    The asymmetric desymmetrisation of meso compounds.

    This thesis is concerned with the asymmetric desymmetrisation of meso compounds. It commences with an introduction to the areas of two-directional synthesis and asymmetric desymmetrisation. This is followed by a review of asymmetric desymmetrisation of achiral and meso compounds, illustrating the power of this technique and the diversity of substrates and reagents that can be employed. Chapter one details the asymmetric desymmetrisation of centrosymmetric meso 2-pyridone [4 + 4]-photodimer derivatives, as a strategy towards the synthesis of functionalised amino acids. The reductive enantioselective desymmetrisation of jY-Boc tetrahydro photodimer 227 using Corey’s oxazaborolidine catalyst 236 was achieved in good yield (76%) and excellent ee (>97%), representing the first ever asymmetric desymmetrisation of a centrosymmetric compound. Chapter two details the synthesis and asymmetric desymmetrisation of meso decalin diallylc alcohols as part of a strategy towards the synthesis of core structures of bioactive Celastracecie sesquiterpenes. The two-directional synthesis of diallylic alcohol 298 (a classical meso compound containing a plane of symmetry) was completed and its subsequent desymmetrisation using a novel Zr modified Sharpless asymmetric epoxidation proceeded in reasonable yield (54%) and excellent ee (>95%

    Phosphorothioate Anti-sense Oligonucleotides: The Kinetics and Mechanism of the Generation of the Sulfurising Agent from Phenylacetyl Disulfide (PADS)

    The synthesis of phosphorothioate oligonucleotides is often accomplished in the pharmaceutical industry by the sulfurisation of the nucleotide-phosphite using phenylacetyl disulfide (PADS) which has an optimal combination of properties. This is best achieved by an initial ‘ageing’ of PADS for 48 hrs in acetonitrile with 3-picoline to generate polysulfides. The initial base-catalysed degradation of PADS occurs by an E1cB-type elimination to generate a ketene and acyldisulfide anion. Proton abstraction to reversibly generate a carbanion is demonstrated by H/D exchange, the rate of which is greatly increased by electron-withdrawing substituents in the aromatic ring of PADS. The ketene can be trapped intramolecularly by an o-allyl group. The disulfide anion generated subsequently attacks unreacted PADS on sulfur to give polysulfides, the active sulfurising agent. The rate of degradation of PADS is decreased by less basic substituted pyridines and is only first order in PADS indicating that the rate-limiting step is formation of the disulfide anion from the carbanion

    Evaluation of portable microscopic devices for the diagnosis of Schistosoma and soil-transmitted helminth infection

    The diagnosis of parasitic worm (helminth) infections requires specialized laboratory settings, but most affected individuals reside in locations without access to such facilities. We tested two portable microscopic devices for the diagnosis of helminth infections in a cross-sectional survey in rural Côte d'Ivoire. We examined 164 stool samples under a light microscope and then re-examined with a commercial portable light microscope and an experimental mobile phone microscope for the diagnosis of Schistosoma mansoni and soil-transmitted helminths. Additionally, 180 filtered urine samples were examined by standard microscopy and compared with the portable light microscope for detection of Schistosoma haematobium eggs. Conventional microscopy was considered the diagnostic reference standard. For S. mansoni, S. haematobium and Trichuris trichiura, the portable light microscope showed sensitivities of 84·8%, 78·6% and 81·5%, respectively, and specificities of 85·7%, 91·0% and 93·0%, respectively. For S. mansoni and T. trichiura, we found sensitivities for the mobile phone microscope of 68·2% and 30·8%, respectively, and specificities of 64·3% and 71·0%, respectively. We conclude that the portable light microscope has sufficient diagnostic yield for Schistosoma and T. trichiura infections, while the mobile phone microscope has only modest sensitivity in its current experimental set-up. Development of portable diagnostic technologies that can be used at point-of-sample collection will enhance diagnostic coverage in clinical and epidemiological setting

    Clinical evaluation for morbidity associated with soil-transmitted helminth infection in school-age children on Pemba Island, Tanzania

    More than 1.5 billion people are infected with soil-transmitted helminths (Ascaris lumbricoides, hookworm, Strongyloides stercoralis, and Trichuris trichiura), causing an estimated global burden in excess of 3 million disability-adjusted life years. However, the relationship between soil-transmitted helminth infection, adverse health consequences, and beneficial effects of deworming are not well understood.; We pursued a detailed longitudinal clinical evaluation of school-age children to evaluate morbidity associated with soil-transmitted helminth infection and responses to treatment. This exploratory study was embedded into a randomized controlled trial. Overall, 434 children, aged 7-14 years, underwent a detailed medical history, physical examination, stool microscopy for soil-transmitted helminths, and hemoglobin (Hb) measurement at baseline. Medical history and stool examination were repeated at 3 and 18 weeks posttreatment. Additionally, Hb measurement was performed at the 18-week treatment follow-up. Logistic regression was employed to assess clinical factors associated with soil-transmitted helminth infection at baseline, and longitudinal data analysis to examine change in health outcomes following treatment over time.; All enrolled children were infected with T. trichiura, and randomized into four different treatment interventions. None of the medical history, physical examination, and laboratory (i.e., Hb) findings were associated with A. lumbricoides, hookworm, or S. stercoralis infection at baseline. A composite of physical exam findings for anemia, including pallor of the conjunctiva, nail beds, and palmar creases predicted lower Hb values (-3.8 g/dl, 95% confidence interval (CI): -6.9, -0.6 g/dl). When examining longitudinal trends, we did not find improvements to Hb or face Wong-Baker Likert scale among children with soil-transmitted helminth infection compared to those without infection, although there was a slight trend toward improving Hb values after treating hookworm infection.; Our study demonstrates the challenges of measuring morbidity in the context of soil-transmitted helminth infection and treatment, thus confirming the mainly subtle morbidity effects of infection

    Double Spin Asymmetry of Electrons from Heavy Flavor Decays in p+p Collisions at sqrt(s)=200 GeV

    We report on the first measurement of double-spin asymmetry, A_LL, of electrons from the decays of hadrons containing heavy flavor in longitudinally polarized p+p collisions at sqrt(s)=200 GeV for p_T= 0.5 to 3.0 GeV/c. The asymmetry was measured at mid-rapidity (|eta|<0.35) with the PHENIX detector at the Relativistic Heavy Ion Collider. The measured asymmetries are consistent with zero within the statistical errors. We obtained a constraint for the polarized gluon distribution in the proton of |Delta g/g(log{_10}x= -1.6^+0.5_-0.4, {mu}=m_T^c)|^2 < 0.033 (1 sigma), based on a leading-order perturbative-quantum-chromodynamics model, using the measured asymmetry.Comment: 385 authors, 17 pages, 15 figures, 5 tables.     Measurement of long-range angular correlation and quadrupole anisotropy of pions and (anti)protons in central dd++Au collisions at sNN\sqrt{s_{_{NN}}}=200 GeV

    We present azimuthal angular correlations between charged hadrons and energy deposited in calorimeter towers in central dd++Au and minimum bias pp++pp collisions at sNN=200\sqrt{s_{_{NN}}}=200 GeV. The charged hadron is measured at midrapidity η<0.35|\eta|<0.35, and the energy is measured at large rapidity (3.7<η<3.1-3.7<\eta<-3.1, Au-going direction). An enhanced near-side angular correlation across Δη>|\Delta\eta| > 2.75 is observed in dd++Au collisions. Using the event plane method applied to the Au-going energy distribution, we extract the anisotropy strength v2v_2 for inclusive charged hadrons at midrapidity up to pT=4.5p_T=4.5 GeV/cc. We also present the measurement of v2v_2 for identified π±\pi^{\pm} and (anti)protons in central dd++Au collisions, and observe a mass-ordering pattern similar to that seen in heavy ion collisions. These results are compared with viscous hydrodynamic calculations and measurements from pp++Pb at sNN=5.02\sqrt{s_{_{NN}}}=5.02 TeV. The magnitude of the mass-ordering in dd++Au is found to be smaller than that in pp++Pb collisions, which may indicate smaller radial flow in lower energy dd++Au collisions.Comment: 424 authors, 8 pages, and 4 figures. v2 is version accepted for publication in Phys.     Cold-nuclear-matter effects on heavy-quark production at forward and backward rapidity in d+Au collisions at sqrt(s_NN)=200 GeV

    The PHENIX experiment has measured open heavy-flavor production via semileptonic decay muons over the transverse momentum range 1 < pT < 6 GeV/c at forward and backward rapidity (1.4 < |y| < 2.0) in d+Au and p+p collisions at ?sNN = 200 GeV. In central d+Au collisions an enhancement (suppression) of heavy-flavor muon production is observed at backward (forward) rapidity relative to the yield in p+p collisions scaled by the number of binary collisions. Modification of the gluon density distribution in the Au nucleus contributes in terms of anti-shadowing enhancement and shadowing suppression; however, the enhancement seen at backward rapidity exceeds expectations from this effect alone. These results, implying an important role for additional cold nuclear matter effects, serves as a key baseline for heavy-quark measurements in A+A collisions and in constraining the magnitude of charmonia breakup effects at the Relativistic Heavy Ion Collider and the Large Hadron Collider.Comment: 424 authors, 69 insitutions, 8 pages, 4 figures.     An aggravated trajectory of depression and anxiety co-morbid with hepatitis C: : A 21 to 62 month follow-up study in 61 South Australian outpatients

    BACKGROUND: This study aimed to explore the course of depression and anxiety in chronic hepatitis C patients. METHODS:   Data were combined from two studies: (1) Hospital Anxiety and Depression Scale (HADS) scores in 395 consecutive Australian outpatients from 2006 to 2010 formed the baseline measurement; and (2) Depression Anxiety Stress Scales (DASS) scores in a survey of a sub-sample of these patients in 2011 formed the follow-up measurement. After converting DASS to HADS scores, changes in symptom scores and rates of case-ness (≥8), and predictors of follow-up symptoms were assessed. RESULTS:   Follow-up data were available for 61 patients (70.5% male) whose age ranged from 24.5 to 74.6 years (M=45.6). The time to follow-up ranged from 20.7 to 61.9 months (M=43.8). Baseline rates of depression (32.8%) and anxiety (44.3%) increased to 62.3% and 67.2%, respectively. These findings were confirmed, independent of the conversion, by comparing baseline HADS and follow-up DASS scores with British community norms. Baseline anxiety and younger age predicted depression, while baseline anxiety, high school non-completion, and single relationship status predicted anxiety. CONCLUSION:  This study demonstrated a worsening trajectory of depression and anxiety. Further controlled and prospective research in a larger sample is required to confirm these findings
