Ocean to the whole Mediterranean Sea. In addition, we extracted temperature values at each occurrence site to define the natural range of this coral, known to maintain its physiological processes at 16\ub0C. Our results document a living temperature range between 3c7\ub0C and 17\ub0C, suggesting that the natural thermal tolerance of this eurybathic coral may represent an advantage for its survival in a progressively warming ocean

    Facility for validating technologies for the autonomous space rendezvous and docking to uncooperative targets

    We present the latest advancements in the air-bearing facility installed at La Sapienza’s GN Lab in the School of Aerospace Engineering. This facility has been utilized in recent times to validate robust control laws for simultaneous attitude control and vibration active damping. The instrumentation and testbed have been restructured and enhanced to enable simulations of close proximity operations. Relative pose determination, accomplished through visual navigation as either an auxiliary or standalone system, is the first building block. Leveraging the acquired knowledge, optimal guidance and control algorithms can be tested for contactless operations (e.g. on-orbit inspection), as well as berthing and docking tasks

    AbstractWe investigated the optical and the electrical conductivity properties of synthetic melanin thin films spray-coated on glass. These films showed a broadband monotonic increase of the absorption coefficient, decreasing the wavelength in the Visible-NIR range. Conductivity as a function of the temperature evidenced a semiconductor like character and a hysteretic behaviour after thermal annealing up to 475 K. Thermal activation energies extrapolated by resistance curves have been explained by using the framework of a band-model as for an amorphous semiconductor

    Immunotherapy in the Treatment of Metastatic Melanoma: Current Knowledge and Future Directions

    Melanoma is one of the most immunologic malignancies based on its higher prevalence in immune-compromised patients, the evidence of brisk lymphocytic infiltrates in both primary tumors and metastases, the documented recognition of melanoma antigens by tumor-infiltrating T lymphocytes and, most important, evidence that melanoma responds to immunotherapy. The use of immunotherapy in the treatment of metastatic melanoma is a relatively late discovery for this malignancy. Recent studies have shown a significantly higher success rate with combination of immunotherapy and chemotherapy, radiotherapy, or targeted molecular therapy. Immunotherapy is associated to a panel of dysimmune toxicities called immune-related adverse events that can affect one or more organs and may limit its use. Future directions in the treatment of metastatic melanoma include immunotherapy with anti-PD1 antibodies or targeted therapy with BRAF and MEK inhibitors

    Interactions between Asaia, Plasmodium and Anopheles: new insights into mosquito symbiosis and implications in malaria symbiotic control

    Background Malaria represents one of the most devastating infectious diseases. The lack of an effective vaccine and the emergence of drug resistance make necessary the development of new effective control methods. The recent identification of bacteria of the genus Asaia, associated with larvae and adults of malaria vectors, designates them as suitable candidates for malaria paratransgenic control. To better characterize the interactions between Asaia, Plasmodium and the mosquito immune system we performed an integrated experimental approach. Methods Quantitative PCR analysis of the amount of native Asaia was performed on individual Anopheles stephensi specimens. Mosquito infection was carried out with the strain PbGFPCON and the number of parasites in the midgut was counted by fluorescent microscopy. The colonisation of infected mosquitoes was achieved using GFP or DsRed tagged-Asaia strains. Reverse transcriptase-PCR analysis, growth and phagocytosis tests were performed using An. Stephensi and Drosophila melanogaster haemocyte cultures and DsRed tagged-Asaia and Escherichia coli strains. Results Using quantitative PCR we have quantified the relative amount of Asaia in infected and uninfected mosquitoes, showing that the parasite does not interfere with bacterial blooming. The correlation curves have confirmed the active replication of Asaia, while at the same time, the intense decrease of the parasite. The 'in vitro' immunological studies have shown that Asaia induces the expression of antimicrobial peptides, however, the growth curves in conditioned medium as well as a phagocytosis test, indicated that the bacterium is not an immune-target. Using fluorescent strains of Asaia and Plasmodium we defined their co-localisation in the mosquito midgut and salivary glands. Conclusions We have provided important information about the relationship of Asaia with both Plasmodium and Anopheles. First, physiological changes in the midgut following an infected or uninfected blood meal do not negatively affect the residing Asaia population that seems to benefit from this condition. Second, Asaia can act as an immune-modulator activating antimicrobial peptide expression and seems to be adapted to the host immune response. Last, the co-localization of Asaia and Plasmodium highlights the possibility of reducing vectorial competence using bacterial recombinant strains capable of releasing anti-parasite molecules

    The practice of deep sedation in electrophysiology and cardiac pacing laboratories: Results of an italian survey promoted by the aiac (italian association of arrhythmology and cardiac pacing)

    The aim of this survey, which was open to all Italian cardiologists involved in arrhythmia, was to assess common practice regarding sedation and analgesia in interventional electrophysiology procedures in Italy. The survey consisted of 28 questions regarding the approach to sedation used for elective direct-current cardioversion (DCC), subcutaneous implantable cardioverter-defibrillator (S-ICD) implantation, atrial fibrillation (AF) ablation, ventricular tachycardia (VT) ablation, and transvenous lead extraction procedures. A total of 105 cardiologists from 92 Italian centres took part in the survey. The rate of centres where DCC, S-ICD implantation, AF ablation, VT ablation and lead extraction procedures were performed without anaesthesiologic assistance was 60.9%, 23.6%, 51.2%, 37.3%, and 66.7%, respectively. When these procedures were performed without anaesthesiologic assistance, the drugs (in addition to local anaesthetics) commonly administered were benzodiazepines (from 64.3% to 79.6%), opioids (from 74.4% to 88.1%), and general anaesthetics (from 7.1% to 30.4%). Twenty-three (21.9%) of the 105 cardiologists declared that they routinely administered propofol, without the supervision of an anaesthesiologist, in at least one of the above-mentioned procedures. In current Italian clinical practice, there is a lack of uniformity in the sedation/analgesia approach used in interventional electrophysiology procedures

    Human Papillomavirus 16 E5 Induces Bi-Nucleated Cell Formation By Cell-Cell Fusion

    Human Papillomaviruses (HPV) 16 is a DNA virus encoding three oncogenes – E5, E6, and E7. The E6 and E7 proteins have well-established roles as inhibitors of tumor suppression, but the contribution of E5 to malignant transformation is controversial. Using spontaneously immortalized human keratinocytes (HaCaT cells), we demonstrate that expression of HPV16 E5 is necessary and sufficient for the formation of bi-nucleated cells, a common characteristic of precancerous cervical lesions. Expression of E5 from non-carcinogenic HPV6b does not produce bi-nucleate cells. Video microscopy and biochemical analyses reveal that bi-nucleates arise through cell-cell fusion. Although most E5-induced bi-nucleates fail to propagate, co-expression of HPV16 E6/E7 enhances the proliferation of these cells. Expression of HPV16 E6/E7 also increases bi-nucleated cell colony formation. These findings identify a new role for HPV16 E5 and support a model in which complementary roles of the HPV16 oncogenes lead to the induction of carcinogenesis
